Oxidative stress plays an important role in the development of complications in diabetes mellitus. Antioxidant therapy has been thought to decrease oxidative stress. The objective of the present study was to explore the effects of melatonin (MLT) on oxidative stress in diabetic rat eye and brain tissue by using immunohistochemical methods. Diabetes was induced by streptozotocin, (STZ, 55 mg/kg/i.p) in adult rats. MLT was given 10 mg/kg/i.p once a day for 2 weeks beginning from the sixth week. Six weeks later, rats were divided into three groups: control (CR), STZ-induced diabetic (STZ), and STZ-induced diabetic group received melatonin (STZ+MLT). Although no significant difference was observed with respect to antioxidant status, NOS activity tended to be higher in the untreated diabetic rats than in the treated rats. It was observed that MLT treatment improved the histopathological changes including apoptosis and oxidative stress in brain and eye in diabetic rat.

The purpose of this study was to investigate the effects of vitamin B6 (Vit B6) on oxidant and antioxidant status in streptozotocin-induced diabetic rats. Thirty-two Wistar rats were divided into four groups: control (C), control + Vit B6 group (C + Vit B6), diabetes (D), and diabetes + Vit B6 group (D + Vit B6). Vit B6 (4 mg/kg body weight) was administered in drinking water for 4 weeks after the induction of diabetes. Vitamin B6 reduced serum total cholesterol level in the C + Vit B6 (P< 0.01) and D + Vit B6 (P< 0.05) groups. Plasma and tissue malondialdehyde levels were reduced in the C + Vit B6 and D + Vit B6 groups. Whole blood glutathione peroxidase (GSH-Px) and erythrocyte superoxide dismutase (SOD) activities were higher in the D group (P< 0.05). GSH-Px and SOD activities were increased in C + Vit B6 group while these parameters decreased in the D + Vit B6 group. Paraoxonase and arylesterase activities were decreased in the D group while they were increased in C + Vit B6 and D + Vit B6 groups. The results of present study suggest that vitamin B6 supplementation might be a promising adjunctive agent for improving oxidative stress and metabolic disturbances and for preventing diabetic complications including atherogenesis.

The present study was designed to investigate the protective effect of the aqueous extract of Portulaca oleracea against hyperglycemic, oxidative damage and inflammation in the serum of streptozotocin (STZ)-induced diabetic rats. In the present study, the rats were divided into the following groups of 8 animals each: control, untreated diabetic, 3 Portulaca oleracea (100, 200, 400 mg/kg/d)–treated diabetic groups. At the end of the 4-week period, glucose, interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), malondialdehyde (MDA), glutathione (GSH), and total antioxidant status (TAS) levels were measured. STZ caused an elevation in the serum levels of glucose, MDA, IL-6, and TNF-α with reduction in the levels of GSH and TAS ( P < .01). Portulaca oleracea ameliorated glucose, MDA, IL-6, TNF-α, GSH, and TAS levels in diabetic groups versus to the untreated groups ( P < .05). Taken together, Portulaca oleracea prevented hyperglycemia by preventing the oxidative stress and inflammation.

Diabetes mellitus (DM) is a serious metabolic disorder with altered carbohydrate, fat, and protein metabolism. In the last four decades, India has emerged as an epicenter of the global diabetes mellitus pandemic. Rapid changes in the developmental scenario, demographic changes, and living style in the Indian subcontinent have led to the explosive increase in diabetes. Present research probes with ethanolic extract of Gymnema sylvestre (500 mg/kg.b.w) for treatment of hyperglycemia and related oxidative stress caused by Alloxan (100 mg/kg.b.wt), as a diabetogenic agent. 25 rats were included in the research divided into 5 groups, each containing 5 rats. Group 1 (normal rats), Group II (Diabetic rats (DM), Group III (DM+ treated for 10 days), Group IV (DM+20 days treated), Group V (DM+30 days treated). Blood samples and pancreatic tissues were collected at each interval of time. The blood sample was used for biochemical parameter and tissues were used for the anti-oxidant assay. Gymnema sylvestre extract (GSE) showed glucose-lowering property meanwhile, insulin secretion also increased as compared to Diabetic rats. Other tests like amylase, lipase, ALT, and AST also showed significant recovery after the extract administration. Oxidative stress was found in the Diabetic group, but after extract treatment concentration of superoxide dismutase, Glutathione-S-transferase, catalase, Glutathione peroxidase, Glutathione, and Total thiol was regained. Imbalance in serum electrolyte recovered and dysregulated hematological parameters due to stress and hyperglycemia showed convincing results. The finding suggests Gymnema sylvestre could be used as a hypoglycemic as well as an anti-oxidant agent in diabetes.
