Abstract Background Diabetes mellitus is a metabolic disorder of glucose metabolism and management of blood glucose level is the hallmark in the treatment of this disease. The present study investigated chemical composition, in vitro antioxidant and antidiabetic activity of different fractions of 80% methanol Piper guineense leaves extract. Materials and methods The crude methanolic extract of P. guineense was obtained following 80% methanol cold extraction and was successively partitioned with dichloromethane (DCM), ethyl acetate (EtOAc), n-butanol (nBuOH) and aqueous solvents to give four fractions. The chemical composition of the fractions from P. guineense was determined using gas chromatography coupled with mass spectrometry (GC-MS) and their potentials as antioxidant and anti-diabetes were evaluated. Results The percentage yields were 3.16, 2.22, 0.68 and 0.66% (w/w) in n-butanol, DCM, aqueous and ethyl acetate fractions of P. guineense methanolic extract, respectively. The GC-MS analyses identified a total of 71 and 34 phytochemicals in n-butanol and ethyl acetate fractions respectively. Tributyl acetylcitrate (10.95%) and phytol (9.11%) were the major components in the n-butanol fraction while ethyl piperonyl cyanoacetate (27.35%) and phytol (15.17%) were the major constituents in the ethyl acetate fraction. Ethyl acetate fraction had the highest ferric reducing antioxidant power with a value of 53.96 ± 0.40 mgAAE/g while n-butanol fraction possessed highest total antioxidant power (9.98 ± 0.15) followed by aqueous fraction (9.72 ± 0.02). The ethyl acetate and n-butanol fractions with IC50 value of 0.24 ± 0.07 and 0.83 ± 0.15 μg/mL respectively elicited significant inhibitory activities against α-glucosidase while only n-butanol fraction (IC50 = 0.33 ± 0.09 μg/ml) exhibited appreciable inhibition against α-amylase activity. However, none of the four fractions showed significant inhibitory activity towards dipeptidyl-peptidase-IV. Conclusion n-butanol and ethyl acetate fractions of 80% methanol P. guineense leaves extract can be a potential source of bioactive compounds of pharmacological importance in the management of diabetes.

Aims: To characterize the chemical composition of Bryophyllum pinnatum leaf ethyl acetate fraction. Methodology: Quantitative phytochemical composition was assessed using gas chromatography fitted with flame ionization detector (GC-FID), while chemical characterization was via gas chromatography-mass spectrophotometry (GC-MS) analysis. The mass spectra peaks were matched with those found in the National Institute of Standard and Technology (NIST) spectral database. Results: Results revealed a rich presence of proanthocyanin, rutin, quinine, flavan-3-ol, anthocyanin, lunamarin, sapogenin, phenol, flavonones, steroids, epicatechin, kaempferol, phytate, oxalate, resveratol, catechin, flavones, tannin, ribalinidine, naringin, and spartein in varying proportion in B. pinnatum leaves ethylacetate fractions. GC-MS analysis of the fractions revealed the presence of at least 50 constituents. The major constituents were Hexadecanoic acid, methyl ester (24.88%), 10,13-Octadecadienoic acid, methyl ester (29.69%), Tetracosanoic acid, methyl ester (7.84%), Methyl stearate (6.97%), cis-Methyl 11-eicosenoate (6.26%), Methyl 18-methylnonadecanoate (4.99%), Docosanoic acid, methyl ester (3.71%) and  4,7-Methano-1H-indene, octahydro- (2.43%). Conclusion: This rich array of chemical compounds presents B. pinnatum leaves as an important source of potential lead compounds with biological and pharmacological benefits and hence a potential candidate for drug discovery.

The Eugenia genus is rich in bioactive substances with biological potential. Mollicutes are bacteria without cell walls, which are responsible for various human and animal diseases. The aim of this study was to evaluate the chemical composition and antimollicute activity of Eugenia mattosii. Leaves and stems were extracted with methanol, partitioned into fractions of different polarities, and submitted to column chromatography in order to isolate major compounds. Identification and quantification of isolated substances in extracts and fractions were performed by UPLC-PDA/ESI+ - QTOF. Methanolic extracts, ethyl acetate and chloroform fractions and isolated substances were screened for antimycoplasmic activity against five Mycoplasma strains. The methanolic extracts of leaves and stems showed moderate antimycoplasmic activity (MICs = 250-500 μg.mL-1). The fractions exhibited better effect, with MICs = 125-1000 μg.mL-1, especially the ethyl acetate fraction of the leaves, which presented MICs of 125-250 μg.mL-1 for all strains tested. Phytochemical analyses evidenced the presence of some phenolic compounds, including pinostrobin, cryptostrobin and catechin, the first showing promising antimycoplasmic activity. Quantification of the compounds demonstrated higher concentrations of pinostrobin and cryptostrobin in the chloroform fraction. In conclusion, E. mattosii presented antimycoplasmic activity related, at least in part, to the presence of pinostrobin.

The fruit of Rosa multiflora has been used as traditional herbal medicine in Asian countries. The present investigation was undertaken to study the antimicrobial activity and total polyphenol content of hexane, ether, ethyl acetate, water fraction of methanol extract of fruit and flower from Rosa multiflora and Rosa wichuraiana. Antimicrobial activity of the mentioned fractions against 3 gram-positive and 4 gram-negative bacteria using disk diffussion method. The measurement of minimal inhibition concentration (MIC) showed that the ethyl acetate fraction of the two Rosa species is the most effective against the tested bacteria. The total polyphenol content of ethyl acetate fraction of the two Rosa species is higher than the other fractions. The results indicate the antimicrobial activity was related with the total polyphenol content and the fruit and flower of the two Rosa species can be considered as a natural source of antimicrobial agents.

This study aims to find the treatment of diabetes using natural materials by exploring plants in the province of East Nusa Tenggara. his research was conducted out by extracting the Strychnos nitida G.Don stem using a method of maceration by ethanol 70%. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Simplicia from maceration and fractionation results were then tested for antioxidant activity, α-glucosidase inhibition activity and identification of active compounds. The results showed that ethyl acetate fraction had the lowest IC50 value of 86.83 μg / ml. Results of the α-glucosidase activity test showed that ethyl acetate fraction and n-heksan fraction at 900 ppm had the highest percentage of inhibition of 34.23% and 33.89%. Identification using LCMS/MS method showed that ethyl acetate fraction consist of Benzenemethamine, N, N-dioctyl- as an antioxidantcompound and compound 24-methyl-5-cholestone-hexol as an antidiabetic compound. From the results of this study, we concluded that the extract of kayu ular Strychnos nitida G.Don stem has inhibition activity toward α-glucosidase enzyme.
