1. Lambs were removed form their dams at 2 d of age, and at 5 weeks of age eitht of one group were incoulated with a mixed rumen ciliate and seven in a second group were maintained ciliate-free throughout the 61 weeks of the experiment. Performance of the groups was compared when given a 2:1 roughage:concentrate diet as a set ration and withad lib.roughage.2. Mixed ciliate populations developed in all faunated lambs and average-sized populations were maintained. Large numbers of flagellate protozoa developed in the ciliate-free animals after flagellates had been introduced into the building with the ciliate-free animals after flagellates had been introduced into the building with the ciliate inoculum.3. Higher numbers of rumen bacteria were found in the ciliate-free group.4. Only between the 14th and 21st weeks was there a significant different between groups in weight gain and this was infavour of the faunated group. The only significant different in body measurements was greater girth in the ciliate-free lambs.5. Only minor differences were found between the groups in calorimetric trials, digestibility and nitrogen balanes. There were no differences between groups in concentration of total protein N and soluble sugar in the rumen. The ammonis concentration was significantly higher in the faunated group.6. The concentrations of total rumen volatile fatty acids (VFA) were higher in the faunated group. Differences, between groups, in proportions of VFA were attributed to the activity of the rumen bacteria rather than the ciliatesper se.7. No differences between groups were found in the concentrations of blood sugar and haemoglobin.8. It was concluded that the changes due to the presence of rumen ciliates were not great enough to be reflected in animal performance under the conditions of this experiment.

The rumen microbiota is vital for the health and growth performance of the host animal, mainly due to its role in the fermentation of ingested feed within the rumen. Attaining a better understanding of the development of the bacterial community and fermentation in the rumen can provide the theoretical basis for regulating feed utilization. This study analyzed the development of rumen bacteria in lambs from birth to 4 months of age using 16S-rRNA amplicon sequencing data and studied its relationship with ruminal fermentation. Serum levels of metabolites were monitored at 30, 60, 90, and 120 days of age, and the RandomForest approach was used to determine age-related changes in rumen bacteria. Levels of blood metabolites, ruminal fermentation, the rumen bacterial community and its functions were all affected by the age of the lambs (P < 0.05). Based on the Bray-Curtis distance within the age groups of the rumen microbiota, the similarity increased sharply after the lambs were weaned at 60 days of age (P < 0.05). The similarity between the samples collected from birth to 90 days of age and those collected at 120 days of age, increased after 20 days of age, reaching a maximum at 90 days vs. 120 days (P < 0.05). Some age-associated changes in the microbial genera were correlated with changes in the concentrations of volatile fatty acids and the levels of microbial crude protein in the rumen, including positive correlations between main volatile fatty acids and the genera of Prevotella 1, Lachnospiraceae NK3A20 group, Ruminococcus gauvreauii group, Ruminococcaceae UCG-014, and Ruminococcus 2 (P < 0.05). These results indicated that the microbial community and the function of rumen was not well-established before 20 days of age, so there is a degree of plasticity in the rumen bacterial community during the first 20 days of post-natal development in lambs, and this might provide an opportunity for interventions to improve rumen fermentation and, thus, increase their growth performance.

A latin square design was used with four sheep, each equipped with a rumen fistula and re-entrant cannula in the proximal duodenum. The four treatments were intraruminal dosings with different amounts (0, 450, 900 and 1800 mg/day) of nitrilotriacetic acid (NTA). The sheep were fed a pelleted diet with excessive concentrations of manganese, iron, zinc and copper. Fecal excretion of iron and zinc increased, and apparent absorption decreased, with increasing dose of NTA. However, only the differences between means for zinc were statistically significant. Fecal excretion and apparent absorption of manganese and copper were not affected by NTA. Flow of soluble iron through the duodenum increased with increasing dose of NTA. There was no effect of NTA on concentrations of manganese, iron, zinc and copper in the rumen bacteria or on microbial activity in the rumen as measured by the concentration of volatile fatty acids in the rumen and by flow of nitrogen into the small intestine.

