This work aims to study the qualitative composition of biologically active substance (BAS) extracts in vitro callus, cell suspension, and root cultures of the medicinal plant Rhaponticum carthamoides. The research methodology is based on high-performance liquid chromatography, and 1H nuclear magnetic resonance (NMR) spectra, to study the qualitative and quantitative analysis of BAS. The results of the qualitative composition analysis of the dried biomass extracts of in vitro callus, cell suspension and root cultures showed that the main biologically active substances in the medicinal plant Rhaponticum carthamoides are 2-deoxy-5,20,26-trihydroxyecdyson (7 mg, yield 0.12%), 5,20,26-trihydroxyecdyson 20,22-acetonide (15 mg, yield 0.25%), 2-deoxy-5,20,26-trihydroxyecdyson 20,22-acetonide (6 mg, yield 0.10%), 20,26-dihydroxyecdyson 20,22-acetonidecdyson 20,22-acetonide (5 mg, yield 0.09%), and ecdyson 20,22-acetonide (6 mg, yield 0.10%). In the future, it is planned to study the antimicrobial, antioxidant, and antitumor activity of BAS of extracts of in vitro callus, cell suspension, and root cultures of the medicinal plant Rhaponticum carthamoides, for the production of pharmaceuticals and dietary supplements with antitumor, antimicrobial and antioxidant effects.

This article shows the possibility of using a biotechnological method to obtain biologically active compounds based on in vitro cultivation of callus cultures. The callus biomass of Adonis vernalis was obtained in vitro by using the biotechnological method. The extracts based on callus biomass of Adonis vernalis were obtained. The content of flavonoids and phenolic compounds was determined. The antioxidant activity of callus biomass of Adonis vernalis has also been studied. The research results show that the content of biologically active substances and biological activity in callus biomass does not differ from plant raw materials.

The medicinal plant Dactylorhiza maculata (L.) Soó (family Orchidaceae) is used to treat gastritis, colic, gastrointestinal tract, and bladder diseases. This study aimed to investigate the properties and characteristics of the in vitro Dactylorhiza maculata extract. The recommended parameters for producing Dactylorhiza maculata extract were determined: temperature 60 °C, process duration 60 min, hydro module 1:10. It is recommended to carry out the extraction using an aqueous ethyl alcohol solution with a mass fraction of the parent substance of 70%. It was found that such biologically active substances as rutin, quercetin, 3,3’,4’,5,5’,7-hexahydroxyflavonone, 3,3’,4’,5,5’,7-hexahydroxyflavonone-3-O-glycoside, gallic acid, and ferulic acid were dominant in Dactylorhiza maculata. A high phosphorus content was noted (2410.8 mg/kg dry matter). The studied Dactylorhiza maculata samples contained a large number of organic acids and water-soluble vitamins. The tested extracts were safe in terms of the content of heavy metals, pesticides, aflatoxin B1, and radionuclides, as well as pathogenic and opportunistic microorganisms; the content was significantly lower than the threshold limit values. The studied complex of biologically active substances from Dactylorhiza maculata extract samples had antimicrobial properties. It was found that the antioxidant activity of the samples was 217.89 ± 10.89 mg AA/g (AA—ascorbic acid). The high content of bioactive substances and the antimicrobial and antioxidant properties of Dactylorhiza maculata extract samples determine the application potential of this plant as a substitute for growth stimulants and feed antibiotics in the production of feed additives, aiming to increase the physiological and immune status of livestock and poultry.

Objective: This study was undertaken to research of antioxidant properties of extracts of the plants and the callus biomass.Methods: The implementation of the method of cultivating the cells, tissues, and organisms in vitro for the reception of biological material of Carlina аcaulis L. (Asteraceae) is a timely issue for the medical and pharmaceutic industry, as the domestication in vitro creates the opportunity for the year-round production of the plant material as the source of biologically active substances. The seeds and plant materials (roots) of C. acaulis L. collected from natural habitat - meadows mountains Klyva Tarnychkovain Ukraine in the Carpathians. The influence of phytohormones on growth processes of plant cells is researched; the optimal conditions for the cultivation of C. acaulis L. are defined and chosen. Biomass extract is obtained and researched for the presence of biologically active substances and conducted definition of oxygen radical absorbance capacity (ORAC) of extracts of medical herbs and callus biomass of C. acaulis L.Results: C. аcaulis L. was cultivated in vitro. There was set up a scheme of sterilization of seeds with the biggest outcome of aseptic explants. There took place the investigation of the qualitative content of the extracts of callus biomass of C. acaulis L. and the detection of flavonoids and tannins. Conducted definition of ORAC of extracts of medical herbs and callus biomass of C. acaulis L. Conclusion: The obtained data showed that the investigated extracts of callus biomass and biomass of the medical herb C. acaulis L. having antioxidant activity, which makes it possible to continue research in this direction.

