AbstractThe present study was carried out to investigate how plant growth-promoting bacteria (PGPB) influence plant growth and uptake of boron (B) and phosphorus (P) in rapeseed (Brassica napus). Rapeseed was subjected to control, B, P and B + P treatments, either with or without B. pumilus (PGPB) inoculation, and grown in pot culture for 6 weeks. In the absence of B. pumilus, the addition of B, P or both elements improved the growth of rapeseed compared with the control. Interestingly, B. pumilus inoculation inhibited plant growth and enhanced B uptake under B and B + P but not under control and P conditions. In addition, B. pumilus inoculation decreased the pH of soil under B and B + P supplies. Bacillus pumilus inoculation thus increased rapeseed B uptake and inhibited growth under B supply, which suggests that the effects of PGPB on rapeseed growth depend on the addition of B to soil. Bacillus pumilus inoculation may therefore be recommended for the enhancement of rapeseed B levels in B-deficient soils but not in B-sufficient ones.

Growth of canola (Brassica napus) seeds treated with plant growth-promoting bacteria in copper-contaminated and polycyclic aromatic hydrocarbon (PAH)-contaminated soils was monitored. Pseudomonas asplenii AC, isolated from PAH-contaminated soil, was transformed to express a bacterial gene encoding 1-aminocyclopropane-1-carboxylate (ACC) deaminase, and both native and transformed bacteria were tested for growth promotion. Inoculation of seeds, grown in the presence of copper or creosote, with either native or transformed P. asplenii AC significantly increased root and shoot biomass. Native and transformed P. asplenii AC and transformed P. asplenii AC encapsulated in alginate were equally effective at promoting plant growth in copper-contaminated soils. In creosote-contaminated soils the native bacterium was the least effective, and the transformed encapsulated bacterium was the most effective in growth promotion.Key words: plant growth-promoting bacteria, phytoremediation, copper, polycyclic aromatic hydrocarbons, Brassica napus, ethylene, alginate encapsulation.

Plant growth promoting bacteria (PGPB) are a growing subset of agricultural adjuncts which can be used to increase crop yield and plant productivity. Although substantial research has been conducted on the metabolites and active molecules secreted by PGPBs, relatively little is known about their effects on the global transcriptome of the host plant. The present study was carried out to investigate changes in the gene expression landscape of early vegetative Brassica napus following treatment with Pseudomonas chlororaphis PA23. This PGPB was isolated from the soybean rhizosphere and has been extensively studied as a biocontrol agent. However, little is known about its effects on plant growth and development. Using a combination of RNA-sequencing and physiological analyses, we identified increased abundance of mRNA transcripts associated with photosynthesis and phytohormone response. Phenotypically we observed increased photosynthetic rates and larger root and shoot systems in B. napus following P. chlororaphis PA23 treatment. Lastly, we identified auxin production by P. chlororaphis PA23 which likely contributes to changes in gene expression and the observed phenotypic differences in root and shoot structures. Together, the results of our study suggest that PA23 is a potent plant growth promoting agent with the potential for field applications as an agricultural adjunct.

Plants in association with plant growth-promoting rhizobacteria can benefit from lower plant ethylene levels through the action of the bacterial enzyme 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase. This enzyme cleaves the immediate biosynthetic precursor of ethylene, ACC. Ethylene is responsible for many aspects of plant growth and development but, under stressful conditions, it exacerbates stress symptoms. The ACC deaminase-containing bacterium Pseudomonas putida UW4 is a potent plant growth-promoting strain and, as such, was used to elaborate the detailed role of bacterial ACC deaminase in Brassica napus (canola) plant growth promotion. Transcriptional changes in bacterially treated canola plants were investigated with the use of an Arabidopsis thaliana oligonucleotide microarray. A heterologous approach was necessary because there are few tools available at present to measure global expression changes in nonmodel organisms, specifically with the sensitivity of microarrays. The results indicate that the transcription of genes involved in plant hormone regulation, secondary metabolism, and stress response was altered in plants by the presence of the bacterium, whereas the upregulation of genes for auxin response factors and the downregulation of stress response genes was observed only in the presence of bacterial ACC deaminase. These results support the suggestion that there is a direct link between ethylene and the auxin response, which has been suggested from physiological studies, and provide more evidence for the stress-reducing benefits of ACC deaminase-expressing plant growth-promoting bacteria.
