Uptake and Effects of Five Heavy Metals on Seed Germination and Plant Growth in Alfalfa ( Medicago sativa L.)

Recently, heavy metals concentrations increased in some agricultural areas due to the consequences of anthropogenic impacts. The aim of this study was to determine the level of heavy metals (As, Cr, Ni and Pb) in Medicago sativa L. and Trifolium pratense L. grown on fluvisol, in order to obtain information on safety of these nutrients. The total content of Pb, As, Cr and Ni in the samples of fluvisol was above the maximum allowable amount. The content of heavy metals in Medicago sativa L. and Trifolium pratense L. was below the critical and toxic concentrations in all samples originating from contaminated soil. It was concluded that the accumulation of heavy metals in plants did not depend only on the total content in soil, but also the affinity of the plant, and individual and interactive effects of various soil properties. No statistically significant differences in the accumulation of heavy metals between Medicago sativa L. and Trifolium pratense L were observed. It is necessary to further control of heavy metals in the investigated area, in order to prevent their entry into the food chain and provide healthy food.

Genotypic variations of alfalfa (Medicago sativa L.) to both phosphorus (P) deficiency and water deficiency are evident on the Loess Plateau of China. Here, we compare the adaptive mechanisms between an introduced cultivar (Arkaxiya) and a landrace (Longzhong) subjected to P- and water-limited conditions. The two genotypes were grown in a soil medium with 0, 4.2, 8.4 and 16.8 μg applied P per gram dry soil. Three water treatments were imposed (maintained at 75–90%, 45–55% and 30–35% of pot capacity (PC)) 28 days after sowing (DAS). At high soil P and high soil water content (SWC), high rates of net photosynthesis (Pn) contributed to greater plant growth and P-use efficiency (PUE) in the introduced Arkaxiya compared to the landrace Longzhong. However, at low SWC, Longzhong had enhanced antioxidative defense (mainly SOD and CAT) compared to Arkaxiya. In addition, shorter shoot length and greater branching in Longzhong than Arkaxiya may also facilitate adaptation to low SWC. The contrasting adaptive mechanisms of the two genotypes provide a number of early-screening parameters associated with plant growth for the selection and introduction of alfalfa targeted at different rainfall and available P environments.

The contents of heavy metals and trace elements and their accumulation in Medicago sativa L., cultivated on Eutric cambisol along the E75 route section Belgrade-Leskovac, were examined in order to assess the health and safety of animal feed. The samples of soil and aerial parts of the plant material were collected from both sides of lanes at 10, 30, 50 and 400 m perpendicular to the direction of the highway. Soil and plant analyses of the metals content were done according to ICP methodology. The results showed that at the locality L 14, a distance of 30 and 50 meters away from the lanes, the content of total forms of Cr, Ni and Pb in soil was above the maximum permissible concentration. In the plant biomass it was determined the following: in a sample from the location L 14 at a distance of 50 meters from the lanes concentrations of Ni and Co were higher than normal values, and concentrations of Fe and Pb were above toxic levels or maximum tolerance levels for animal feed; determined Fe content in the sample of alfalfa at location L 11, 400 m away from the lanes, and Ni in the sample from the site D 12 at a distance of 50 m from the lanes, was above the normal values, while in the sample from D12 location, at a distance of 30 m from the lanes, the content of Pb was above the toxic levels or maximum tolerance levels for animal feed. The results suggest a caution in the use of alfalfa, grown near the highway route, for animal feed because of the potential entry of heavy metals into the food chain.

Allelopathic effects of cold water extracts of Cuscuta epithymum L. on seed germination and initial development of Medicago sativa L. varieties were investigated under laboratory conditions at the Institute of Forage Crops, Pleven, during 2016-2017. It was found that the water extracts from dry biomass of C. epithymum had a considerably stronger inhibitory effect on the studied M. sativa varieties (IR 32.7-100.0 %), as compared to the extracts of fresh biomass (IR 0.2-40.5%). Depending on the kind of C. epithymum extract (fresh or dry parasitic weed biomass), IRs for seed germination of the tested M. sativa varieties could be conventionally classified into five groups: 1) seed germination stimulation, -1.4% - extracts from fresh biomass affecting the variety ?Multifoliolate?; 2) seed germination inhibition of 0.1-10% - extracts prepared from fresh biomass affecting the varieties ?Prista 3?, ?Pleven 6?, ?Prista 5? and ?Obnova?; 3) seed germination inhibition of 11-20% - extracts from fresh biomass affecting the varieties ?Roly? and ?Victoria?; 4) seed germination inhibition of 30-45% - extracts from dry biomass affecting the variety ?Multifoliolate?; 5) seed germination inhibition of 46-60% - extracts from dry biomass affecting the varieties ?Prista 5?, ?Prista 3?, ?Victoria?, ?Roly?, ?Dara?, ?Pleven 6? and ?Obnova?. The varieties ?Victoria?, ?Prista 5? and ?Multifoliolate? of M. sativa possess some allelopathic tolerance because their germination indexes (GI) range from 80.5 to 88.7 % for the extracts prepared from fresh weed biomass of C. epithymum, and from 47.1 to 48.6% for the extracts from dry weed biomass, compared to control treatment. These varieties can be used as components in future breeding programmes.
