Single-cell protein (SCP) is the term used for crude or pure protein derived from simple single or multi-celled microorganisms. Pineapple peel contains monosaccharides as much as 10.8% so that it can be used as a fermentation medium in single-cell protein production. The purpose of this study was to determine the effect of adding carbon sources of fructose and sucrose on pH, cell dry weight, and protein content in the manufacture of single-cell proteins. This study used a completely randomized design (CRD) with two factors, namely the addition of carbon (fructose, sucrose, and control) and fermentation time (24.48, and 72 hours). The data analysis used the variance test and the Duncan Multiple Range Test (DMRT) continued to test with a confidence level of 95%. The results showed that the addition of carbon to the media had a very significant effect on media pH, cell dry weight, and protein content. In the medium with the addition of fructose it has a pH of 3.81; dry weight 0.4203 grams; and protein content 69.08/L. Whereas in the media with the addition of sucrose, the pH was 4.33, the dry weight of the cells was 0.3385 grams, and the cells had a protein content of 85.55 mg/L. The addition of a fructose carbon source gave the cell dry weight more than the addition of carbon sucrose

Abstract Motivation R Experiment objects such as the SummarizedExperiment or SingleCellExperiment are data containers for storing one or more matrix-like assays along with associated row and column data. These objects have been used to facilitate the storage and analysis of high-throughput genomic data generated from technologies such as single-cell RNA sequencing. One common computational task in many genomics analysis workflows is to perform subsetting of the data matrix before applying down-stream analytical methods. For example, one may need to subset the columns of the assay matrix to exclude poor-quality samples or subset the rows of the matrix to select the most variable features. Traditionally, a second object is created that contains the desired subset of assay from the original object. However, this approach is inefficient as it requires the creation of an additional object containing a copy of the original assay and leads to challenges with data provenance. Results To overcome these challenges, we developed an R package called ExperimentSubset, which is a data container that implements classes for efficient storage and streamlined retrieval of assays that have been subsetted by rows and/or columns. These classes are able to inherently provide data provenance by maintaining the relationship between the subsetted and parent assays. We demonstrate the utility of this package on a single-cell RNA-seq dataset by storing and retrieving subsets at different stages of the analysis while maintaining a lower memory footprint. Overall, the ExperimentSubset is a flexible container for the efficient management of subsets. In the whiting, no significant changes occur in the composition of the muscle substance, either throughout the year, or with increasing age, or during ripening of the gonads.The amount of fat in the liver is very low in immature fishes, increasing greatly with age, and reaching its maximum just after the commencement of the ripening of the gonads. It then falls off, reaching its lowest value amongst mature fishes in the “Spent” stages. Males are richer in liver fat than females.Immature fish have a decidedly higher protein content in the liver than have mature fish.

This study investigated the influence of treated greywater on growth and protein content of multipurpose (forage and ornamental) transplants, Prosopis juliflora L., Prosopis tamarugo L., and Albizia lebbeck L. Transplants of tested species were irrigated with treated greywater, diluted greywater (grey + distilled water, 1:1/by volume), and distilled water (control) for seven months. Water quality analysis showed that the concentrations of nutrients and heavy metals found in the greywater were within the acceptable range compared with Jordan Institution for Standard and Metrology (JISM) and the World Health Organization (WHO) thresholds for safe use of greywater. Escherichia coli found in the greywater were lower compared to JISM and WHO guidelines for the safe use of greywater. Irrigation with treated greywater increased shoot fresh weight by 24–39% and dry weight by 34–40% compared to diluted greywater and control. No significant difference in crude protein was noticed between water treatments. Prosopis species (P. juliflora Albizia lebbeck L. and P. tamarugo Albizia lebbeck L.) had higher shoot fresh (35%) and dry weight the same species had lower crude protein (44%) when compared to Albizia lebbeck Albizia lebbeck L. The reuse of treated greywater for landscaping or forage production alleviates the demand for water resources and reduces the pressure on wastewater treatment plants. However, considering the controversial findings of previous studies on greywater quality (especially, long-term reuse), the reuse of treated greywater needs to be considered with caution and periodic quality analyses and economic assessments are required.

