The susceptibility of laboratory rats (Rattus norvegicus) to experimental inoculation with tachyzoites of Neospora caninum was examined. Groups of female rats were intramuscularly injected with 0, 2, or 4 mg of methylprednisolone acetate (MPA) 7 days prior to, and on the day of, subcutaneous inoculation with 0 or 1.5 × 105 tachyzoites. Clinical signs of disease or deaths did not occur in rats given nothing or only N. caninum tachyzoites. Rats given only 4 mg MPA failed to grow as well as rats given nothing or only N. caninum tachyzoites but were otherwise healthy. All of 20 rats given 4 mg MPA and tachyzoites died of hepatitis and pneumonia within 12 days postinoculation. Hepatic necrosis was the most striking lesion seen in these rats, and other milder lesions consisted of pneumonia, encephalitis, and myositis. The response of rats given 2 mg MPA and tachyzoites was less severe. Three of 20 rats died with encephalitis, myositis, hepatitis, and pancreatitis. Mild lesions, but no N. caninum tachyzoites, were seen in 3 of 14 rats inoculated only with tachyzoites. Rats given the 4 mg MPA treatment and inoculated with N. caninum tachyzoites appear to be suitable subjects for examining acute neosporosis and could be used in studies designed to examine treatment of acute disease.

Abstract This study was conducted to improve the quality of oocytes in old-female rats by using pregnant mare serum gonadotrophin (PMSG). Female rats at productive age were used as a control.  The experimental rats were injected with 4 doses of PMSG i.e., 0, 2.5, 5.0, and 10 IU PMSG. After 2 weeks of acclimation to the experimantal condition, the experimental rats were injected with PGF2α at a dose of 25µg/g BW two times with 2 days interval to synchronize estrous cycle. PMSG injections were conducted at the same time with the second PGF2α injection.  After PMSG injection, the experimental rats were divided into two groups rats, i.e., rats without mating for masurement of oocyte qualities and rats mated for measurement of offspring qualities.  Therefore, 16 experimental rats from each age group were sacrifized for maeasurement of oocyte qualities.  The other 16 experimental rats for each age group were mated for measurement of offspring qualities. Parameters measured were hematological profile, uterine and ovarian weights, the qualities of oocytes, the qualities of the offsspring born by using swimming test and rat maze test. The collected data were analyzed by analysis of variance (ANOVA) and continued with Duncan test with a 95% confidence level. The data were analyzed using SSPS. The results showed that the improvement in the quality of oocytes in old female rats using the PMSG hormone in this study showed an increase in the quality of oocytes in old and productive age rats. The highest number of oocyte quality was found in rats of productive age (3.25) and significantly different (P< 0.05) from the other oocyte qualities. The quality of offspring born to old age and productive age rats injected with PMSG were improved.  It was concluded that the improvement of oocyte quality by using PMSG also improves offspring qualities.

This study evaluated the effect of maternal restraint stress during the gestation period on behaviors, biochemical parameters, and antioxidant capacities of offspring rats (Rattus norvegicus (Berkenhout,1769)) at weaning age. Behaviors, plasma biochemical indices, and antioxidant ability of the liver, soleus muscle, and gastrocnemius muscle of mother and (or) offspring rats were analyzed. Significant increases were found in the immobility and swinging behavior frequencies of offspring male rats; no difference was found in behaviors of female rats. The antioxidant indices including superoxide dismutase, nitric oxide synthase, and total antioxidant capacity in the soleus muscle of offspring male rats were significantly decreased in the restraint group. Female offspring rats showed significant lower glutathione and higher malondialdehyde levels in the gastrocnemius muscle and liver, respectively. No difference was found in the productive performance and plasma biochemical indices of maternal rats, nor in the biochemical parameters of the two groups of weaning rats. The results suggested that maternal chronic stresses negatively affected the behaviors and antioxidant abilities of offspring rats, and that these effects possibly have a greater impact on offspring male rats than on female rats.

Red calliandra (Calliandra calothyrsus Meissn.) leaf have the potential as phytoestrogens. In this study, adult female ovariectomized rats (as a model of menopause women) were given calliandra leaf extract containing estrogenic flavonoids. This study was an experimental design with a Completely Randomized Design (CRD). Forty-five ovariectomized female rats were divided into five groups i.e. untreated control (K), and the treatment group given 80% of C. calothyrsus ethanolic leaves extract doses 25 mg/ kg bw (P1), 50 mg/ kg bw (P2), 75 mg/ kg bw (P3) and 100 mg/ kg bw (P4). The test suspension was given once a day at 24-hour intervals by gavage as much as 0.5 ml/rat/day for two months. At the end of the treatments, blood was collected for blood biochemical tests including hemoglobin levels, hematocrit values, blood glucose and proteins levels. The results showed that C. calothyrsus leaf extract did not negatively affect the health status of the animals through blood biochemical parameters, i.e. hemoglobin levels, hematocrit values, and glucose and protein levels of ovariectomized female rats.

