Gold@palladium–gold nanocrystals with a tunable branched shape are prepared via seeded growth, where the use of a syringe pump allows the manipulation over reaction kinetics as coupled by surface diffusion and strain caused by lattice mismatch.

Serotonin receptors play important roles in neuronal excitation, emotion, platelet aggregation, and vasoconstriction. The serotonin receptor subtype 2A (5-HT2AR) is a Gq-coupled GPCR, which activate phospholipase C. Although the structures and functions of 5-HT2ARs have been well studied, little has been known about their real-time dynamics. In this study, we analyzed the intramolecular motion of the 5-HT2AR in living cells using the diffracted X-ray tracking (DXT) technique. The DXT is a very precise single-molecular analytical technique, which tracks diffraction spots from the gold nanocrystals labeled on the protein surface. Trajectory analysis provides insight into protein dynamics. The 5-HT2ARs were transiently expressed in HEK 293 cells, and the gold nanocrystals were attached to the N-terminal introduced FLAG-tag via anti-FLAG antibodies. The motions were recorded with a frame rate of 100 μs per frame. A lifetime filtering technique demonstrated that the unliganded receptors contain high mobility population with clockwise twisting. This rotation was, however, abolished by either a full agonist α-methylserotonin or an inverse agonist ketanserin. Mutation analysis revealed that the “ionic lock” between the DRY motif in the third transmembrane segment and a negatively charged residue of the sixth transmembrane segment is essential for the torsional motion at the N-terminus of the receptor.

ABSTRACTThe biomass accumulation and movement of biofilms in a microchannel is monitored by optical microscopy. First, the average optical density of the biofilm is monitored in time as a measure of biofilm thickness and structural heterogeneity. These results are used as inputs to calculate changing flow velocities due to resulting excluded volume. Next the displacement velocity of moving biofilm segments was recorded in different places in the microchannel. Quantitative analysis by a particle tracking routine showed differences in displacement velocity near and far from the microchannel corner, which is believed to be related to the local shear forces which vary depending on the height of the biofilm segment and its position in the microchannel. The effect of changing biofilm thickness and different hydrodynamic environments in the microchannel are then discussed in terms of their effects on molecular loading rates. Finally, a demonstration of a flow-templated growth approach as a means to homogenize the growth environment.

This paper addresses a special zone design problem for economic census investigators that is motivated by a real-world application. This paper presented a heuristic multikernel growth approach via Constrained Delaunay Triangulation (CDT). This approach not only solved the barriers problem but also dealt with the polygon data in zoning procedure. In addition, it uses a new heuristic method to speed up the zoning process greatly on the premise of the required quality of zoning. At last, two special instances for economic census were performed, highlighting the performance of this approach.
