Abstract Resistance to citrus tristeza virus (CTV) was evaluated in 554 progeny of 10 populations derived from Poncirus trifoliata. A dominant gene (Ctv) controlled CTV resistance in P. trifoliata. Twenty-one dominant PCR-based DNA markers were identified as linked to Ctv by bulked segregant analysis. Of the 11 closest markers to Ctv, only 2 segregated in all populations. Ten of these markers were cloned and sequenced, and codominant RFLP markers were developed. Seven RFLP markers were then evaluated in 10 populations. Marker orders were consistent in all linkage maps based on data of single populations or on combined data of populations with similar segregation patterns. In a consensus map, the six closest marker loci spanned 5.3 cM of the Ctv region. Z16 cosegregated with Ctv. C19 and AD08 flanked Ctv at distances of 0.5 and 0.8 cM, respectively. These 3 markers were present as single copies in the Poncirus genome, and could be used directly for bacterial artificial chromosome library screening to initiate a walk toward Ctv. BLAST searches of the GenBank database revealed high sequence similarities between 2 markers and known plant disease resistance genes, indicating that a resistance gene cluster exists in the Ctv region in P. trifoliata.

`Chandler' pummelo [Citrus maxima (Burm.) Merrill] was found to be citrus tristeza virus (CTV)–resistant. The inheritance of this resistance in 84 progeny of two crosses derived from `Chandler' pummelo and trifoliate orange [Poncirus trifoliata (L.) Raf.] was controlled by a single dominant gene designated Ctv2. Progeny analysis of four molecular markers closely linked to the Ctv gene, which confers resistance to CTV in trifoliate orange, demonstrated that Ctv2 was an independently assorting gene from Ctv.

Citrus tristeza virus (CTV) [genus Closterovirus; family Closteroviridae] is one of the most important, economically devastating viruses of citrus worldwide. On citrus trees grafted onto sour orange rootstock, typical CTV symptoms include dieback and defoliation, stunting, curling and chlorotic leaves, stem-pitting, and pinholes below the bud union on the inner face of the bark (Moreno et al. 2008). This single-stranded, positive-sense RNA virus is most efficiently transmitted by the brown citrus aphid (Toxoptera citricida), but it can also be transmitted by other aphid species and through grafting of infected plant material onto healthy plants (Moreno et al 2008; Herron et al. 2006). In Fall 2020, leaf material for virus testing was collected from 13 navel orange trees (Citrus × sinensis) grafted onto Poncirus trifoliata rootstocks (including ‘Flying Dragon’) located in a citrus research orchard in Tifton, GA. Trees ranged in age from 2 to 10 years, with the younger trees having been grafted from cuttings taken from the older trees. The oldest of these trees was derived from cuttings taken in 2009 from an orange tree growing locally in a residential yard in Tifton; this parent tree was more than 15 years old when these cuttings were obtained and was no longer available for sampling as of 2020. Symptoms or other visible signs of disease had not been noted on any of the tested trees, and trees were chosen for testing prior to the further dissemination of this plant material. The presence of CTV was verified via molecular and serological testing. CTV infection was initially confirmed in 8 of 13 tested samples using the ImmunoStrip® for CTV assay (Agdia® Inc., Elkhart, IN, cat no: ISK 78900/0025) according to the manufacturer’s instructions. RNA was extracted from leaf material collected from the 13 sampled trees using the RNeasy Plant Mini Kit (Qiagen, Valencia, CA). Following cDNA synthesis, samples were tested for the presence of CTV by reverse-transcription PCR using primer pair AR18F (5’-ATGTCAGGCAGCTTGGGAAATT-3’) and AR18R (5’-TTCGTGTCTAAGTCRCGCTAAACA-3’) which produces a 511 bp amplicon (Roy et al., 2005). PCR reactions confirmed the presence of CTV, with the same eight samples that had previously tested positive via Immunostrip® producing PCR fragments of the expected size. Amplified products from two of these samples were then sequenced using Sanger sequencing (Retrogen Inc, San Diego, CA, USA) and subjected to BLAST analysis (https://blast.ncbi.nlm.nih.gov/Blast.cgi) for further identification. Sequence analysis revealed that the obtained partial sequences (MW540805) from the p18 gene of both isolates were 100% identical to one another and shared 100% identity to corresponding sequences from CTV strain N4 (MK779711.1). To the best of our knowledge, this is the first report of CTV infecting citrus plants in Georgia. CTV could pose an imminent threat to the emerging citrus industry in Georgia if it were to become established in commercial citrus plantings either via the dissemination of infected plant material or via vector transfer of the virus under field conditions. While the brown citrus aphid is not known to be widespread in Georgia at this time, other CTV vectors are prevalent including the cotton aphid (Aphis gossypii) and the black citrus aphid (T. aurantia). Georgia citrus growers and plant propagators should be aware of this virus and take appropriate control measures to prevent the spread of this viral diseas.

