The article deals with the requirements and needs for long-term digital preservation in different areas of scholarly work. The concept of long-term digital preservation is introduced by comparing it to digitization and archiving concepts and defined with the emphasis on dynamic activity within a certain time line. The structure of digital preservation is presented with regard to the elements of the activity as understood in Activity Theory. The life-cycle of digitization processes forms the basis of the main processing of preserved data in preservation archival system.The author draws on the differences between humanities and social sciences on one hand and natural and technological science on the other. The empirical data characterizing the needs for digital preservation within different areas of scholarship are presented and show the difference in approaches to long-term digital preservation, as well as differences in selecting the items and implementing the projects of digital preservation. Institutions and organizations can also develop different understanding of preservation requirements for digital documents and other objects.The final part of the paper is devoted to some general problems pertaining to the longterm digital preservation with the emphasis of the responsibility for the whole process of safe-guarding the cultural and scholarly heritage for the re-use of the posterior generations. It is suggested that the longevity of the libraries in comparison with much shorter life-span of private companies strengthens the claim of memory institutions to playing the central role in the long-term digital preservation.

Optical coherence tomography angiography (OCTA) is a non-invasive tool for imaging and quantifying the retinal and choroidal perfusion state in vivo. This study aimed to evaluate the acute effects of isometric and dynamic exercise on retinal and choroidal sublayer perfusion using OCTA. A pilot study was conducted on young, healthy participants, each of whom performed a specific isometric exercise on the first day and a dynamic exercise the day after. At baseline and immediately after the exercise, heart rate (HR), mean arterial pressure (MAP), superficial capillary plexus perfusion (SCPP), deep capillary plexus perfusion (DCPP), choriocapillaris perfusion (CCP), Sattlers’s layer perfusion (SLP), and Haller’s layer perfusion (HLP) were recorded. A total of 34 eyes of 34 subjects with a mean age of 32.35 ± 7.87 years were included. HR as well as MAP increased significantly after both types of exercise. Both SCPP and DCPP did not show any significant alteration due to isometric or dynamic exercise. After performing dynamic exercise, CCP, SLP, as well as HLP significantly increased. Changes in MAP correlated significantly with changes in HLP after the dynamic activity. OCTA-based analysis in healthy adults following physical activity demonstrated a constant retinal perfusion, supporting the theory of autoregulatory mechanisms. Dynamic exercise, as opposed to isometric activity, significantly changed choroidal perfusion. OCTA imaging may represent a novel and sensitive tool to expand the diagnostic spectrum in the field of sports medicine.

This article examines early modern learning through Ashkenazic responsa. Beyond explicit evidence from published responsa collections, implicit insights dwell in what these publications lack. These missing features shed light on sixteenth-century scholarly practices. The works’ organizational inconsistencies must be understood in context of learned archives. Such an adjustment offers a corrective to the regnant narrative, which views the introduction of print as a sharp rupture from earlier modes of transmission. This article suggests instead that the culture of printed books coexisted with older approaches, and that print was complemented by more disorderly and unconstrained forms of transmission. Comparing rabbinic “paper-ware” with contemporaneous humanist practices highlights the rabbi's working papers, focusing on a culture's dynamic activity rather than its stable output. This shift in perspective allows us to see rabbinic writings not merely as a collection of books but as a mode of scholarship.

The selection of reward-seeking and aversive behaviors is controlled by two distinct D1 and D2 receptor-expressing striatal medium spiny neurons, namely the direct pathway MSNs (dMSNs) and the indirect pathway MSNs (iMSNs), but the dynamic modulation of signaling cascades of dMSNs and iMSNs in behaving animals remains largely elusive. We developed an in vivo methodology to monitor Förster resonance energy transfer (FRET) of the activities of PKA and ERK in either dMSNs or iMSNs by microendoscopy in freely moving mice. PKA and ERK were coordinately but oppositely regulated between dMSNs and iMSNs by rewarding cocaine administration and aversive electric shocks. Notably, the activities of PKA and ERK rapidly shifted when male mice became active or indifferent toward female mice during mating behavior. Importantly, manipulation of PKA cascades by the Designer Receptor recapitulated active and indifferent mating behaviors, indicating a causal linkage of a dynamic activity shift of PKA and ERK between dMSNs and iMSNs in action selection.

