A field experiment was conducted during kharif, 2005 at Agricultural Research station, Devihosur, Haveri, Karnataka to assess the progress of powdery mildew at different time interval of sowing dates. Totally 20 different dates of sowings were imposed in the experiment at an interval of 10 days. The crop sown on last week of May to mid of June recorded minimum disease severity compared to rest of the date of sowings. This clearly indicated that crop sown during this period suffers less, which may be due to low inoculum potential, whereas late sown crop suffers more because of the readily available inoculum in the early sown crops. Low disease severity in last week of May to mid of June sowing may be attributed to the non-congenial weather factors for the development of the disease.

Abstract Synthetic transcription factors have great promise as tools to help elucidate relationships between gene expression and phenotype by allowing tunable alterations of gene expression without genomic alterations of the loci being studied. However, the years-long timescales, high cost, and technical skill associated with plant transformation have limited their use. In this work we developed a technology called VipariNama (ViN) in which vectors based on the Tobacco Rattle Virus (TRV) are used to rapidly deploy Cas9-based synthetic transcription factors and reprogram gene expression in planta. We demonstrate that ViN vectors can implement activation or repression of multiple genes systemically and persistently over several weeks in Nicotiana benthamiana, Arabidopsis (Arabidopsis thaliana), and tomato (Solanum lycopersicum). By exploring strategies including RNA scaffolding, viral vector ensembles, and viral engineering, we describe how the flexibility and efficacy of regulation can be improved. We also show how this transcriptional reprogramming can create predictable changes to metabolic phenotypes, such as gibberellin biosynthesis in N. benthamiana and anthocyanin accumulation in Arabidopsis, as well as developmental phenotypes, such as plant size in N. benthamiana, Arabidopsis, and tomato. These results demonstrate how ViN vector-based reprogramming of different aspects of gibberellin signaling can be used to engineer plant size in a range of plant species in a matter of weeks. In summary, VipariNama accelerates the timeline for generating phenotypes from over a year to just a few weeks, providing an attractive alternative to transgenesis for synthetic transcription factor-enabled hypothesis testing and crop engineering.

In many circles, brown coal continues to be viewed as a cheap source of energy, resulting in numerous investments in new opencast brown coal mines. Such a perception of brown coal energy is only possible if the external costs associated with mining and burning coal are not considered. In past studies, external cost analysis has focused on the external costs of coal burning and associated emissions. This paper focuses on the extraction phase and assesses the external costs to agriculture associated with the resulting depression cone. This paper discusses the difficulties researchers face in estimating agricultural losses resulting from the development of a depression cone due to opencast mineral extraction. In the case of brown coal, the impacts are of a geological, natural-climatic, agricultural-productive, temporal, and spatial nature and result from a multiplicity of interacting factors. Then, a methodology for counting external costs in crop production was proposed. The next section estimates the external costs of crop production arising from the operation of opencast mines in the Konin-Turek brown coal field, which is located in central Poland. The analyses conducted showed a large decrease in grain and potato yields and no effect of the depression cone on sugar beet levels. Including the estimated external costs in the cost of producing electricity from mined brown coal would significantly worsen the profitability of that production.

Arabidopsis thaliana (L.) Heynh. is a model plant species in which to study plant gene functions. Recently developed virus-induced gene silencing (VIGS) offers a rapid and high-throughput technique platform for gene function analysis. In this paper we report optimisation of tobacco rattle virus (TRV)-induced gene silencing in Arabidopsis. The parameters potentially affecting the efficiency of VIGS in Arabidopsis were investigated. These included the concentration and pre-incubation of Agrobacterium inocula (agro-inocula), the concentration of acetosyringone included in agro-inocula, the Agrobacterium inoculation (agro-inoculation) method, the ecotypes and the growth stages of Arabidopsis plants for agro-inoculation, and the growth temperature of agro-inoculated plants. The optimised VIGS procedure involves preparing the agro-inocula with OD600 of 2.0, pre-incubating for 2 h in infiltration buffer containing 200 μm acetosyringone, agro-inoculating by vacuum infiltration, and growth of agro-inoculated plants at 22 −24°C. Following this procedure consistent and highly efficient VIGS was achieved for the genes encoding phytoene desaturase (PDS) and actin in Arabidopsis. The silencing phenotype lasts for at least 6 weeks, and is applicable in at least seven ecotypes, including Col-0, Cvi-0, Sd, Nd-1, Ws-0, Bay-0 and Ler. TRV-induced VIGS was expressed not only in leaves, but also in stems, inflorescences and siliques. However, VIGS was not transmissible through seed to the subsequent generation. The optimised procedure of the TRV-induced gene silencing should facilitate high-throughput functional analysis of genes in Arabidopsis.

In a field experiment with strawberries, pre-plant treatments with broad-spectrum fumigants methyl bromide-chloropicrin (450 kg/ha) or methyl isothiocyanate-dichloropropene (500 l/ha) (and 300 l/ha) controlled wilt caused by Verticillium dahliae Kleb and resulted in increased yields. Soil fumigation with the nematicide ethylene dibromidz (105 l/ha) also improved yields. It controlled the root-knot nematode (Meloidogyne hapla Chitwood), delayed the onset of wilt symptoms and reduced the severity of disease. This indicated a nematode-fungus interaction and is the first report of a Meloidogyne-Verticillium interaction in strawberry.

The experiment testing N residues (made on a clay-loam soil) clearly showed that N applied for potatoes benefited the following wheat crop, but that N applied for wheat benefited the following potato crop little. There were no worth-while 1. An experiment on a clay-loam soil measured responses to three amounts of nitrogen on alternate crops of wheat and potatoes; these dressings were tested in all combinations with three rates of N applied 1 and 2 years previously. Nitrogen applied for potatoes consistently increased yields of following wheat. The residue from applying 1·5 cwt. N/acre for potatoes was equivalent to topdressing the wheat with 0·55 cwt. N/acre; the value of the residue was decreased by applying N to the wheat. Potato yields were increased little by applying N to the preceding wheat crop and the residues were of little significance when compared with the responses to new N. There was no gain from N applied 2 years previously for either crop.
