Abstract Several mammalian peptide hormones and proteins from plant and animal origin contain an N-terminal pyroglutamic acid (pGlu) residue. Frequently, the moiety is important in exerting biological function in either mediating interaction with receptors or stabilizing against N-terminal degradation. Glutaminyl cyclases (QCs) were isolated from different plants and animals catalyzing pGlu formation. The recent resolution of the 3D structures of Carica papaya and human QCs clearly supports different evolutionary origins of the proteins, which is also reflected by different enzymatic mechanisms. The broad substrate specificity is revealed by the heterogeneity of physiological substrates of plant and animal QCs, including cytokines, matrix proteins and pathogenesis-related proteins. Moreover, recent evidence also suggests human QC as a catalyst of pGlu formation at the N-terminus of amyloid peptides, which contribute to Alzheimer's disease. Obviously, owing to its biophysical properties, the function of pGlu in plant and animal proteins is very similar in terms of stabilizing or mediating protein and peptide structure. It is possible that the requirement for catalysis of pGlu formation under physiological conditions may have triggered separate evolution of QCs in plants and animals.

Biotinylation of antibodies to be used in immunolocalization studies is a well established approach to mapping epitopes in biological specimens at both the light and ultrastructural level. Biotinylation of antibodies does not appear to significantly alter their interaction with antigen. A similar approach has been proposed for examining ligand interaction with receptors. Producing a soluble biotinylated ligand that behaves similarly to unlabeled ligand avoids the problems introduced by particulate labels such as ferritin, gold and latex. Particulate labels for ligands can be especially problematic in studies of receptor behavior in live cells since viable cells may directly react to the particulate probe itself. The interaction of plasma fibrinogen with its receptor on blood platelets, GPIIb-IIIa, is required for platelet-platelet cohesion and subsequent formation of a hemostatic platelet plug at sites of vascular injury. To study the events that follow binding of fibrinogen to its receptor5 we have produced a series of fibrinogens biotinylated with molar ratios of biotin:fibrinogen that range from 5 to 50.

An optical AND logic device based on chiral surface plasmon polaritons (SPPs) is theoretically proposed. The AND logic device is made up of five Y-shape nanowires with two input ports and one output port. Chiral SPPs excited at the end of input ports propagate along the nanowire. By adding two gold blocks on one side of the AND logic device, the chirality of the device is amplified. By modifying parameters, we find suitable parameters to design the structure with a large chirality at the working wavelength. By setting the threshold, we achieve the AND logic devices. Our structure may provide a significant component in signal transmission and processing devices.
