The increase of temperature might affect the distribution and reproduction of seagrass. This research aims to determine the seagrass bed coverage and the ecosystem condition. Three line transects were established perpendicular to the coastal line with the distance of 50-100 m, or up to the border of the intertidal area. In each transect, sampling points were determined with a distance of 10-20 m. At the sampling points, a plot of 50 cm x 50 cm was established to measure the coverage percentage of seagrass vegetation. The seagrass species were also observed and recorded along the line transects. The percentage of seagrass coverage was measured using a method from Saito and Atobe (1994). The results showed that the coastal area of Jelenga has the highest percentage of seagrass coverage (>60%, healthy) among other coastal areas. This may be caused by the characteristic of Jelenga coast which was relatively calm, few visitors, low water turbidity, and high light penetration. While, other transects have percentage coverage of less than 60% (less healthy). There was one transect on Maluk coast which has coverage percentage of less than 29% (lack of seagrass species). The small coverage percentage on Maluk coast can be caused by the high number of visitors and high activity of fishing boats around the coast which results in high turbidity. Abstract Lucinid bivalves harbor environmentally acquired, chemosynthetic, gammaproteobacterial gill endosymbionts. Lucinid gill microbiomes, which may contain other gammaproteobacterial and/or spirochete taxa, remain under-sampled. To understand inter-host variability of the lucinid gill microbiome, specifically in the bacterial communities, we analyzed the microbiome content of Stewartia floridana collected from Florida. Sampled gills contained a monospecific gammaproteobacterial endosymbiont expressing lithoautotrophic, mixotrophic, diazotrophic, and C1 compound oxidation-related functions previously characterized in similar lucinid species. Another low-abundance Spirochaeta-like species in ∼72% of the sampled gills was most closely related to Spirochaeta-like species in another lucinid Phacoides pectinatus and formed a clade with known marine Spirochaeta symbionts. The spirochete expressed genes were involved in heterotrophy and the transport of sugars, amino acids, peptides, and other substrates. Few muscular and neurofilament genes from the host and none from the gammaproteobacterial and spirochete symbionts were differentially expressed among quadrats predominantly covered with seagrass species or 80% bare sand. Our results suggest that spirochetes are facultatively associated with S. floridana, with potential scavenging and nutrient cycling roles. Expressed stress- and defense-related functions in the host and symbionts also suggest species-species communications, which highlight the need for further study of the interactions among lucinid hosts, their microbiomes, and their environment.

AbstractSeagrass conservation and management requires scientific understanding of spatial and temporal variability, information that is currently limited for the Eastern Tropical Pacific (ETP). Here, we analysed seagrass presence based on previous reports, herbarium collections and stakeholder knowledge, combined with field characterization in Golfo Dulce, southern Pacific coast of Costa Rica. Seagrasses were found at multiple locations along a narrow border close to shore and in up to 6 m depth within Golfo Dulce, dating back to 1969. Two seagrass species were found, Halophila baillonii and Halodule beaudettei. Seagrass biomass values for Golfo Dulce (12.0 ± 8.5 g DW m−2) were lower and water nutrient concentrations were higher than previously reported in the gulf. Shoot density (1513 ± 767 shoots m−2) was similar to previous reports. Stable isotope values in seagrass were −11.3 ± 1.0‰ δ13C and 1.2 ± 0.9‰ δ15N; while those in sediments were −26.1 ± 1.3 and 2.5 ± 0.9‰. In Golfo Dulce, isotopic values of both seagrass species do not overlap with other known primary producers. Management strategies should aim to minimize known seagrass stressors, protect potential seagrass habitat, and take into account the dynamic life strategies of the two seagrass species found.

In India 14 seagrass species can be found with monospecific genera (Enhalus, ThalassiaandSyringodium),Cymodoceawith two species andHalophilaandHalodulerepresented by more than two taxonomically complex species. Considering this, the present study was made to understand the level and pattern of genetic variability among these species collected from Tamilnadu coast, India. Random amplified polymorphic DNA (RAPD) analysis was used to evaluate the level of polymorphism existing between the species. Out of the 12 primers tested, 10 primers amplified 415 DNA fragments with an average of 41.5 fragments per primer. Of the total 415 amplified fragments only 123 (29.7%) were monomorphic and the remaining 292 (70.3%) were polymorphic for Indian seagrass species. Among the 10 primers used four are identified as the key primers capable of distinguishing all the Indian seagrasses with a high degree of polymorphism and bringing representative polymorphic alleles in all the tested seagrasses. From the present investigation, this study shows that the RAPD marker technique can be used not only as a tool to analyse genetic diversity but also to resolve the taxonomic uncertainties existing in the Indian seagrasses. The efficiency of these primers in bringing out the genetic polymorphism or homogeneity among different populations of theHalophilaandHalodulecomplex still has to be tested before recommending these primers as an identification tool for Indian seagrasses.