The potential risk to animal and human health in the use of antibiotic feed additives for modifying rumen fermentation has necessitated the search for natural alternatives which are generally regarded as safe. The aim of this study was to evaluate the potentials of turmeric powder in rumen manipulation using the in vitro method. Substrate of Panicum maximum and concentrate in ratio 6: 4 with turmeric inclusion at four levels of 0, 5, 10 and 15 mg/g dry matter (DM) were used for this study. The experiment was arranged in a completely randomized design. Approximately 200 mg of substrate in each treatment was weighed separately into 100 mL glass syringes into which 30 mL of rumen fluid and buffer solution (1:2 v/v) were added. The quantities of total gas, methane, ammonia, total volatile fatty acids production and substrate degraded were determined 48-h post incubation. Rumen bacteria, protozoa, fungi population were determined and microbial biomass was estimated. Some phytochemical constituents of turmeric were also determined using standard methods. Turmeric had a higher percentage of curcumin relative to other phytochemical contents determined. Turmeric effectively and consistently (p < 0.05) reduced gas production at levels above 5 mg/g of substrate inclusion throughout the 48-h incubation period. Similarly, turmeric reduced (p < 0.05) methane, carbon-dioxide, ammonia and total volatile fatty acids production, and substrate degradation at 10 – 15 mg/g inclusion. Rumen bacteria and protozoa reduced when turmeric was included at 10 – 15 mg/g while fungi reduction was observed at 15 mg/g of inclusion. Reduction in microbial biomass was observed at 15 mg/g of turmeric inclusion. It can be concluded from this study that turmeric inclusion above 5 mg/g DM of substrate, can modify the rumen by causing a reduction in fermentation end-products. The reduction of ammonia production at 15 mg/g which significantly reduced microbial biomass has implication for lowering microbial protein synthesis.     Le risque potentiel pour la santé animale et humaine dans l'utilisation d'additifs alimentaires antibiotiques pour modifier la fermentation du rumen a nécessité la recherche d'alternatives naturelles qui sont généralement considérées comme sûres. Le but de cette étude était d'évaluer les potentiels de la poudre de curcuma dans la manipulation du rumen en utilisant la méthode in vitro. Substrat de Panicum maximum et concentré dans le rapport 6: 4 avec l'inclusion de curcuma à quatre niveaux de 0, 5, 10 et 15 mg/g de matière sèche (DM) ont été utilisés pour cette étude. L'expérience a été organisée dans une conception complètement randomisée. Environ 200 mg de substrat dans chaque traitement ont été pesés séparément dans des seringues en verre de 100 mL dans lesquelles 30 mL de liquide rumen et de solution tampon (1:2 v/v) ont été ajoutés. Les quantités totales de gaz, de méthane, d'ammoniac, de production totale d'acides gras volatils et de substrat dégradé ont été déterminées 48 h après incubation. La bactérie Rumen, le protozoaire, la population de champignons ont été déterminés et la biomasse microbienne a été estimée. Certains constituants phytochimiques du curcuma ont également été déterminés à l'aide de méthodes standard. Le curcuma avait un pourcentage plus élevé de curcumine par rapport à d'autres contenus phytochimiques déterminés. Le curcuma a réduit efficacement et systématiquement (p < 0,05) la production de gaz à des niveaux supérieurs à 5 mg/g d'inclusion du substrat tout au long de la période d'incubation de 48 h. De même, le curcuma a réduit (p < 0,05) le méthane, le dioxyde de carbone, l'ammoniac et la production totale d'acides gras volatils, et la dégradation du substrat à 10 à 15 mg/g d'inclusion. Les bactéries rumen et le protozoaire ont diminué lorsque le curcuma a été inclus à 10 – 15 mg/g tandis que la réduction des champignons a été observée à 15 mg/g d'inclusion. La réduction de la biomasse microbienne a été observée à 15 mg/g d'inclusion de curcuma. On peut conclure de cette étude que l'inclusion de curcuma au- dessus de 5 mg/g de DM de substrat, peut modifier le rumen en causant une réduction des produits finaux de fermentation. La réduction de la production d'ammoniac à 15 mg/g, ce qui a considérablement réduit la biomasse microbienne, a des répercussions sur l'abaissement de la synthèse des protéines microbiennes.

Chain elongation of volatile fatty acids for medium chain fatty acids production (e.g. caproate) is an attractive approach to treat wastewater anaerobically and recover resource simultaneously. Undefined microbial consortia can be tailored to achieve chain elongation process with selective enrichment from anaerobic digestion sludge, which has advantages over pure culture approach for cost-efficient application. Whilst the metabolic pathway of the dominant caproate producer, Clostridium kluyveri, has been annotated, the role of other coexisting abundant microbiomes remained unclear. To this end, an ethanol-acetate fermentation inoculated with fresh digestion sludge at optimal conditions was conducted. Also, physiological study, thermodynamics and 16 S rRNA gene sequencing to elucidate the biological process by linking the system performance and dominant microbiomes were integrated. Results revealed a possible synergistic network in which C. kluyveri and three co-dominant species, Desulfovibrio vulgaris, Fusobacterium varium and Acetoanaerobium sticklandii coexisted. D. vulgaris and A. sticklandii (F. varium) were likely to boost the carboxylates chain elongation by stimulating ethanol oxidation and butyrate production through a syntrophic partnership with hydrogen (H2) serving as an electron messenger. This study unveils a synergistic microbial network to boost caproate production in mixed culture carboxylates chain elongation.

ООО «БИОТРОФ», Санкт-Петербург Аннотация: Приведены данные исследования методом газожидкостной хроматомасс-спектрометрии метаболитов, синтезируемых пробиотическими бактериями, входящими в состав кормовой добавки «Профорт®». Проведен опыт в условиях интенсивного промышленного птицеводства по применению кормовой добавки «Профорт®» (50 тыс. голов бройлеров в группе). Среди метаболитов пробиотических штаммов Enterococcus sp. и Bacillus sp. обнаружены такие полезные вещества, как молочная кислота, уксусная, пропионовая и другие короткоцепочечные (летучие) жирные кислоты, активные пептиды. Результаты зоотехнического опыта показали, что при скармливании бройлерам пробиотика «Профорт®» (500 г/т) живая масса при убое в 40 дней была выше контроля на 6,9%, конверсия корма улучшилась на 3,0%, а европейский индекс продуктивности бройлеров - на 5,69%.