To examine the capability of genetic regulatory systems for complex dynamic activity, we developed simple kinetic models that incorporate known features of these systems. These include autoregulation and stimulus-dependent phosphorylation of transcription factors (TFs), dimerization of TFs, crosstalk, and feedback. The simplest model manifested multiple stable steady states, and brief perturbations could switch the model between these states. Such transitions might explain, for example, how a brief pulse of hormone or neurotransmitter could elicit a long-lasting cellular response. In slightly more complex models, oscillatory regimes were identified. The addition of competition between activating and repressing TFs provided a plausible explanation for optimal stimulus frequencies that give maximal transcription. Such optimal frequencies are suggested by recent experiments comparing training paradigms for long-term memory formation and examining changes in mRNA levels in repetitively stimulated cultured cells. In general, the computational approach illustrated here, combined with appropriate experiments, provides a conceptual framework for investigating the function of genetic regulatory systems.

Background: Changes in knee kinematics after anterior cruciate ligament (ACL) injury may alter loading of the cartilage and thus affect its homeostasis, potentially leading to the development of posttraumatic osteoarthritis. However, there are limited in vivo data to characterize local changes in cartilage thickness and strain in response to dynamic activity among patients with ACL deficiency. Purpose/Hypothesis: The purpose was to compare in vivo tibiofemoral cartilage thickness and cartilage strain resulting from dynamic activity between ACL-deficient and intact contralateral knees. It was hypothesized that ACL-deficient knees would show localized reductions in cartilage thickness and elevated cartilage strains. Study Design: Controlled laboratory study. Methods: Magnetic resonance images were obtained before and after single-legged hopping on injured and uninjured knees among 8 patients with unilateral ACL rupture. Three-dimensional models of the bones and articular surfaces were created from the pre- and postactivity scans. The pre- and postactivity models were registered to each other, and cartilage strain (defined as the normalized difference in cartilage thickness pre- and postactivity) was calculated in regions across the tibial plateau, femoral condyles, and femoral cartilage adjacent to the medial intercondylar notch. These measurements were compared between ACL-deficient and intact knees. Differences in cartilage thickness and strain between knees were tested with multiple analysis of variance models with alpha set at P < .05. Results: Compressive strain in the intercondylar notch was elevated in the ACL-deficient knee relative to the uninjured knee. Furthermore, cartilage in the intercondylar notch and adjacent medial tibia was significantly thinner before activity in the ACL-deficient knee versus the intact knee. In these 2 regions, thinning was significantly influenced by time since injury, with patients with more chronic ACL deficiency (>1 year since injury) experiencing greater thinning. Conclusion: Among patients with ACL deficiency, the medial femoral condyle adjacent to the intercondylar notch in the ACL-deficient knee exhibited elevated cartilage strain and loss of cartilage thickness, particularly with longer time from injury. It is hypothesized that these changes may be related to posttraumatic osteoarthritis development. Clinical Relevance: This study suggests that altered mechanical loading is related to localized cartilage thinning after ACL injury.

Background:Shape writing is relatively new technology for on-screen keyboards that enable users of mobile touch-screen devices to input text by drawing continuous lines. With growth of touch-screen device usage, there has risen the need to investigate potential risks that may occur during prolonged usage. Objective: The biomechanical strain on upper limb muscles were assessed while study-participants used Swype technology on a tablet touch screen device and compared with traditional/regular input methods. Methods: Study-participants performed typing tasks (email and text) using Swype and regular input methods under counterbalanced conditions with sEMG data collected to measure muscle activity during tasks. Results: Email & Text had the same exertion for all muscles except the Extensor. The interaction between task and muscle was significant, F (1.6, 27.5) = 15.39, p < .001, ηp2 = 0.48. The interaction between muscle, task and method was also significant, F (2.19, 37.19) = 3.6, p = 0.03, ηp2= 0.18. Exertion was lower for Swype but with marginal significance. Overall, Email resulted in less dynamic activity than Text with Main effects F(1, 17) = 10.07, p = 0.006, ηp2 = 0.37. Extensor has more dynamic activity than other muscles with main effect F(1.8, 29.9) = 16.51, p < 0.001, ηp2 = 0.49. Conclusion: Results indicate that Swype presents no more biomechanical strain than regular input for most muscles. Swype may result in less exertion for the Extensor muscles in the lower arm. This may be particularly true for tasks requiring interactions like those found in the email task.
