The persistence of 2,4-D [(2,4-dichlorophenoxy) acetic acid] and 2,4,5-T [(2,4,5-trichlorophenoxy)acetic acid] was studied in the chaparral environment. Chamise (Adenostoma fasciculatumH. & A.), grass and forbs, soil surface litter, and soil were sampled for up to 360 days after herbicide (4.5 kg/ha) application. Over 50% of the 2,4-D and 2,4,5-T recovered was found on soil surface litter while 18 to 31% was found on vegetation. Much less herbicide (0.1 to 0.2%) was found in soil (0 to 5 cm) than in foliage or litter. No herbicide residues were found below the 0 to 5 cm soil sample zone. Herbicide residues on foliage and litter decreased rapidly (up to 93%) within 30 days after application but remained constant thereafter until winter rainfall began. Residues of both herbicides were evident in chamise foliage (0.01 to 0.02%), surface litter (0.01 to 0.03%), and soil (0.01%) up to 360 days after application.

Placentomes, taken from ewes at 101-141 days gestation, were homogenized and subjected to differential and density gradient centrifugation. In the presence or absence of protease inhibitors, choriomammotropin (ovine chorionic somatomammotropin, oCS) was detected by radioimmunoassay, and banded at two positions in 30-60% w/v sucrose gradients. The first band (soluble form) eluted in the sample zone of the gradient and contained 33�6� 1 % (mean�s.e.m., n = 10) of the total oCS present in the gradient. The remainder (48� 7�6%) of the oCS (particulate form) sedimented to 37�5-43% w/v sucrose. Both these bands became sharper with decreasing protein loaded onto the gradients. 125I_oCS, pre-equilibrated with a placentome preparation, eluted in the same position as the first band of endogenous oCS. When loaded onto a second gradient the second band eluted at 38-41 % w /v sucrose, but after being subjected to ultrasonic disruption or treatment with Triton X-I00 this oCS eluted in the sample zone. There was no correlation between gestational age of the ewe and the percentage oCS in either the soluble or particulate form, the concentration of oCS, or enrichment by protein.

There are many situations when it is necessary to separate rapidly and efficiently a cytosolic and a membrane vesicle fraction from yeast, cultured cells, or from bacteria. This Protocol Article describes the flotation of the vesicles through a self-generated gradient from a dense sample zone using the low-viscosity medium iodixanol. As the sample is exposed to the gmaxthe tendency of the proteins to sediment overcomes any diffusion in the opposite direction and are therefore completely separated from the vesicles.
