Free-Radical-Scavenging and Antioxidant Activities of Secondary Metabolites from Reddened Cv. Annurca Apple Fruits

In this research study, quantitative determination of phytoconstituents in seed coat, dehusked raw dal, cooked dal and whole seed of locally grown pulses viz. pigeonpea, lentil, desi and kabuli chickpea, and ricebean was carried out. Among the four treatments of all the pulses, highest yield in methanol was found in ricebean. Total phenolic content (mg GAEg-1) varied from 1.72-103.62 in all of the extracts. Similarly, flavonoids (mg CAEg-1) ranged from 1.13 to 33.66 which also found to be reduced in dehulled and cooked extracts. o-Dihydric phenols, hydrophobic and hydrophilic phenols followed order as: seed coat> whole seed> raw dal> cooked dal. Results obtained by DPPH method showed that free radical scavenging capacities ranged from 53.80 to 92.00% while antioxidant activity ranged from 51.00 to 78.60% by FTC method. A highly significant correlation (plessthan0.05) between antioxidant activities/free radical scavenging efficiencies and phenolic contents as well as flavonoids was observed in most of the extracts under study. The results concluded that the pulse seed extracts may be valuable natural source of secondary metabolites for nutraceutical industry.

Objectives: The present study was carried out to determine the phytochemical constituents and in vitro antioxidant activities of leaf extracts of Alternanthera brasiliana (L). Kuntze (A. brasiliana) and Alternanthera bettzickiana regel (A. bettzickiana).Methods: Preliminary phytochemical screening was performed in petroleum ether, chloroform, acetone, ethanol, hydroethanolic and water extracts of leaves of A. brasiliana and A. bettzickiana. The level of alkaloids, flavonoids, total phenolic content, tannins, Vitamin C, Vitamin E, GSH, and total proteins were determined in hydroethanolic, ethanol, and water extracts. Antioxidant activity of the hydroethanolic leaf extracts of A. brasiliana and A. bettzickiana were determined by 2,2-diphenyl-1-picryl-hydrazyl-hydrate free radical scavenging assay, nitric oxide scavenging assay, superoxide anion scavenging assay, ferric reducing antioxidant power assay, total antioxidant capacity, and reducing power assay.Results: The phytochemical screening of six different extracts of A. brasiliana and A. bettzickiana revealed the presence of various phytonutrients. Quantitative analysis of secondary metabolites in ethanol, hydroethanolic and water extracts of leaves of A. brasiliana and A. bettzickiana showed the presence of high amount of secondary metabolites in the hydroethanolic extract. In vitro, antioxidant assay of two plant extracts revealed that A. bettzickiana was more potent than A. brasiliana in scavenging free radicals.Conclusion: The different extracts from A. brasiliana and A. bettzickiana and specifically the hydroethanolic extract of A. bettzickiana revealed several properties such as rich source of phytonutrients, higher free radical scavenging properties, and significant antioxidant capabilities. Therefore, the bioactive compound should be isolated in future studies and could be used as a safe and serve as a potential source of natural antioxidants.

A series of 20 new biologically active derivatives of 2-{4, 5-(substituted diphenyl)-4H-1,2,4-triazol-3-ylthio}acetyl chloride has been synthesized, with the aim to investigate antimicrobial, free radical scavenging activity. All the synthesized compounds were characterized by spectroscopic data and elemental analysis. The final compounds were tested for antibacterial activity against Gram-positive bacteria: Staphylococcus aureus MTCC 3160, Bacillus subtilis MTCC 441; Gram-negative bacteria: Escherichia coli MTCC 443, and, for antifungal activity, against Candida albicans MTCC 227 and Aspergillus niger MTCC 281, taking ciprofloxacin as antibacterial and fluconazole as antifungal standard drugs. Compound 7a6 was found to be the most effective antibacterial (MIC = 3.12 µg/ml), and compounds 7a2 and 7d1 (MIC = 3.12 and 6.25 µg/ml) had the most effective antifungal effects on the selected strains, as compared to the standard drugs. The results of antioxidant studies revealed that compound 7b1 was found to be most active antioxidant, with 40.4±0.687 µg/ml, and compounds 7b3, 7d7, and 7d4 also showed promising free radical scavenging activity, as compared with the standard drug ascorbic acid.

