AbstractPseudomonas donghuensis P482 is a tomato rhizosphere isolate with the ability to inhibit growth of bacterial and fungal plant pathogens. Herein, we analysed the impact of the carbon source on the antibacterial activity of P482 and expression of the selected genes of three genomic regions in the P482 genome. These regions are involved in the synthesis of pyoverdine, 7-hydroxytropolone (7-HT) and an unknown compound (“cluster 17”) and are responsible for the antimicrobial activity of P482. We showed that the P482 mutants, defective in these regions, show variations and contrasting patterns of growth inhibition of the target pathogen under given nutritional conditions (with glucose or glycerol as a carbon source). We also selected and validated the reference genes for gene expression studies in P. donghuensis P482. Amongst ten candidate genes, we found gyrB, rpoD and mrdA the most stably expressed. Using selected reference genes in RT-qPCR, we assessed the expression of the genes of interest under minimal medium conditions with glucose or glycerol as carbon sources. Glycerol was shown to negatively affect the expression of genes necessary for 7-HT synthesis. The significance of this finding in the light of the role of nutrient (carbon) availability in biological plant protection is discussed.

This paper discusses the structural difference and role of xylan, procedures involved in the production of xylooligosaccharides (XOS), and their implementation into animal feeds. Xylan is non-starch polysaccharides that share a β-(1-4)-linked xylopyranose backbone as a common feature. Due to the myriad of residues that can be substituted on the polymers within the xylan family, more anti-nutritional factors are associated with certain types of xylan than others. XOS are sugar oligomers extracted from xylan-containing lignocellulosic materials, such as crop residues, wood, and herbaceous biomass, that possess prebiotic effects. XOS can also be produced in the intestine of monogastric animals to some extent when exogenous enzymes, such as xylanase, are added to the feed. Xylanase supplementation is a common practice within both swine and poultry production to reduce intestinal viscosity and improve digestive utilization of nutrients. The efficacy of xylanase supplementation varies widely due a number of factors, one of which being the presence of xylanase inhibitors present in common feedstuffs. The use of prebiotics in animal feeding is gaining popularity as producers look to accelerate growth rate, enhance intestinal health, and improve other production parameters in an attempt to provide a safe and sustainable food product. Available research on the impact of xylan, XOS, as well as xylanase on the growth and health of swine and poultry, is also summarized. The response to xylanase supplementation in swine and poultry feeds is highly variable and whether the benefits are a result of nutrient release from NSP, reduction in digesta viscosity, production of short chain xylooligosaccharides or a combination of these is still in question. XOS supplementation seems to benefit both swine and poultry at various stages of production, as well as varying levels of XOS purity and degree of polymerization; however, further research is needed to elucidate the ideal dosage, purity, and degree of polymerization needed to confer benefits on intestinal health and performance in each respective species.

The effectiveness of enhanced biological phosphorus removal (EBPR) systems is directly affected by the competition of polyphosphate accumulating organisms (PAOs) and glycogen accumulating organisms (GAOs). This study investigated the short-term effects of carbon source on PAO and GAO performance. The tests were designed to clearly determine the impact of volatile fatty acid (VFA) composition on the performance of two types of biomass, one enriched for PAOs and the other for GAOs. The two populations were enriched in separate reactors using identical operating conditions and very similar influent compositions with acetate as the sole carbon source. The only difference was that a very low level of phosphorus was present in the influent to the GAO reactor. The abundance of PAOs and GAOs was quantified using fluorescence in-situ hybridisation. The results clearly show that there are some very distinctive differences between PAOs and GAOs in their ability to utilise different carbon substrates. While both are able to take up acetate rapidly and completely, the GAOs are far slower at consuming propionate than the PAOs during short-term substrate changes. This provides a potentially highly valuable avenue to influence the competition between PAOs and GAOs. Other VFAs studied seem to be less usable in the short term by both PAOs and GAOs, as indicated by their much lower uptake rates.

