ABSTRACT Recognition of virus presence via RIG-I (retinoic acid inducible gene I) and/or MDA5 (melanoma differentiation-associated protein 5) initiates a signaling cascade that culminates in transcription of innate response genes such as those encoding the alpha/beta interferon (IFN-α/β) cytokines. It is generally assumed that MDA5 is activated by long molecules of double-stranded RNA (dsRNA) produced by annealing of complementary RNAs generated during viral infection. Here, we used an antibody to dsRNA to show that the presence of immunoreactivity in virus-infected cells does indeed correlate with the ability of RNA extracted from these cells to activate MDA5. Furthermore, RNA from cells infected with encephalomyocarditis virus or with vaccinia virus and precipitated with the anti-dsRNA antibody can bind to MDA5 and induce MDA5-dependent IFN-α/β production upon transfection into indicator cells. However, a prominent band of dsRNA apparent in cells infected with either virus does not stimulate IFN-α/β production. Instead, stimulatory activity resides in higher-order structured RNA that contains single-stranded RNA and dsRNA. These results suggest that MDA5 activation requires an RNA web rather than simply long molecules of dsRNA.

A high-order compact-differencing and filtering algorithm, coupled with the classical fourth-order Runge–Kutta scheme, is developed and implemented to simulate aeroacoustic phenomena on curvilinear geometries. Several issues pertinent to the use of such schemes are addressed. The impact of mesh stretching in the generation of high-frequency spurious modes is examined and the need for a discriminating higher-order filter procedure is established and resolved. The incorporation of these filtering techniques also permits a robust treatment of outflow radiation condition by taking advantage of energy transfer to high-frequencies caused by rapid mesh stretching. For conditions on the scatterer, higher-order one-sided filter treatments are shown to be superior in terms of accuracy and stability compared to standard explicit variations. Computations demonstrate that these algorithmic components are also crucial to the success of interface treatments created in multi-domain and domain-decomposition strategies. For three-dimensional computations, special metric relations are employed to assure the fidelity of the scheme in highly curvilinear meshes. A variety of problems, including several benchmark computations, demonstrate the success of the overall computational strategy.

The recent construct of Self-leadership, which includes cognitive and behavioral strategies of managing oneself, has yet to be examined for associations with central personality dimensions such as the Big Five and their higher-order factors (Alpha, Beta). It was hypothesized that Self-leadership and its subfacets would be significantly correlated with all Big Five traits except Agreeableness, albeit higher with Extraversion and Openness to Experiences as it should pertain more strongly to agentic than communal traits. Analyses in university students ( N = 168) indicated that Self-leadership and its facets were more strongly related to Beta (Agency) than Alpha (Communion), and, although there were mostly positive correlations, Self-leadership should be distinguished from the Big Five traits. Findings are discussed regarding Self-leadership's associations with the Big Five traits and higher-order factors.