The research was conducted to explore the potency of pluchea leaves-black tea drink as antioxidant at various proportions. The research used a single factor randomized block design of pluchea leaves and black tea proportions, including 100:0; 75:25; 50:50; 25:75; and 0:100% (w/w). Each of it was repeated five times. The parameters observed in this study were secondary metabolites, total phenolic, total flavonoids, free radical DPPH scavenging activity, and iron reducing power. The data were statistically analyzed using Analysis of Variance (ANOVA) at α = 5%, if the analysis showed a significant effect then it was continued with Duncan's Multiple Range Test (DMRT). Data was stated as mean ± standard deviation. The results showed that the secondary metabolites containing in drink from pluchea leaves and black tea at various proportions were alkaloids, flavonoids, phenolics, saponins, tannins, and cardiac glycosides. The increasing of black tea proportion in samples added the intensity of alkaloids, phenolics, flavonoids, saponins, and cardiac glycosides compounds detected, but the tannins were decreased. These secondary metabolites were correlated with total phenolic content (TPC) and total flavonoid content (TFC). The increasing of black tea proportion in drink significantly decreased DPPH free radical scavenging activity and iron ion reducing power, except for 100% black tea proportion. Tannin compounds seems determining antioxidant activity.  Based on coefficient correlation between TPC or TFC and DPPH free radical scavenging activity or iron ion reducing power, the result showed that DPPH free radical scavenging activity was dominantly contributed by TPC and iron ion reducing power was determined by TPC and TFC. ABSTRAKPenelitian ini dilakukan untuk mengetahui potensi minuman daun beluntas teh hitam sebagai antioksidan pada berbagai proporsi. Penelitian ini menggunakan rancangan acak kelompok satu faktor, yaitu proporsi daun beluntas teh hitam meliputi 100:0; 75:25; 50:50; 25:75; dan 0:100% (b/b). Tiap faktor diulang sebanyak 5 kali. Parameter yang diamati pada penelitian ini meliputi kandungan metabolit sekunder, total fenolik (TPC), total flavonoid (TFC), kemampuan menangkal radikal bebas DPPH, dan kemampuan mereduksi ion besi. Data dianalisis secara statistik dengan Analysis of Variance (ANOVA) pada α = 5%, jika terdapat beda signifikan dilanjutkan dengan uji Duncan's Multiple Range Test (DMRT). Data dinyatakan dengan rata-rata ± standar deviasi.  Hasil menunjukkan bahwa metabolit sekunder yang terkandung dalam minuman daun beluntas teh hitam pada berbagai proporsi adalah alkaloid, flavonoid, fenolik, saponin, tannin, dan kardiak glikosida. Peningkatan proporsi teh hitam menambah intensitas senyawa alkaloid, fenolik, flavonoid, saponin, dan kardiak glikosida yang terdeteksi, tetapi kandungan senyawa tannin berkurang. Kandungan metabolit sekunder ini berkorelasi dengan total fenolik (TPC) dan total flavonoid (TFC). Peningkatan proporsi teh hitam dalam minuman menurunkan kemampuan menangkal radikal bebas DPPH dan kemampuan mereduksi ion besi, kecuali pada proporsi teh hitam 100%. Kandungan senyawa tannin dalam minuman menentukan aktivitas antioksidan. Berdasarkan koefisien korelasi antara TPC atau TFC dan kemampuan menangkal radikal bebas DPPH atau kemampuan mereduksi ion besi, hasil menunjukkan bahwa kemampuan menangkal radikal bebas DPPH dominan dikontribusi oleh TPC dan kemampuan mereduksi ion besi ditentukan oleh TPC dan TFC.

Processing of legumes before consumption has several effects on micronutrients, macronutrients and phytonutrients. This study was undertaken to investigate the effect of domestic processing on antioxidant activities and phenolic phytochemicals of the red bambara groundnuts and red beans. The study employed in vitro antioxidant assays (DPPH and FRAP) to screen for antioxidant properties, HPLC-PDA-ESI-MS and Folin Ciocalteu assay to screen for phenolic phytochemical profiles. Domestic cooking displayed positive effects on the antioxidant activity and phenolic phytochemical profiles of the two legumes. The free radical scavenging speed increased 10-fold in the methanolic extract from cooked red bambara groundnuts compared to uncooked. By contrast, the free radical scavenging speed increased 20-fold in the methanolic extract from cooked red beans compared to uncooked. HPLC-PDA-ESI-MS profiles of the cooked red bambara groundnuts and red beans revealed a number of emergent phenolic compounds, mainly flavonoids. These data indicate that cooking appear to enhance the nutraceutical profiles of the legumes investigated.

ABSTRACTObjective: The objective of this study is to evaluate and compare the free radical scavenging activities of selective dopamine agonist namely ropiniroleand pramipexole.Methods: The antioxidant activity of ropinirole and pramipexole at various concentrations was done by 1, 1-diphenyl-2picrylhydrazyl (DPPH) freeradical scavenging assay comparing it with ascorbic acid which was taken as standard.Results: The free radical scavenging property as measured by DPPH method showed that ropinirole and pramipexole have got a potent free radicalscavenging activity with that of ascorbic acid.Conclusion: Novel drugs such as pramipexole and ropinirole are promising molecules in the field of oxidative damage related neurodegenerativedisorders providing us an optimistic targeted approach toward neuroprotection.Keywords: Free radical scavenging, 1, 1-diphenyl-2picrylhydrazyl assay, Anti-Parkinson’s, Dopamine agonist.
