Litter decomposition is the key link between material circulation and energy flow in ecosystems, resulting from the activity of resident microbes and various enzymes. This study investigated enzyme activity in litter and associated microbial community characteristics to help clarify the internal mechanisms associated with litter decomposition, while also providing researchers a scientific basis for soil remediation in mining areas. Results confirmed that the nutrient content of Bothriochloa ischaemum litter significantly increased as phytoremediation years progressed, while enzyme activities in litter varied over different phytoremediation years. During the litter decomposition process, cellulase predominated in the early phytoremediation stage and catalase predominated in the intermediate phytoremediation stage. Obvious differences were found in bacterial community structure and diversity over progressive phytoremediation years. Predominant bacterial genera mainly included Massilia, Sphingomonas, Curtobacterium, Amnibacterium, and Methylobacterium. Moreover, Methylorosula and Jatrophihabitans had relatively higher betweenness centrality, and played important roles in bacterial community positive interactions. Additionally, total nitrogen (TN) and total zinc in soil, sucrase and catalase activity in litter were the main environmental factors that affected the structural framework of bacteria in B. ischaemum litter. However, TN had the greatest overall effect on the structural framework of bacteria in litter. Results from this study can help our understanding of the role that litter plays in degraded ecosystems. Our results also provide a scientific basis for improving poor quality soil in areas affected by copper tailings while also amending ecological restoration efficiency.

AbstractThe vermicompost collected from dewatered domestic sludge as packing material in biofilter was investigated for hydrogen sulfide (H2S) removal. No nutrients or microbial inoculation was added throughout the experiment. The corresponding bacterial community characteristics in the vermicompost biofilter of different spatial levels were evaluated by Miseq high-throughput sequencing technique. The results showed that the vermicompost biofilter performed well during operation. The H2S removal efficiency reached nearly 100% under condition of the inlet concentration <350 mg m−3 and 0.25−0.35 m3 h−1 gas flow rate. The maximum elimination capacity of 20.2 g m−3 h−1 was observed at a flow rate of 0.35 m3 h−1. Furthermore, the amounts of biodegraded products and pH varied accordingly. In addition, the results from high-throughput sequencing revealed pronouncedly spatial variation of the vermicompost, and the Rhodanobacter, Halothiobacillus, Mizugakiibacter as well as Thiobacillus, which can play an important role in removing H2S, were predominant in the final vermicompost. These results imply that the vermicompost with diverse microbial communities has a good potential for eliminating H2S.

Plant and microbiome interactions are necessary for plant nutrient acquisition. However, relatively little is known about the responses of roots, bulk, and rhizosphere soil microbial communities in different artificial vegetation types (woody and herbaceous) in riparian areas of massive dams and reservoirs. Therefore, this study aims to assess such responses at elevations of 165–170 m a.s.l. in the riparian zones of the Three Gorges Dam Reservoir, China. The samples were collected containing the rhizosphere soil, bulk soil, and roots of herbaceous and woody vegetation at different emergence stages in 2018. Then, all the samples were analyzed to quantify the soil properties, bacterial community characteristics, and their interaction in the early and late emergence phases. In different periods, the weight of dominant soil bacteria, including Proteobacteria, Acidobacteria, Actinobacteria, Chloroflexi, and Cyanobacteria, was higher, and their composition was different in the rhizosphere, bulk soil, and endophytes. Moreover, the soil co-occurrence networks indicated that the weight of soil physical properties was higher than chemical properties in the early emergence stage. In contrast, the weight of chemical properties was relatively higher in the late emergence stage. Furthermore, the richness and diversity of the bacterial community were mainly affected by soil organic matter. This study suggests that these herbaceous and woody vegetation are suitable for planting in reservoir areas affected by hydrology and human disturbance in light of soil nutrients and soil microbial communities, respectively. Additionally, these results provide valuable information to inoculate the soil with key microbiota members by applying fertilizers, potentially improving plant health and soil production.

To understand dynamic changes in rhizosphere microbial community in consecutive monoculture, Illumina MiSeq sequencing was performed to evaluate the V3-V4 region of 16S rRNA in the rhizosphere of newly planted and three-year ratooning sugarcane and to analyze the rhizosphere bacterial communities. A total of 126,581 and 119,914 valid sequences were obtained from newly planted and ratooning sugarcane and annotated with 4445 and 4620 operational taxonomic units (OTUs), respectively. Increased bacterial community abundance was found in the rhizosphere of ratooning sugarcane when compared with the newly planted sugarcane. The dominant bacterial taxa phyla were similar in both sugarcane groups. Proteobacteria accounted for more than 40% of the total bacterial community, followed by Acidobacteria and Actinobacteria. The abundance of Actinobacteria was higher in the newly planted sugarcane, whereas the abundance of Acidobacteria was higher in the ratooning sugarcane. Our study showed that Sphingomonas, Bradyrhizobium, Bryobacter, and Gemmatimonas were dominant genera. Moreover, the richness and diversity of the rhizosphere bacterial communities slightly increased and the abundance of beneficial microbes, such as Bacillus, Pseudomonas, and Streptacidiphilus, in ratooning sugarcane were more enriched. With the consecutive monoculture of sugarcane, the relative abundance of functional groups related to energy metabolism, glycan biosynthesis, metabolism, and transcription were overrepresented in ratooning sugarcane. These findings could provide the way for promoting the ratooning ability of sugarcane by improving the soil bacterial community.
