SummaryThe effect of thrombin on the phosphorylating activity of platelet membranes was compared to that of trypsin. Preincubation of non-32P phosphorylated platelet membranes with or without either of these two enzymes resulted in a considerable loss of membrane protein kinase activity which was most severe when trypsin was used. Protein kinase activity and endogenous protein acceptors decreased in parallel. 32P-phosphorylated membranes showed a slow but progressive loss of label which was accelerated by trypsin. Thrombin under these conditions prevented the loss of 32P-phosphate. These results are interpreted to indicate a thrombin-induced destruction of a phosphoprotein phosphatase. The protein kinase activity of phosphorylated platelet membranes using endogenous or exogenous protein substrates showed a significant reduction compared to non-phosphorylated membranes suggesting a deactivation of protein kinase by phosphorylation of platelet membranes. Neither thrombin nor trypsin caused a qualitative change in the membrane polypeptides accepting 32P-phosphate but resulted in quantitative alterations of their ability to become phosphorylated.

The article discusses the results of the latest interdisciplinary research of Czermno stronghold and its immediate surroundings. The site is mentioned in chroniclers’ entries referring to the stronghold Cherven’ (Tale of Bygone Years, first mention under the year 981) and the so-called Cherven’ Towns. Given the scarcity of written records regarding the history of today’s Eastern Poland, Ukraine, and Belarus in the 10th and 11th centuries, recent archaeological research, supported by geoenvironmental analyses and absolute dating, brought a significant qualitative change. In 2014 and 2015, the remains of the oldest rampart of the stronghold were uncovered for the first time. A series of radiocarbon datings allows us to refer the erection of the stronghold to the second half/late 10th century. The results of several years’ interdisciplinary research (2012-2020) introduce qualitatively new data to the issue of the Cherven’ Towns, which both change current considerations and confirm the extraordinary research potential in the archeology of the discussed region.

This paper looks at the qualitative change in the foreign policy discourse by the European Union towards the Middle East, as well as the EU’s overall degree of consistency between words and deeds. By looking at European Council Conclusions as well as General Affairs Council conclusions, it will be argued that on a discursive level the Union has taken stock of the emergence of new threats to its security, and has started shifting its attention from state failure and regional conflicts to the threats posed by terrorism and non-conventional proliferation. Secondly, by differentiating among three kinds of coherence, it will be shown that the main source of incoherence in the Union external action in the Middle East is not to be found in its institutional or horizontal dimensions, but in its vertical level, that is between the Union and member states. Examples will be provided in order to substantiate an overall claim: the EU security discourse might have changed; its policies however remain driven by the difficult balancing exercise between Brussels and national capitals.   Full text available at: https://doi.org/10.22215/rera.v3i2.184

AbstractThe unprecedented efforts to minimize the effects of the COVID-19 pandemic introduce a new arena for human face recognition in which faces are partially occluded with masks. Here, we tested the extent to which face masks change the way faces are perceived. To this end, we evaluated face processing abilities for masked and unmasked faces in a large online sample of adult observers (n = 496) using an adapted version of the Cambridge Face Memory Test, a validated measure of face perception abilities in humans. As expected, a substantial decrease in performance was found for masked faces. Importantly, the inclusion of masks also led to a qualitative change in the way masked faces are perceived. In particular, holistic processing, the hallmark of face perception, was disrupted for faces with masks, as suggested by a reduced inversion effect. Similar changes were found whether masks were included during the study or the test phases of the experiment. Together, we provide novel evidence for quantitative and qualitative alterations in the processing of masked faces that could have significant effects on daily activities and social interactions.

Abstract The analysis of the production and yield of sunflower seeds in Ukraine for the period from 2000 to 2019 was conducted in the article. The comparative analysis of the gross harvest of sunflower seeds and the export of sunflower oil for the years under research was carried out. The dependence of exports on gross harvest was revealed and its share was calculated. It was determined that the export of sunflower oil has increased over the years under research, which indicates a significant Ukraine’s export potential. It was found that the increase in the share of exports by 15.9% was made possible by a qualitative change in yield, that was ensured by the changes in the cultivation technology and by the selection of sunflower hybrids that are better adapted to climate changes. The recommendations for further improvement of cultivation technology in connection with climate change in order to further increase yields and the export potential of Ukraine were given.

