AbstractMetallic Platinum dissolves under oxygen atmosphere in aqueous histidine solution. About 0.1 to 0.3 μmol of a water soluble histidine platinum complex per ml solution have been found by atomic absorption spectroscopy.

Since the discovery of the platinum based complex, cisplatin, medicinal inorganic chemistry has attracted much more attention and a large number of platinum complexes with promising pharmacological properties have been synthesized. In this work a new platinum complex of N-(5-indanyl(methylene)anthranilic acid(5-indanyl methylene)-hydrazide (HL) has been synthesized and characterized by physical and spectral techniques, as elemental analysis, IR, EI-MS, 1H-NMR, thermal analysis, transmittance electron microscope (TEM) and magnetic moment. The results indicated that the ligand binds to Pt(II) in the enol form. Square-planar stereochemistry was suggested for the Pt(II) complex. The morphological characterization showed nano-sized spherical particles with average size 92 nm of the isolated complex. The synthesized Pt(II) complex exhibited a significant cytotoxic activity against HCT116 and HEPG2. Also in vivo study of the Pt(II) complex showed cytotoxic activity towards Ehrlich ascites carcinoma (EAC).

Proteasome inhibitors are useful for biochemical research and clinical treatment. In our previous study, we reported that the 4N-coordinated platinum complexes with anthracenyl ring and heterocycle exhibited proteasome-inhibitory activity. In the present study, the structure–activity relationships and characterization of these complexes were determined for the elucidation of the role of aromatic ligands. Lineweaver–Burk analysis revealed that the chemical structure of heterocycles affects the binding mode of platinum complexes. Platinum complexes with anthracenyl ring and pyridine showed competitive inhibition, although platinum complexes with anthracenyl ring and phenanthroline showed non-competitive inhibition. The structure–activity relationships demonstrated that anthracenyl moiety plays a crucial role in proteasome-inhibitory activity. The platinum complexes with naphthyl or phenyl rings exhibited lower inhibitory activities than the platinum complex with anthracenyl ring. The reactivity with N-acetylcysteine varied according to the chemical structure of complexes.

Three platinum complexes with cis and trans configuration cis -[Pt(TCEP) 2 Cl 2 ], cis -[Pt(tmTCEP) 2 Cl 2 ] and trans -[Pt(TCEP) 2 Cl 2 ], where TCEP is tris(2-carboxyethyl)phosphine, have been synthesized and fully characterized by usual techniques including single-crystal X-ray diffraction for trans -[Pt(TCEP) 2 Cl 2 ] and cis -[Pt(tmTCEP) 2 Cl 2 ]. Here, we also report on an esterification process of TCEP, which takes place in the presence of alcohols, leading to a platinum complex coordinated to an ester tmTCEP (2-methoxycarbonylethyl phosphine) ligand. The stability in solution of the three compounds and their interaction with biological models such as DNA (pBR322 and calf thymus DNA) and proteins (lysozyme and RNase) have also been studied.

A platinum (II) complex stabilized by a pyridine and an N-heterocyclic carbene ligand featuring an anthracenyl moiety was prepared. The compound was fully characterized and its molecular structure was determined by single-crystal X-ray diffraction. The compound demonstrated high in vitro antiproliferative activities against cancer cell lines with IC50 ranging from 10 to 80 nM. The presence of the anthracenyl moiety on the N-heterocyclic carbene (NHC) Pt complex was used as a luminescent tag to probe the metal interaction with the nucleobases of the DNA through a pyridine-nucleobase ligand exchange. Such interaction of the platinum complex with DNA was corroborated by optical tweezers techniques and liquid phase atomic force microscopy (AFM). The results revealed a two-state interaction between the platinum complex and the DNA strands. This two-state behavior was quantified from the different experiments due to contour length variations. At 24 h incubation, the stretching curves revealed multiple structural breakages, and AFM imaging revealed a highly compact and dense structure of platinum complexes bridging the DNA strands.

Activated carbon and alumina catalysts were prepared by impregnation with solutions of platinum compounds differing substantially in the nature of their interaction with the supports: chloroplatinic acid (dissolved in water or acetone), tetraammineplatinum dichloride, and tetrakis (triphenylphosphine)platinum complex. The relations between the course of impregnation, distribution of platinum across the catalyst grain, and the activity of the catalyst in liquid phase hydrogenations were studied. Lower adsorption rates and amounts of the platinum compound adsorbed on the support bring about a more uniform platinum distribution across the grain. Catalysts with a uniform distribution exhibit higher activities than those with nonuniform distribution. The highest activity was achieved with Pt/alumina catalysts prepared from triphenylphosphine platinum comlex thermally treated in oxygen atmosphere prior to the reduction.