Na presente pesquisa objetivou-se estudar os efeitos do gossipol (C30H30O8) na gestação e consequente lactação, de 40 ratas com idade média de 120 dias e peso variando entre 200 e 250 gramas, oriundas do Biotério Central da Universidade Federal do Paraná. Os animais foram acompanhados diariamente até a confirmação da cobertura através de esfregaço de lavado vaginal (esfregaço positivo - presença de espermatozóides) e separados em quatro grupos de dez animais: G I - controle, G II – gossipol 20 mg.kg-1, G III – gossipol 40 mg.kg-1, G IV - gossipol 80 mg.kg-1 respectivamente de acordo com a concentração de gossipol presente na suspensão utilizada (gossipol 20 mg.mL-1, gossipol 40 mg.mL-1 e gossipol 80 mg.mL-1). O gossipol ácido acético (GAA 97,8 % de pureza) diluído em óleo vegetal de canola (veículo) foi administrado oralmente para todos os animais no volume de 1 mL de suspensão para cada quilo de peso vivo. A administração da droga iniciou-se no 5o dia de gestação estendendo-se até o 20º. O peso das ratas foi aferido diariamente, do dia da cobertura até o dia do parto, a fim de se observar possíveis efeitos tóxicos durante a gestação. Durante a lactação as ratas e os filhotes eram pesados a cada 3 dias para acompanhar o desenvolvimento dos filhotes e recuperação materna. No desmame (21o dia pós-parto) todas as fêmeas foram sacrificadas para a observação dos pontos uterinos de implantação, comparando-se o número de fetos nascidos com o número de fetos gerados. Durante o experimento sete ratas do grupo G IV e uma do G III morreram, além de abortos e sinais de canibalismo na parturição. Observou-se como sinais de toxicidade: diarréia, inapetência, pelos eriçados, taquipnéa e perda de peso. Nenhum filhote apresentou alterações morfológicas macroscópicas. Concluiu-se que o gossipol (GAA) causou intoxicação grave nos animais que receberam maior dosagem, bem como interferiu no ganho de peso das ratas durante a gestação, peso ao nascimento e peso à desmama dos filhotes. Contudo não interferiu no processo de manutenção da gestação e no número de filhotes nascidos quando administrado entre o 5o e 20º dia de gestação. Gossypol toxicity in pregnant and nursing rats – Rattus rattus norvegicus Abstract In order to study the effects of gossypol (C30H30O8) in pregnancy and lactation, 40 female rats (120 days old) weighing between 200 and 250 grams, were used in this experiment. The animals were divided in four different group GI (n = 10), GII (n = 10), GIII (n = 10) and GIV (n = 10). The daily oral dose administered by gavage to the animals of each group was of 0, 20, 40 and 80 mg.kg-1 respectively of gossypol dissolved in canole oil. All rats were weighed daily from the day of copulation (0 day) to the day of parturition, for evaluation of the toxic effects during pregnancy. All rats and their offspring were weighed on days 3, 6, 9, 12, 15, 18 and 21 (day of weaning) of lactation. At the day of weaning, all rats were killed in order to observe the number of uterine implantation sites. Eight rats (1 from GIII group and 7 from GIV) died during the period of drug administration besides the occurrence of abortions and cannibalism at the parturition. Clinical symptoms of toxicity were diarrhea, inappetency, bristly hair, tachypnea, loss of weight. Neither pup displayed either macroscopic or morphological alterations. Heavy poisoning has been induced in rats that received higher doses of gossypol acetate as well as a dose-dependent decrease in body weight gain in pregnant rats, lesser weight at birth and at weaning. Nevertheless, gossypol did not interfere in the maintenance of pregnancy and in the number of pups born when administered from day 5 through day 20 of pregnancy.

Nusantara Bioscience 12: 55-58. Menopause is the transition from productive to non-productive times due to reduced estrogen and progesterone and has an impact on improving cardiovascular problems. The fulfillment of estrogen needs in menopause can use a type of phytoestrogens from black soybeans. The purpose of research is to prove the administration of black soy extract (Glycine soja) can increase the levels of estradiol in the rat blood (Rattus norvegicus) ovariectomy. Design research uses post-test only control group design. Twenty-five female rats were randomly divided into 5 groups (n = 5 tails per group), negative controls, positive and 3 treatments. Ovariectomy was performed in a group of positive controls and treatment. Thirty-day post ovariectomy rats were given appropriate group treatment (negative control, positive, and treatment with the administration of black soy extract dose of 50, 100, 150 mg/200  g/day for 30 days). The estradiol test used the Enzyme-linked immunosorbent assay (ELISA). Anova's test results with a significant degree of α = 0.05 established that the p-value of 0.000 was less than α = 0.05, meaning there was a significant effect of giving black soy extract on increased levels of estradiol in the rat blood ovariectomy. Post - Hoc Tukey testing shows increased the highest estradiol levels in the dose treatment group 100 mg/200  g/day. This research proves that the administration of black soy extract (Glycine soja) can increase the levels of estradiol in the rat blood (Rattus norvegicus) ovariectomy.

