Candida auris is a novel and major fungal pathogen that has triggered several outbreaks in the last decade. The few drugs available to treat fungal diseases, the fact that this yeast has a high rate of multidrug resistance and the occurrence of misleading identifications, and the ability of forming biofilms (naturally more resistant to drugs) has made treatments of C. auris infections highly difficult. This review intends to quickly illustrate the main issues in C. auris identification, available treatments and the associated mechanisms of resistance, and the novel and alternative treatment and drugs (natural and synthetic) that have been recently reported.

Acinetobacter baumannii is classified as a top priority pathogen by the World Health Organization (WHO) because of its widespread resistance to all classes of antibiotics. This makes the need for understanding the mechanisms of resistance and virulence critical. Therefore, tools that allow genetic manipulations are vital to unravel the mechanisms of multidrug resistance (MDR) and virulence in A. baumannii. A host of current strategies are available for genetic manipulations of A. baumannii laboratory-strains, including ATCC® 17978TM and ATCC® 19606T, but depending on susceptibility profiles, these strategies may not be sufficient when targeting strains newly obtained from clinic, primarily due to the latter’s high resistance to antibiotics that are commonly used for selection during genetic manipulations. This review highlights the most recent methods for genetic manipulation of A. baumannii including CRISPR based approaches, transposon mutagenesis, homologous recombination strategies, reporter systems and complementation techniques with the spotlight on those that can be applied to MDR clinical isolates.

ABSTRACTMultidrug resistance (MDR) plasmids frequently carry antibiotic resistance genes conferring qualitatively different mechanisms of resistance. We show here that the antibiotic concentrations selecting for the RK2 plasmid inEscherichia colidepend upon the sociality of the drug resistance: the selection for selfish drug resistance (efflux pump) occurred at very low drug concentrations, just 1.3% of the MIC of the plasmid-free antibiotic-sensitive strain, whereas selection for cooperative drug resistance (modifying enzyme) occurred at drug concentrations exceeding the MIC of the plasmid-free strain.

Antibiotics are the ' wonder medicines ' used for battling microbes. Numerous types of antibiotics have been not only used for therapeutic purposes for decades, but have been used prophylactically across other fields such as livestock and animal husbandry. The emergence of multidrug resistance among pathogenic bacteria jeopardizes the importance of antibiotics which have transformed medical sciences before. A growing list of infections, i.e. pneumonia, tuberculosis, and gonorrhea, is becoming more difficult and sometimes impossible to treat as antibiotics become less effective. Antibiotic-resistant infections are correlated with antibiotic intake levels. It is mainly the non-judicial use of antibiotics that makes the bacteria immune. Extensive efforts are required to reduce the rate of resistance by researching emerging microorganisms, mechanisms of resistance and antimicrobial agents. Multidisciplinary strategies are required across health care environments, as well as across sectors of the environment and agriculture. Conservative new approaches including probiotics, antibodies, and vaccines have demonstrated positive results in future trials that suggest the role of these alternative options as preventative or adjunct therapies.

In recent years, multidrug resistance ofEscherichia colihas become a serious problem. However, resistance to fosfomycin (FOM) has been low. We screenedE. coliclinical isolates with reduced susceptibility to FOM and characterized molecular mechanisms of resistance and reduced susceptibility of these strains. Ten strains showing reduced FOM susceptibility (MIC ≥ 8 μg/mL) in 211 clinical isolates were found and examined. Acquisition of genes encoding FOM-modifying enzyme genes (fosgenes) and mutations inmurAthat underlie high resistance to FOM were not observed. We examined ability of FOM incorporation via glucose-6-phosphate (G6P) transporter andsn-glycerol-3-phosphate transporter. In ten strains, nine showed lack of growth on M9 minimum salt agar supplemented with G6P. Eight of the ten strains showed fluctuated induction by G6P ofuhpTthat encodes G6P transporter expression. Nucleotide sequences of theuhpT,uhpA, glpT,ptsI, andcyaAshared several deletions and amino acid mutations in the nine strains with lack of growth on G6P-supplemented M9 agar. In conclusion, reduction ofuhpTfunction is largely responsible for the reduced sensitivity to FOM in clinical isolates that have not acquired FOM-modifying genes or mutations inmurA. However, there are a few strains whose mechanisms of reduced susceptibility to FOM are still unclear.
