Abstract Assimilation of continuously streamed monitored data is an essential component of a digital twin; the assimilated data are used to ensure the digital twin represents the monitored system as accurately as possible. One way this is achieved is by calibration of simulation models, whether data-derived or physics-based, or a combination of both. Traditional manual calibration is not possible in this context; hence, new methods are required for continuous calibration. In this paper, a particle filter methodology for continuous calibration of the physics-based model element of a digital twin is presented and applied to an example of an underground farm. The methodology is applied to a synthetic problem with known calibration parameter values prior to being used in conjunction with monitored data. The proposed methodology is compared against static and sequential Bayesian calibration approaches and compares favourably in terms of determination of the distribution of parameter values and analysis run times, both essential requirements. The methodology is shown to be potentially useful as a means to ensure continuing model fidelity.

Studies of sewage sludge conditioning by ultrasonic field concentrate on determination of the increase of water removal effect, which depends on kind of sludge and chemical compounds used in the dewatering process. An attempt was made to find new methods of sludge preparation before dewatering. Tests presented here focused on digested and difficult dewatered sludge. The sludge was dewatered on a vacuum filter after conditioning with polyelectrolytes and the ultrasonic field. The microscopic analysis was an additional criterion to evaluate changes in the sludge structure after preparation. The polyelectrolyte dose of 3 mg/g d.m. sonicated within 15 sec. resulted in the 50% decrease of sludge volume. Results presented confirmed our previous experiences, concerning the relation between conglomerates of sludge and the effect of dewatering.

Reliable cytopathological diagnosis requires new methods and approaches for the rapid and accurate determination of all cell types. This is especially important when the number of cells is limited, such as in the cytological samples of fine-needle biopsy. Immunoplasmonic-multiplexed- labeling may be one of the emerging solutions to such problems. However, to be accepted and used by the practicing pathologists, new methods must be compatible and complementary with existing cytopathology approaches where counterstaining is central to the correct interpretation of immunolabeling. In addition, the optical detection and imaging setup for immunoplasmonic-multiplexed-labeling must be implemented on the same cytopathological microscope, not interfere with standard H&E imaging, and operate as a second easy-to-use imaging method. In this article, we present multiplex imaging of four types of nanoplasmonic markers on two types of H&E-stained cytological specimens (formalin-fixed paraffin embedded and non-embedded adherent cancer cells) using a specially designed adapter for SI dark-field microscopy. The obtained results confirm the effectiveness of the proposed optical method for quantitative and multiplex identification of various plasmonic NPs, and the possibility of using immunoplasmonic-multiplexed-labeling for cytopathological diagnostics.

Most of the mathematical models that were developed to study the UK 2001 foot-and-mouth disease epidemic assumed that the infectiousness of infected premises was constant over their infectious periods. However, there is some controversy over whether this assumption is appropriate. Uncertainty about which farm infected which in 2001 means that the only method to determine if there were trends in farm infectiousness is the fitting of mechanistic mathematical models to the epidemic data. The parameter values that are estimated using this technique, however, may be influenced by missing and inaccurate data. In particular to the UK 2001 epidemic, this includes unreported infectives, inaccurate farm infection dates and unknown farm latent periods. Here, we show that such data degradation prevents successful determination of trends in farm infectiousness.

Apart from traditional optimization techniques, e.g. progressive optimality algorithm (POA), modern intelligence algorithms, like genetic algorithms, differential evolution have been widely used to solve optimization problems. This paper deals with comparative analysis of POA, GA and DE and their applications in a reservoir operation problem. The results show that both GA and DES are feasible to reservoir operation optimization, but they display different features. GA and DE have many parameters and are difficult in determination of these parameter values. For simple problems with mall number of decision variables, GA and DE are better than POA when adopting appropriate parameter values and constraint handling methods. But for complex problem with large number of variables, POA combined with simplex method are much superior to GA and DE in time-assuming and quality of optimal solutions. This study helps to select proper optimization algorithms and parameter values in reservoir operation.
