Random amplified polymorphic DNA (RAPD) markers were identified for self-incompatibility (SI) alleles that will allow marker-assisted selection of desired S-alleles and assist in cloning the locus responsible for the sporophytic SI displayed in hazelnut (Corylus avellana L.). DNA was extracted from young leaves collected from field-planted parents and 27 progeny of the cross OSU 23.017 (S1 S12) × VR6-28 (S2 S26). Screening of 10-base oligonucleotide RAPD primers was performed using bulked segregant analysis. DNA samples from six trees each were pooled into four “bulks,” one for each of the following: S1 S2, S1 S26, S2 S12, and S12 S26. “Super bulks” of twelve trees each for S1, S2, S12, and S26 then were created for each allele by combining the appropriate bulks. The DNA from these four super bulks and also the parents was used as a template in the PCR assays. Amplification products were electrophoresed on 2% agarose gels and photographed under UV light after ethidium bromide staining. 200 primers were screened and one RAPD marker each was identified for alleles S2 (OPI-07700) and S1 (OPJ-141700).

A linkage map for European hazelnut (Corylus avellana L.) was constructed using random amplified polymorphic DNA (RAPD) and simple sequence repeat (SSR) markers and the 2-way pseudotestcross approach. A full-sib population of 144 seedlings from the cross OSU 252.146 × OSU 414.062 was used. RAPD markers in testcross configuration, segregating 1:1, were used to construct separate maps for each parent. Fifty additional RAPD loci were assigned to linkage groups as accessory markers whose exact location could not be determined. Markers in intercross configuration, segregating 3:1, were used to pair groups in one parent with their homologues in the other. Eleven groups were identified for each parent, corresponding to the haploid chromosome number of hazelnut (n = x = 11). Thirty of the 31 SSR loci were able to be assigned to a linkage group. The maternal map included 249 RAPD and 20 SSR markers and spanned a distance of 661 cM. The paternal map included 271 RAPD and 28 SSR markers and spanned a distance of 812 cM. The maps are quite dense, with an average of 2.6 cM between adjacent markers. The S-locus, which controls pollen-stigma incompatibility, was placed on chromosome 5S where 6 markers linked within a distance of 10 cM were identified. A locus for resistance to eastern filbert blight, caused by Anisogramma anomala, was placed on chromosome 6R for which two additional markers tightly linked to the dominant allele were identified and sequenced. These maps will serve as a starting point for future studies of the hazelnut genome, including map-based cloning of important genes. Ptychobothridean genera like Senga and Circumoncobothrium are the common parasites of fresh water fishes. The genotypic study of these parasites was taken by RAPD. The RAPD profile of these two parasites were not similar to each other as depicted by the band pattern in picture. These results suggest the presence of inter-specific polymorphism among cestode parasites of two different genera for RAPD analysis. The present study demonstrated that genetic differentiation of cestode parasites could be accomplished on the basis of genomic variation with polymorphic band pattern using RAPD. All the detected bands (PCR product) were polymorphic and band size ranged from 500-5000 bp in length. The RAPD of profiles using GBO-31, GBO-32, GBO-33, GBO-34, GBO-35 and GBO-36. Primers were able to characterize inter-specific polymorphism among the two genus ( Senga and Circumoncobothrium ). Genetic analysis suggests that Senga and Circumoncobothrium show genetic diversity with respect to RAPD patterns using all the six primers used for the present study. The genetic distance between the analyzed genuses ranged from 0.14 to 0.80. The differentiation of the two parasites on the basis of genetic markers could greatly facilitate study on the biology of these parasites.

Показана актуальность изучения экологической пластичности древесных растений в связи с необходимостью подбора адаптированного видового состава деревьев и кустарников для защитных лесных насаждений в условиях климатических изменений. Установлено, что экологическая пластичность и пределы толерантности растений связаны со свойством организмов адаптироваться к тому или иному диапазону факторов среды. В статье представлен таксономический состав орехоплодных культур коллекций ФНЦ агроэкологии РАН. Это шесть видов рода Juglans: J. regia, J. mandshurica, J. cinerea, J. rupestris, J. ailanthifolia, J. nigra и три вида рода Corylus: С. avellana L., С. аmericana W., сорта С. pontica C. Koch Президент, Футкурами, Черкесский2. Приведены сведения о положительном опыте культивирования Corylus avellana L. в плантационных насаждениях (1,6 га, посадка рядовая, схема размещения 55 м) в условиях южных черноземов. Цель исследований изучить экологическую пластичность орехоплодных кустарников коллекций ФНЦ агроэкологии РАН. На примере родового комплекса Corylus получены материалы по экологической пластичности орехоплодных кустарников в условиях каштановых (кадастр. 34:36:0000:14:0178), светлокаштановых почв (34:34:000000:122 34:34:060061:10). Установлены уровни изменчивости морфологических признаков ассимиляционного аппарата и плодов С. avellana L., сортов С. pontica C. Koch., которые согласуются с зимо и засухоустойчивостью. Кластерный анализ позволил выявить корреляцию признаков при 5 уровне значимости. Для целей защитного лесоразведения и озеленения засушливых районов рекомендованы Corylus avellana и Черкесский2 с выраженной вариабельностью морфологических признаков, которая указывает на их широкую экологическую валентность и адаптационные возможности в рассматриваемых условиях. The urgency of studying the ecological plasticity of woody plants in connection with the need to select an adapted species composition of trees and shrubs for protective forest plantations in the context of climate change is shown. It is established that the ecological plasticity and tolerance range of plants is associated with the ability of organisms to adapt to a particular range of environmental factors. The article presents the taxonomic composition of nut crops in the collections of FSC Agroecology RAS. Six species of Juglans: J. regia, J. mandshurica, J. cinerea, J. rupestris, J. ailanthifolia, J. nigra and three species of the genus Corylus: С. avellana L., С. аmericana W., varieties of C. pontica C. Koch the President, Futkurami, Circassian2. Data on positive experience of cultivation of Corylus avellana L. in plantation plantings (1,6 hectares, landing ordinary, the scheme of placement of 55 m) in the conditions of southern chernozems are given. The purpose of the research is to study the ecological plasticity of nutbearing shrubs of the collections of the Federal scientific center for Agroecology Russian Academy of Sciences. For example, a generic complex Corylus submissions received on the environmental plasticity of nut bushes in the conditions of brown (the cadaster nubmer 34:36:0000:14:0178), light chestnut soils (34:34:000000:122 34:34:060061:10). The levels of variability of morphological features of the assimilation apparatus and fruits of Corylus avellana L., С. pontica C. Koch varieties were established., which are consistent with winter and drought resistance. Cluster analysis revealed the correlation of features at 5 significance level. Corylus avellana and Circassian2 with a pronounced variability of morphological features, which indicates their broad ecological valence and adaptation capabilities in the conditions under consideration, are recommended for the purposes of protective afforestation and greening of dry areas.