The citrus tristeza virus resistance gene (Ctv) is a single dominant gene in Poncirus trifoliata, a sexually compatible relative of citrus. To clone this gene, a bacterial artificial chromosome (BAC) library has been constructed from an individual plant that was homozygous for Ctv. This library contains 45 696 clones with an average insert size of 80 kb, corresponding to 9.6 genome equivalents. Screening of the BAC library with five chloroplast DNA probes indicated that 0.58% of the BAC clones contained chloroplast-derived inserts. The chromosome walk across the Ctv locus was initiated using three closely linked genetic markers: C19, AD8, and Z16. The walk has been completed and a contig of ca. 1.2 Mb was constructed. Based on new data, the genetic map in the Ctv region was revised, with Ctv being located between AD8-Z16 and C19 at distances of 1.2 and 0.6 cM, respectively. Utilizing DNA fragments isolated from the contig as RFLP markers, the Ctv locus was further mapped to a region of ca. 300 kb. This contig contains several putative disease-resistance genes similar to the rice Xa21 gene, the tomato Cf-2 gene, and the Arabidopsis thaliana RPS2 gene. This library will therefore allow cloning of Ctv and other putative disease-resistance genes.Key words: Poncirus, citrus tristeza virus, chromosome walk, resistance gene.

O objetivo deste trabalho foi avaliar a produtividade e as características agronômicas de laranjeira 'Valência', enxertadas em porta-enxertos de híbridos de trifoliata (Poncirus trifoliata). A produção de frutos, a de sólidos solúveis totais por planta, as dimensões e a eficiência produtiva de copas de laranjeira 'Valência', enxertadas em 13 híbridos de trifoliata, cultivados sem irrigação, foram avaliados por períodos que variaram de três a oito anos. As plantas também foram avaliadas, visualmente, quanto à manifestação dos sintomas de tristeza (Citrus tristeza virus) e de declínio-dos-citros, e foi utilizado o teste diagnóstico "dot immunobinding assay" (DIBA), para detecção da ocorrência do declínio antes do aparecimento dos sintomas. As plantas tinham oito anos de idade, no início das avaliações. Verificou-se que o citrandarin 'Sunki' x 'English' induziu as maiores produções de frutos em oito colheitas, sem diferir significativamente do citrange 'Troyer'. Em três anos de análise, o citrandarin 'Sunki' x 'English', sem diferir dos citranges 'Troyer' e 'Carrizo', também induziu as maiores produções de frutos e sólidos solúveis por planta. O citrentin 'Clementina' x trifoliata, os citrandarins 'Cleópatra' x 'Swingle' (715) e (1.614), 'Cleópatra' x 'Rubidoux' (1.600) e 'Cleópatra' x 'Christian' induziram a formação de laranjeiras da cultivar Valência com alturas iguais ou inferiores a 2,5 m. Nenhuma das plantas apresentou sintomas de tristeza ou declínio-dos-citros. Foi constatada a incompatibilidade entre a cultivar Valência e o trangpur 'Cravo' x 'Carrizo'.