Introduction. Geroprotectors are biologically active substances that inhibit the aging process. Many plant species are natural geroprotectors. For instance, Thymus vulgaris and Trifolium pratense are callus cultures with strong geroprotective properties. Study objects and methods. The present research featured T. vulgaris and T. pratense grown in vitro on various nutrient media. Their extracts were obtained by aqueous-alcoholic extraction using the following parameters: water-ethanol solvent Se = 30, 50, and 70 %; temperature Te = 30, 50, and 70°C; time τe = 2, 4, and 6 h. The quantitative and qualitative analysis was based on high-performance liquid mass spectrometry, gas mass spectrometry, and thin-layer chromatography. Results and discussion. The optimal extraction parameters for T. vulgaris were τe = 4 h, Te = 50°C, Se = 70 %, for T. pratense – τe = 6 h, Te = 70°C, Se = 70 %. The chromatography detected flavonoids, phenylpropanoids, simple phenols, higher fatty acids, mono- and sesquiterpenes, and aliphatic hydrocarbons. T. vulgaris appeared to have the highest content of thymol (23.580 ± 1.170 mg/mL); its thymol, apigenin, gallic, chlorogenic, and caffeic components demonstrated geroprotective properties. The extract of T. pratense possessed the highest rutin content (10.05 ± 0.35 mg/mL), and it owed its geroprotective characteristics to rutin, chlorogenic and p-coumaric acids. Conclusion. The callus cultures of T. vulgaris and T. pratense proved to be promising sources of geroprotective biologically active substances.

Turnip (latin Brassica rapa) is a herbaceous plant from the genus Cruciferous. The root crop is a traditional product that has been producing in Russia since ancient times. This unpretentious herbaceous plant contains many different nutrients. The composition of turnips determines its value as a dietary product. From an economic point of view, the root crop, along with broccoli, can act as a source of biologically active substances like sulforaphane. Processing root crops using extraction will expand the scope of its application. The work presents a laboratory way for producing an extract of sulforaphane, followed by its quantitative determination from turnip varieties Petrovskaya-1. It was shown that the content of sulforaphane turnip extract (0.83 ± 0.03) mg in 1 cm3 extract. Next, the antioxidant ability of the extract was determined. A spectrophotometric study of antioxidant activity using a model adrenaline autooxidation reaction (in vitro) revealed that the aqueous extract has a pronounced antioxidant effect. It was registered that at an exposure time of 5 min, the AOA of the extract was 49.0%, at 10 min it was 13.0%. We can say that the processing of root crops is promising as a source of biologically active substances.

In this paper, the results of studies on the release of biological active compounds from their encapsulated forms under conditions of enzymatic hydrolysis in vitro are presented. In the phase of the model «small intestine» swelling of the capsules and their subsequent decay occurs, which allows to speak about the controlled release of encapsulated bioactive components. It was revealed that almost 90 % of the residual quantity of essential ingredients was released from the capsules in the model phase of the artificial «small intestine». At the end of the experiment, the capsules released all the encapsulated biologically active substances, regardless of the content of fish oil and phenolic compounds in them. It was noted that the poly-capsules had the greatest propensity to withstand the aggressive environment of the «model stomach» and concentrate in themselves the maximum amount of biologically active substances. Mathematical modeling confirms the direct transport of biologically active compounds and the role of the swelling of capsules in the release of biologically active compounds. two mathematical models describing the classical theory of diffusion from capsules and incorporating the material relaxation coefficient demonstrate a combination of empirical and theoretical approaches in controlling the properties of encapsulated biologically active substances. the obtained data are promising in the field of development of improved and functional food products, as well as the dry ingredients and concentrates
