Objective: To isolate the antibacterial proteins/peptides from Ficus glomerata leaf.Methods: Present study was designed to investigate antibacterial activity of proteins/peptides isolated from Ficus glomerata leaf. The isolated proteins/peptides were further checked for antibacterial activity against, Pseudomonas aeruginosa, Bacillus subtilis, Escherichia coli and Salmonella entrica bacterial pathogens.Results: The results indicates that a 35kDa of protein were identified and exhibit good antibacterial activity against bacterial pathogen among all strains, Salmonella entrica and Pseudomonas aeruginosa exhibit good results with a clear zone of inhibition.Conclusion: Ficus glomerata is popular for its medicinal properties against therapeutic potential. In the present study a novel protein with broad spectrum antibacterial activity. Microbes cause severe damage to plants which results in a large economic loss so; this protein can be use as an active agent in agriculture for plant protection and also in the development of novel therapeutic agents.

The antibacterial activity of geraniol and its effect in combination with ampicillin, amoxicillin and clarithromycin against Staphylococcus aureus, Escherichia coli and Helicobacter pylori was tested. The minimum inhibitory concentrations (MICs) and combinatory effects of geraniol against the bacteria were assessed by using the modified broth microdilution and checkerboard assay, respectively. The combinatory effect is expressed as fractional inhibitory concentration index (FICI). The MIC of geraniol against S. aureus, E. coli and H. pylori was found to be 11200, 5600, and 7325 μg/mL, respectively. A significant synergistic effect was observed with geraniol and ampicillin against S. aureus with FICI in the range 0.19 to 0.32. Geraniol and ampicillin exhibited a partial synergistic effect against E. coli. A similar effect was observed with geraniol and clarithromycin against S. aureus. A partial synergistic effect was observed with clarithromycin and geraniol against H. pylori with the FICI value in the range 0.86 to 0.89. An additive effect was observed with geraniol and amoxicillin combination against H. pylori. However, the amoxicillin and clarithromycin dose was reduced by thirty-two fold when combined with geraniol against H. pylori. The anti- H. pylori effect of geraniol with clarithromycin and amoxicillin could be of potential interest in the treatment of H. pylori infection and associated ulcers in humans. Further, geraniol, in combination with other antibiotics, has substantial therapeutic potential against S. aureus and E.coli infection.

Abstract Typhoid fever is a significant health problem in developing countries like Pakistan. Salmonella Typhi the causative agent of typhoid has developed resistant to almost all recommended antibiotics. Emergence of resistance to third generation cephalosporins has further complicated the situation and such strains are called as extensively drug resistant (XDR) Salmonella Typhi. Currently only available options are azithromycin and cabapenems. Recently few reports of azithromycin resistance have emerged from countries like Pakistan, India, Bangladesh and Nepal. As azithromycin is the only oral option available to treat XDR Typhoid, development of resistance may change treatment strategy altogether from out patient management to hospitalization of every patient. This may increase the burden on already weak health care system of countries like Pakistan. So there is dire need to look for the alternative treatment options. Manuka honey is well known for its therapeutic potential against wide range of bacteria including Salmonella Typhimurium. In this study 3 azithromycin resistant isolates were isolated and identified using disc diffusion, E-test and broth micro dilution methods and antibacterial activity, MIC and MBC of manuka honey was performed by agar well diffusion assay and broth micro dilution assay respectively. Manuka honey manifested significant antibacterial activity against all test isolates with zone of inhibition ranging from 7.3mm to 7.5mm, MIC and MBC values were between 10 to 15% v/v Here, we conclude that Manuka honey possess potent antibacterial activity and might be used as an alternative treatment option against azithromycin resistant XDR Typhid. However, further clinical trials are mandatory to validate our initial findings.

Aim: Antibiotic resistance is an alarming issue, as multidrug-resistant bacteria are growing worldwide, hence the decrease of therapeutic potential of available antibiotic arsenal. Among these bacteria, Staphylococcus aureus was pointed by the WHO in the pathogens list to be prioritized in drug development. Methods: We report the use of chemical similarity models for the virtual screening of new antibacterial with structural similarity to known inhibitors of FabI. The potential inhibitors were experimentally evaluated for antibacterial activity and membrane disrupting capabilities. Results & conclusion: These models led to the finding of four new compounds with antibacterial activity, one of which having antimicrobial activity already reported in the literature.

Background and Objectives: Probiotics are nonpathogenic and beneficial viable microorganisms that exhibit potential health welfare for human beings. Probiotics are found in various food products. They also occur as natural microflora in the intestine of mammals. Main goal of this study was to isolate probiotics conferring antibacterial activity and cholesterol lowering ability from different fruits. Materials and Method: Present research reveals the usefulness of probiotics, in which twenty one bacterial cultures were isolated from different fruit samples including figs, coconut water and grapes. These strains were explored for their antibacterial and cholesterol reduction ability by conducting in vitro experiments. Results and Conclusion: Among twenty one isolates, nine probiotic cultures FgC2, FgC7, FgC14, G2C5, G1C,GrC18 and StCW showed maximum antibacterial activity against different human clinical pathogens. This suggests that these microbes produce inhibitory metabolites which are extracellular and diffusible. For cholesterol assimilation assay, six strains FgC2, FgC7,FgC12, FgC13, GrC7 and GrC18 presented remarkable cholesterol lowering efficacy (up to 98%) when grown in the presence of bile salts. Only potential probiotic cultures were identified and characterized as lactic acid bacteria (LAB), on the basis of Bergey’s Manual of Determinative Bacteriology. Thus this study is helpful to exploit the bioactive and therapeutic potential of beneficial microorganisms so that they can be utilized in the generation of functional food and other health promoting products.

Background. External infections involving the skin and wound are the most frequent complications affecting humans and animals. Medicinal plants play great roles in the treatment of skin and wound infections. This study was aimed to evaluate thein vitroantibacterial activity of crude methanolic extracts of nine medicinal plants.Methods. Agar well diffusion and broth dilution methods were used to determine the antibacterial activity of nine Ethiopian plants against four bacterial species includingStaphylococcus aureus,Pseudomonas aeruginosa,Escherichia coli, andKlebsiella pneumoniae.Results. Among the tested plants, seven (Cissus quadrangularis,Commelina benghalensis,Euphorbia heterophylla,Euphorbia prostrate,Momordica schimperiana,Trianthemaspp., andSolanum incanum) were found to exhibit considerable antibacterial activity against at least one of the test bacteria. The extracts ofC. quadrangularis,E. heterophylla, andE. prostratahad a wide spectrum of antibacterial activities against test bacterial strains while the extracts ofGrewia villosaandSchinus molledid not show any inhibitory activity. Clinical isolate and laboratory strain of S.aureusshowed the highest susceptibility to highest concentration (780 mg/mL) ofE. prostratawith a zone of inhibition of 21.0mm and 22.3mm, respectively.Conclusion. This study indicates clear evidence supporting the traditional use of seven plants in treating skin and wound infections related to bacteria.