Due to their unpredictability and variable effects, injectable anesthetic regimens in laboratory rodent species warrant refinement. In our study we sought to evaluate alfaxalone, which has gained recent popularity in veterinary medicine, alone and in combination with dexmedetomidine to evaluate their anesthetic ability in Sprague–Dawley rats when administered intraperitoneally. Three doses of alfaxalone only and 4 dose combinations of alfaxalone-dexmedetomidine were tested in males and female rats. The time to induction, anesthetic duration, pulse rate, respiratory rate, temperature, and time to recovery were recorded by a blind observer. The level of anesthesia induced by the various anesthetic protocols was assessed by using pedal withdrawal reflex to a noxious stimulus and scored according to the response. Dependent on the treatment group, atipamezole or saline was administered intraperitoneally once animals reached 60 min of anesthesia. Regardless of the dose, alfaxalone alone achieved only a sedative level of anesthesia, whereas all alfaxalone-dexmedetomidine combinations led to a surgical level of anesthesia in all animals. Anesthesia regimens using alfaxalone alone and in combination with dexmedetomidine demonstrated sex-associated differences, with female rats maintaining longer durations of sedation or anesthesia than their male counterparts. Both male and female rats displayed decreases in physiologic parameters consistent with the effects of dexmedetomidine. Given the results described herein, we recommend 20 mg/kg alfaxalone for sedation and 30 mg/kg alfaxalone combined with 0.05 mg/kg dexmedetomidine for surgical anesthesia in female rats. Appropriate doses of alfaxalone only and alfaxalone-dexmedetomidine for male rats were not determined in this study and need further evaluation.

Objective: This study aimed to verify the effect of oral siper-metrin exposure to decrease serum estradiol 17b levels and increased malondialdehyde (MDA) in the uterus level of female Wistar strain rats (Rattus norvegicus).Materials and Methods: The method of this study was true experimental post test only control group in vivo using 24 female rats, divided into 3 groups treated by administering a dose of 5, 10 and 20 mg/kg sipermetrin for 28 days and one control group. Then blood samples were taken from the heart for measurement of serum estradiol 17b levels by ELISA and uterine organs were taken for measurement of Malondialdehyde (MDA) with spectro-photometry method.Results: The results of the measurement of serum estradiol 17b and uterus malondialdehyde (MDA) levels of female Wistar strain rats (Rattus norvegicus) showed an opposite pattern, where there was a decline in serum estradiol 17b levels and an increase in uterus malondialdehyde (MDA) level. There was a significant difference (p=0.000<alpha) in 17b estradiol serum and uterus Malondialdehyde (MDA) levels of female rats between control group and group exposed to sipermetrin treatment for 28 days.Conclusions: Oral sipermetrin exposure can decrease serum levels of estradiol 17b and increase uterine levels of malondi-aldehyde (MDA) of female Wistar strain rats (Rattus norvegicus).

The aims of this research was to determine the density level of lumbal vertebrae on ovariectomized rats administered with Cissus qudrangula Salisb stem extract with various doses. In this research twelve female rats were randomizedly divided into 4 groups with 3 treatment repetition. K1-groups were ovariectomized rats without Cissus quadrangula Salisb trunct extract administration; K2, K3, and K4, were ovariectomized and given with Cissus quadrangula Salisb extract  with 500 mg/kg/Bw, 700 mg/kg/Bw, and 900 mg/kg/Bw as doses perday for 30 days. On day-31, rats were euthanized using chloroform and the os lumbal-5 were collected, followed with histological preparations. Research parameters were the density of active osteoblasts, passive osteoblasts, and osteoclasts. The results showed that the damage in K1 was clearly marked with the high number of osteoclasts, K2 and K3 indicate lower osteoclasts levels than K1 and had repaired bone structure characterized with active cuboidal shaped  osteoblast  activity and passive flat shaped osteoblasts and K4 showed increasing  level of active osteoblasts. The administration of Cissus quadrangula Salisb stem extract at 900 mg/kg/bw for 30 days showed bone repair due to ovariectomy indicated with high level of active osteoblasts.
