Fish muscle, which accounts for 15%–25% of the total protein in fish, is a desirable protein source. Their hydrolysate is in high demand nutritionally as a functional food and thus has high potential added value. The hydrolysate contains physiologically active amino acids and various essential nutrients, the contents of which depend on the source of protein, protease, hydrolysis method, hydrolysis conditions, and degree of hydrolysis. Therefore, it can be utilized for various industrial applications including use in nutraceuticals and pharmaceuticals to help improve the health of humans. This review discusses muscle protein hydrolysates generated from the muscles of various fish species, as well as their amino acid composition, and highlights their functional properties and bioactivity. In addition, the role of the amino acid profile in regulating the biological and physiological activities, nutrition, and bitter taste of hydrolysates is discussed.

Fish populations play an active role in the maintenance of aquatic ecosystems biodiversity. Their intestinal microbiota and fillet chemistry depend on abiotic and biotic factors of the water environments that they inhabit. The present study investigated the grey mullets’ gut microbiota from a transitional aquatic ecosystem (Santa Giusta Lagoon, Sardinia, Italy) by a multidisciplinary approach which refers the results of (1) gut cultivable microbiota analyses (MA), (2) the trace metal assessment of fish muscle (TM), (3) the physico-chemical water monitoring (PC). MA detected the greatest number of total aerobic heterotrophic bacteria, Enterobacteriaceae and coliforms in Autumn (mean values 1.3 × 105, 2.4 × 104, 1.1 × 104 cfu g−1, respectively) when the accumulated rain and mean values of nutrients (reactive phosphorous and silica) were the highest. Marine bacteria were more numerous in Summer (mean value 7.4 × 105 cfu g−1) when the highest mean values of water temperature and salinity were registered. The gut bacteria were identified as Pseudomonas spp. (64%), Aeromonas spp. (17%), Ochrobactrum pseudogrignonense (10%), Providencia spp. (5%), Enterobacter ludwigii (2%) and Kocuria tytonicola (2%). TM showed that Ca, Na, B and Ni increased their concentrations in Winter while maxima of P, Zn, Cu and Fe were found in muscles of fish sampled in Summer. This study highlighted that the fish intestinal microbiota and metal composition of the fillet reflected the seasonal aquatic environmental variability.

White skeletal muscle of crucian carp contains a single isoenzyme of glycogen phosphorylase, which was purified approximately 300-fold to a specific activity of approximately 13 mumol.min-1.mg protein-1 (assayed in the direction of glycogen breakdown at 25 degrees C). Tissue extracts of crucian muscle produced three distinct peaks of phosphorylase activity when separated on DEAE-Sephacel. Peaks 1 and 3 were identified, in terms of kinetic properties and by interconversion experiments, as phosphorylase b and a, respectively. Peak 2 was shown to be a phospho-dephospho hybrid. The three interconvertible forms of phosphorylase were purified and shown to be dimeric molecules at 20 degrees C. At 5 degrees C, a and the hybrid tended to form tetramers. The Mr of the subunit was estimated to be 96,400 from sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The hybrid is kinetically homogeneous, and its kinetic properties are intermediate between those of b and a forms. The b, hybrid, and a forms of phosphorylase can be isolated from rapidly frozen muscle of crucian but in different proportions, depending on whether fish were anesthetized or forced to muscular activity for 20 s. Muscle of anesthetized crucian had 36, 36, and 28% of phosphorylase b, hybrid, and a forms, respectively, whereas the corresponding values for exercised fish were 12, 37, and 51%. Results suggest that three interconvertible forms of phosphorylase exist simultaneously in crucian muscle and that hybrid phosphorylase is active in contracting muscle in vivo.

The extent of energy depletion was assessed in Atlantic cod (Gadus morhua) in spring and early summer (1993-1995) to assess relationships between poor condition and natural mortality. Several indices of condition were compared in wild fish in the northern Gulf of St. Lawrence and in fish exposed to a prolonged period of starvation in laboratory experiments. Discriminant analyses classified only a small fraction of the wild fish as similar to cod that did not survive and a much larger fraction as similar to cod that survived starvation. This percentage increased from April to May and peaked in June 1993 and 1994. Condition factor and muscle somatic index allowed a clear distinction between live and dead fish. Muscle lactate dehydrogenase activity suggested that cod had experienced a period of negative growth early in 1993, 1994, and 1995. Fish classified as similar to starved individuals were characterized by a higher gonad to liver mass ratio than others. Reproduction may have a negative impact on survival not only in spring but also later into summer, as some individuals were found not to have recovered by late summer. This study shows that natural mortality from poor condition contributed to lower production in the early 1990s.