A mixture of wheat bran with maize bran as a carbon source and addition of (NH4)SO4 as nitrogen source was found to significantly increase production of feruloyl esterase (FAE) enzyme compared with wheat bran as a sole carbon and nitrogen source. The optimal conditions in conical flasks were carbon source (30 g) to water 1 : 1, maize bran to wheat bran 1 : 2, (NH4)SO4 1.2 g and MgSO4 70 mg. Under these conditions, FAE activity was 7.68 mU/g. The FAE activity on the mixed carbon sources showed, high activity against the plant cell walls contained in the cultures.

The research work was carried out to investigate the efficacy of different mother culture media viz., rice straw (T-1), rice straw and rice bran (T-2), rice husk (T-3), rice grain (T-4), maize grain (T-5), and rice straw and wheat bran (T-6) and the impact of age of mother culture and substrate sterilization techniques viz., sun dried for 8 hrs covering with transparent polythene (A-1), black polythene (A-2), blue polythene (A-3) sheet, autoclave for two hrs at 121°C (A-4), and hot water for one hr (A-5) for the commercial cultivation of Volvariella volvacea (Bull.) Singer. The maximum mycelium run rate and minimum days required for completing the mother culture were recorded in T-4. The lowest days required for primordial initiation (DRFPI) was 6 in T-1 and T-2, whereas highest DRFPI was recorded in T-3. The maximum (13.33) days required for first harvest (DRFFH) and lowest (109) number of effective fruiting bodies (NEFB) were recorded in T-3. The minimum (10.67 days) DRFFH was found in T-2 and maximum (239.30) NEFB was recorded in T-1. The lowest length and diameter (LFB and DFB) were recorded in T-5 (3.03 cm) and T-1 (1.66 cm), while highest LFB and DFB were observed in T-3 (3.20 and 2.39 cm). Maximum biological yield and efficiency were observed in rice straw and wheat bran materials. The highest NEFB, DFB, biological yield and efficiency were recorded in 30 days old of mother culture. The results revealed that combined rice straw and wheat bran were the excellent mother culture medium and 30 days old was the best age for the commercial production of paddy straw mushroom. Considering the experimental results on the sterilization techniques it may be suggested that hot water sterilization of rice straw substrate was the best sterilization technique for the commercial production and yield improvement of V. volvacea.

A new P-N containing the flame retardant, which was namely N,N′-dibutyl-phosphate diamide (DBPDA), was synthesized and it was assembled into the cavity of β-cyclodextrin (β-CD) to form an inclusion complex (IC). The structure and properties of IC were characterized by Fourier transform infraredspectroscopy (FTIR), wide-angle X-ray diffraction (WAXD), 1H nuclear magnetic resonance (1H NMR), scanning electron microscopy with X-ray microanalysis (SEM-EDS), differential scanning calorimeter (DSC) and thermal gravimetric analysis (TGA). 1H NMR and SEM-EDS were also used to identify the molar ratio of β-CD/DBPDA in IC and the results from the analyses indicated that their molar ratio was 1:1. In order to test the flame retardant effect of IC, it was added to epoxy (EP). IC was proposed to be able to act as an intumescent flame retardant (IFR) system in EP through a combination of β-CD and DBPDA properties during the combustion process. β-CD is a biomass carbon source, which has the advantages of environmental protection and low cost. Furthermore, DBPDA is both a source of acid and gas. When IC was heated, IC had the advantage of acting as both a carbon source and foam forming agent, while the DBPDA component were able to directly generate phosphoric acid and NH3 in situ. The impact of IC in low additive amounts on flame retardancy of EP was studied by the cone calorimeter test. When only 3 wt % IC was incorporated, the peak values of heat release rate (pHRR) and smoke production rate (pSPR) of EP were reduced by 22.9% and 33.3% respectively, which suggested that IC could suppress the heat and smoke release efficiently.