Cold tolerance of the springtail Gomphiocephalus hodgsoni Carpenter (Collembola: Hypogastruridae) was studied at Cape Bird, Ross Island, Antarctica (77°13′S, 166°26′E). Microclimate temperatures indicate a highly seasonal thermal environment, with winter minima <–39°C. Snow cover significantly buffers both minimum temperatures and cooling rates. Gomphiocephalus hodgsoni survives low temperatures by avoiding freezing. Mean low group supercooling points (SCPs) ranged from –35.4°C in October to –28.3°C in January. The lowest SCP measured was –38.0°C. The high SCP group was very small, making up only 18% of the population in January. In October, G. hodgsoni had a very high glycerol content (>80 μg mg−1 dry weight), although this declined rapidly to low levels (c. 7–10 μg mg−1 dry weight) in January. Quantities of glucose and trehalose were low during October, but steadily increased throughout the summer. Haemolymph osmolality was exceptionally high (up to 1755 mOsm kg−1) at the end of November, but this rapidly declined to c. 500 mOsm kg−1 by late December. The presence of thermal hystersis proteins was indicated by both osmometry on haemolymph samples and recrystallization inhibition studies of springtail homogenates. There was a strong relationship between glycerol content and SCP, but the relationship between haemolymph osmolality, SCP and carbohydrates is uncertain.

The human aging process is associated with molecular changes and cellular degeneration, resulting in a significant increase in cancer incidence with age. Despite their potential correlation, the relationship between cancer- and ageing-related transcriptional changes is largely unknown. In this study, we aimed to analyze aging-associated transcriptional patterns in publicly available bulk mRNA-seq and single-cell RNA-seq (scRNA-seq) datasets for chronic myelogenous leukemia (CML), colorectal cancer (CRC), hepatocellular carcinoma (HCC), lung cancer (LC), and pancreatic ductal adenocarcinoma (PDAC). Indeed, we detected that various aging/senescence-induced genes (ASIGs) were upregulated in malignant diseases compared to healthy control samples. To elucidate the importance of ASIGs during cell development, pseudotime analyses were performed, which revealed a late enrichment of distinct cancer-specific ASIG signatures. Notably, we were able to demonstrate that all cancer entities analyzed in this study comprised cell populations expressing ASIGs. While only minor correlations were detected between ASIGs and transcriptome-wide changes in PDAC, a high proportion of ASIGs was induced in CML, CRC, HCC, and LC samples. These unique cellular subpopulations could serve as a basis for future studies on the role of aging and senescence in human malignancies.

Ocean warming driven bleaching is one of the greatest threats to zooxanthellate cnidarians in the Anthropocene. Bleaching is the loss of Symbiodiniaceae, chlorophyll, or both from zooxanthellate animals. To quantify bleaching and recovery, standardised methods for quantification of Symbiodiniaceae and chlorophyll concentrations have been developed for reef-building scleractinian corals, but no such standard method has been developed for octocorals. For stony corals, quantification of Symbiodiniaceae and chlorophyll concentrations often relies on normalisation to skeletal surface area or unit of biomass [i.e., protein, ash-free dry weight (AFDW)]. Stiff octocorals do not change their volume, as such studies have used volume and surface area to standardise densities, but soft-bodied octocorals can alter their size using water movement within the animal; therefore, Symbiodiniaceae and chlorophyll cannot accurately be measured per unit of surface area and are instead measured in units of Symbiodiniaceae and chlorophyll per μg of host protein or AFDW. Though AFDW is more representative of the full biomass composition than host protein, AFDW is more time and resource intensive. Here, we provide a streamlined methodology to quantify Symbiodiniaceae density, chlorophyll concentration, and protein content in soft-bodied octocorals. This technique uses minimal equipment, does not require freeze-drying or burning samples to obtain ash weight, and is effective for down to 0.2 g wet tissue. Bulk samples can be centrifuged, the Symbiodiniaceae pellet washed, and the supernatant saved for protein analysis. This efficient technique allows for clean, easy to count samples of Symbiodiniaceae with minimal animal protein contamination. Chlorophyll a and c2 extractions occurs at different rates, with chlorophyll a taking 24 h to extract completely at 4°C and chlorophyll c2 taking 48 h. Finally, we found that where necessary, wet weight may be used as a proxy for protein content, but the correlation of protein and wet weight varies by species and protein should be used when possible. Overall, we have created a rapid and accurate method for quantification of bleaching markers in octocorals.

Abstract The cause of global warming is the increasing carbon concentration arising from industrial activities, burning of fossils, and land-use change. The purpose of this research was to find out the allometric equation to calculate the local bamboo biomass and then to be able to calculate how much carbon sequestration at bamboo riparian forest since this area was rarely being explored. The parameters observed were the height and diameter of the bamboo stem at 1.3 m height of 6 types of local bamboo using destructive sampling, along with the measurement of bamboo weight. The carbon content of the bamboo biomass, litter, and soil was measured to complement the estimation of total carbon sequestration. The results showed that the allometric equation for estimating local bamboo biomass is Y=0.6396 X1.6162 with R2=0.77, obtained from the relationship equations between dry weight and the diameter. Total carbon sequestration of this system ranged between 81 to 215 tons C ha−1.
