AbstractMeta-analyses of neuroimaging studies have not found a clear relationship between the orbitofrontal cortex and obesity, despite animal and human studies suggesting the contrary. Our primary meta-analysis examined what regions are associated with reduced gray matter volume, given increased body mass index. We identified 23 voxel-based morphometry studies examining the association between gray matter volume and body mass index. In a sample of 6,788 participants, we found that greater body mass index is associated with decreased gray matter volume in the right Brodmanns’ area 10 and 11, forming part of the right orbitofrontal cortex (FWE, p=0.05). Use of Brodmanns’ areas 10 and 11 as seeds in a Neurosynth network coactivation and text decoding analysis revealed that these regions are associated with studies of emotional regulation and processing, clinical symptoms and disorder, ‘mentalizing’ and social cognition, and the Default mode network. Our finding uniquely contributes to the literature in showing a relationship between the orbitofrontal cortex and obesity and showing the wide-ranging impact these differences may have on social, mental, and emotional functioning as well as on the Default mode network. Exploratory analyses suggest the need for studies examining the effect of age on these findings.

Aims/hypothesis: Diabetes mellitus (DM) is associated with comorbid brain disorders. Neuroimaging studies in DM revealed neuronal degeneration in several cortical and subcortical brain regions. Previous studies indicate more pronounced brain alterations in type 2 diabetes mellitus (T2DM) than in type 1 diabetes mellitus (T1DM). However, a comparison of both types of DM in a single analysis has not been done so far. The aim of this meta-analysis was to conduct an unbiased objective investigation of neuroanatomical differences in DM by combining voxel-based morphometry (VBM) studies of T1DM and T2DM using dual disorder anatomical likelihood estimation (ALE) quantification.Methods: PubMed, Web of Science and Medline were systematically searched for publications until June 15, 2020. VBM studies comparing gray matter volume (GMV) differences between DM patients and controls at the whole-brain level were included. Study coordinates were entered into the ALE meta-analysis to investigate the extent to which T1DM, T2DM, or both conditions contribute to gray matter volume differences compared to controls.Results: Twenty studies (comprising of 1,175 patients matched with 1,013 controls) were included, with seven studies on GMV alterations in T1DM and 13 studies on GMV alterations in T2DM. ALE analysis revealed seven clusters of significantly lower GMV in T1DM and T2DM patients relative to controls across studies. Both DM subtypes showed GMV reductions in the left caudate, right superior temporal lobe, and left cuneus. Conversely, GMV reductions associated exclusively with T2DM (>99% contribution) were found in the left cingulate, right posterior lobe, right caudate and left occipital lobe. Meta-regression revealed no significant influence of study size, disease duration, and HbA1c values.Conclusions/interpretation: Our findings suggest a more pronounced gray matter atrophy in T2DM compared to T1DM. The increased risk of microvascular or macrovascular complications, as well as the disease-specific pathology of T2DM may contribute to observed GMV reductions.Systematic Review Registration: [PROSPERO], identifier [CRD42020142525].

Bilingualism is of great interest to the neuroscience of language, and understanding the anatomical changes associated with second language learning help inform theories of bilingual advantage across the lifespan. While the literature on structural differences between bilinguals and monolinguals is robust, relatively few studies of gray matter volume (GMV) have directly compared bilinguals with monolinguals in a whole-brain analysis. Overall, this and heterogeneity of study samples and methodology have led to a lack of clear anatomical support for major theories. Here, we engage in an activation likelihood estimate (ALE) meta-analysis of voxel-based morphometry (VBM) studies of GMV for cases that directly compare bilingual and monolingual subjects in a whole-brain analysis. The analysis (sixteen foci, from ten contrasts across eight studies) resulted in one cluster located primarily within the anterior lobe of the right cerebellum. However, when the one pediatric study was removed, the analysis revealed no consistent results across the studies included in this meta-analysis. This suggests that for VBM studies of bilingual and monolingual adults there is considerable heterogeneity of results that complicate the understanding of the bilingual brain. Future studies will need to include larger, more well-defined samples and interrogate more fine-grained anatomical features such as cortical thickness and surface area in order to more fully examine the anatomical changes associated with bilingualism across the lifespan.

AbstractMotivation theories of obesity suggest that one of the brain mechanisms underlying pathological eating and weight gain is the dysregulation of dopaminergic circuits. While these dysregulations occur likely at the microscopic level, studies on gray matter volume reported macroscopic differences associated with obesity. One region suggested to play a key role in the pathophysiology of obesity is the nucleus accumbens (NAcc). We performed a meta-analysis of findings regarding NAcc volume and overweight/obesity. We additionally examined whether gray matter volume in the NAcc and other mesolimbic areas depends on the longitudinal trajectory of obesity, using the UK Biobank dataset. To this end, we analysed the data using a latent growth model, which identifies whether certain variables of interest (e.g. NAcc volume) is related to another variable’s (BMI) initial values or longitudinal trajectories. Our meta-analysis showed that, overall, NAcc volume is positively related to BMI. However, further analyses revealed that the relationship between NAcc volume and BMI is dependent on age. For younger individuals such relationship is positive, while for older adults it is negative. This was corroborated by our analysis in the UK Biobank dataset, which includes older adults, where we found that higher BMI was associated with lower NAcc and thalamus volume. Overall, our study suggests that increased NAcc volume in young age might be a vulnerability factor for obesity, while in the older age decreased NAcc volume with increased BMI might be an effect of prolonged influences of neuroinflammation on the brain.