Multiple auditory steady-state responses were evoked by eight tonal stimuli (four per ear), with each stimulus simultaneously modulated in both amplitude and frequency. The modulation frequencies varied from 80 to 95 Hz and the carrier frequencies were 500, 1000, 2000, and 4000 Hz. For air conduction, the differences between physiologic thresholds for these mixed-modulation (MM) stimuli and behavioral thresholds for pure tones in 31 adult subjects with a sensorineural hearing impairment and 14 adult subjects with normal hearing were 14 ± 11, 5 ± 9, 5 ± 9, and 9 ± 10 dB (correlation coefficients .85, .94, .95, and .95) for the 500-, 1000-, 2000-, and 4000-Hz carrier frequencies, respectively. Similar results were obtained in subjects with simulated conductive hearing losses. Responses to stimuli presented through a forehead bone conductor showed physiologic-behavioral threshold differences of 22 ± 8, 14 ± 5, 5 ± 8, and 5 ± 10 dB for the 500-, 1000-, 2000-, and 4000-Hz carrier frequencies, respectively. These responses were attenuated by white noise presented concurrently through the bone conductor.

Analysis of the practice of planning is increasingly being used to develop planning theory, The papers by Roweis and Forester in the second issue of Environment and Planning D: Society and Space base analysis of planning practice on hermeneutic, linguistic, and phenomenological approaches, as an alternative to the technical -rational approach to planning theory, In the present paper, I argue that the approaches adopted by these two authors create more problems than they solve, and a critique of Roweis's and Forester's theoretical ideas is made, It is argued that these approaches rest upon idealist ontological assumptions, rendering explanation of qualitative change (development) impossible. Discussion of Giddens's concept of structuration and of the negative consequences for scientific explanation of Habermas's epistemological position is presented, as both approaches are used by Roweis and Forester. Criticism is also made of the separation of territorial relations from relations of substance. Finally, the serious consequences of their approaches for scientific and social practice are outlined. I conclude that this type of approach cannot provide a satisfactory basis for planning theory, and furthermore, that the approach is inherently conservative. Some ideas arc presented concerning planning theory based on materialist ontological foundations.

The present study examined the development of calcium binding protein-containing neurons in a timed series of fetal neocortical transplants. The immunoexpression of parvalbumin and calbindin, which are subpopulations of GABAergic neurons, have been widely studied in normal development and in disease and injury states. Because of their purported resistance to oxidative injury by their ability to buffer Ca++ influx, these neurons have been particularly studied following ischemia. Because it is likely that oxidative stress is associated with the grafting procedure, we sought to determine if these neurons displayed enhanced survival characteristics. Normally, parvalbumin and calbindin represent about 5-10% of cortical neurons. Within 2-4 wk after grafting the expression of both proteins increased markedly in that a relatively larger number of neurons (27% for parvalbumin) were immunopositive. This increase was transitory, however, and by 4 mo and beyond, confocal microscopic data showed a reduction of over 50% of parvalbumin (+) neurons and processes. Calbindin (+) processes showed a qualitative change in that they were smaller with less terminal branching. Electron microscopy confirmed a substantial reduction in parvalbumin synaptic contacts. Interestingly, in older grafts, remaining parvalbumin neurons were those that were strongly NSE (+) suggesting a link between normal metabolism and Ca++ buffering in grafted neurons. It is possible that in early grafts certain neuronal populations transiently upregulated calcium binding proteins as a defensive mechanism against Ca++ influx associated with oxidative stress. Over time, however, following physiological normalization within grafts, the calcium binding protein (+) neurons are diminished, possibly due to lack of appropriate afferent input to the interneuronal pool.

Insulin/IGF-I-dependent signals play important roles for the regulation of proliferation, differentiation, metabolism, and autophagy in various cells, including hematopoietic cells. Although the early protein kinase activation cascade has been intensively studied, the whole picture of intracellular signaling events has not yet been clarified. To identify novel downstream effectors of insulin-dependent signals in relatively early phases, we performed high-resolution two-dimensional electrophoresis (2-DE)-based proteomic analysis using human hematopoietic cells 1 h after insulin stimulation. We identified SRp20, a splicing factor, and CLIC1, an intracellular chloride ion channel, as novel downstream effectors besides previously reported effectors of Rho-guanine nucleotide dissociation inhibitor 2 and glutathione S-transferase-pi. Reduction in SRp20 was confirmed by one-dimensional Western blotting. Moreover, MG-132, a proteasome inhibitor, prevented this reduction. By contrast, upregulation of CLIC1 was not observed in one-dimensional Western blotting, unlike the 2-DE results. As hydrophilic proteins were predominantly recovered in 2-DE, the discrepancy between the 1-DE and 2-DE results may indicate a certain qualitative change of the protein. Indeed, the nuclear localization pattern of CLIC1 was remarkably changed by insulin stimulation. Thus insulin induces the proteasome-dependent degradation of SRp20 as well as the subnuclear relocalization of CLIC1.
