Increase consumption of high fat diet was found to alter blood sugar level similar to diabetes and contributed to the development of obesity and affected the reproductive function of both sexes. The study aimed to clarify the influence of diabetes and or hypercholesterolemia on the cytological picture of cells of the anterior lobe of pituitary gland of male albino rats. Eighteen male albino rats weighing approximately 120 gram body weight were divided into three main groups; control, diabetes, hypercholesterolemia, diabetes (single i.p. 40 mg streptozotocin/kg B.wt plus 100mg. nicotinamide /kg body weight) and hypercholesterolemia (diet containing 3% cholesterol). Dietary feeding on cholesterol and diabetes were carried out for 12 weeks. At the end of treatment, animals were sacrificed, and pituitary glands were separated and their anterior lobe was processed for cytological investigations by transmission electron microscopy. The present study revealed that the rats subjected to experimental diabetes and/ or hypercholesterolemia exhibited a decrease of the secretory granules within the gonadotroph cells somatotroph and corticotrophin cells. There was a detected intracellular accumulation of fat globules in both the gonado- and sommatotroph cells. The authors reported that the altered cytological structures of the secretory function of the anterior pituitary gland led to marked impairment of the male hormonal level and causing infertility.

SUMMARY A study on the effect of synthetic thyrotrophic releasing factor (TRF) on different areas of the central nervous system and on the anterior pituitary gland was performed in a system in vitro. These observations were combined with ultrastructural studies of the anterior pituitary gland after intracarotid TRF administration. TRF specifically decreased the incorporation in vitro of labelled amino acids into proteins of the medial hypothalamus, whereas the anterior pituitary gland showed enhanced protein synthesis. At the electron microscope level, only thyrotrophs showed enhanced signs of activation after 5, 15 and 30 min of TRF administration, which consisted in hypertrophy of the rough endoplasmic reticulum and Golgi complexes associated with a marked increase in the number of secretory granules undergoing extrusion into the perivascular and extracellular spaces. These observations indicate that the medial hypothalamus may be related to regulatory mechanism(s) of TRF secretion. They provide further morphological identification of the thyrotrophs and corroborate that TRF acts on these cells by promoting release and synthesis of thyrotrophin.

Abstract. OT is synthetized in the hypothalamus. These neurons project to the posterior lobe of the pituitary and the external zone of the median eminence. In order to localize OT in the male rat anterior pituitary we have used immunocytochemistry on ultrathin sections in target cell(s) obtained by cryoultramicrotomy. OT-like immunoreactivity was observed in lactotropes only. No immunoreactivity was observed in gonadotropes, somatotropes, corticotropes or thyrotropes. In lactotropes, immunoreactivity was localized at the plasma membrane level, in the cytoplasmic matrix and around the secretory granules, but not in the other organelles, and in the nucleus. No reaction was observed by using either non-immune serum or anti-OT serum incubated with OT. No modification of OT-like immunoreactivity was observed by using antiOT serum incubated with heterologous peptides. These results 1) provide immunocytological evidence for the presence of OT in the anterior pituitary gland; 2) indicate the presence of this peptide in one particular cell type, and 3) support the hypothesis that OT could have a direct participation in the regulation of the PRL release.
