The effects of stress and starvation on brain levels of serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were studied in Arctic charr (Salvelinus alpinus). Three experimental protocols were used to elucidate (1) the effect of stress in fish given food, (2) the effect of starvation, and (3) the effect of stress in fish deprived of food. In the stress experiments, fish were stressed three times a day over a four-week period, and in the starvation experiment the fish were starved for a four-week period. Stressed fish, whether given food or not, showed significantly higher concentrations of 5-HIAA, the main 5-HT metabolite, in both the telencephalon and the brain stem. The 5-HIAA/5-HT ratio (an index of serotonergic activity) was also significantly increased in the brain of stressed fish. In the telencephalon of starved fish, the 5-HT concentration was significantly decreased. However, starvation had no effect on 5-HIAA concentrations or 5-HIAA/5-HT ratios in either the telencephalon or the brain stem. These results suggest that stress increases brain serotonergic activity in Arctic charr, while starvation has no effect on the utilization of this transmitter system. It is suggested that stress could be a mediator of the increased 5-HTAA levels and 5-HIAA/5-HT ratios recently observed in low-ranking Arctic charr in a dominance hierarch.

Within a species, different males may display different mating strategies. For example, some males may be selected to invest in attractiveness and mate guarding, whereas others are selected for increased sperm production and sneaky breeding. In systems with a hierarchical structure, dominant males are expected to adopt mate-guarding behaviour and subordinate males sneaky-breeding behaviour. In this study, we kept wild-caught and sexually ripe male Arctic charr (Salvelinus alpinus) in size-matched pairs and determined social rank from the number of aggressive encounters. After 4 days, subordinate males showed symptoms of stress, with higher blood glucose and erythrocyte levels than dominant males. There were no differences between dominant and subordinate males in parasite intensities or immune activity, measured as levels of granulocytes and lymphocytes in blood. Although subordinate males had smaller testes than dominant males, they still had a higher density of sperm cells and higher sperm numbers relative to the size of their testes. These results can be explained as indicating adaptation of subordinate males for reproduction in an unfavourable role, always exposed to sperm competition and out of synchrony with females' egg release. Our results suggest that rapid changes in social rank may affect ejaculate production.

Abstract Arctic charr (Salvelinus alpinus L.) were exposed to five levels of acidity between pH 6 and pH 3.8. Swimming performance as determined by critical swimming speeds was 67.5 cm · sࢤ1 or 4.4 body lengths per second for untreated fish (pH 7.8). Performance declined sharply below pH 4.5; at pH 3.8 it was reduced by 35% after 7 days of exposure. Tailbeat frequencies and ventilation rates showed no dose-response effects. At swimming speeds between 20 and 50 cm · sࢤ1, ventilation rates at all levels of acidity were higher than at the control level.

Incorporation of genomic technologies into fish breeding programs is a modern reality, promising substantial advances regarding the accuracy of selection, monitoring the genetic diversity and pedigree record verification. Single nucleotide polymorphism (SNP) arrays are the most commonly used genomic tool, but the investments required make them unsustainable for emerging species, such as Arctic charr (Salvelinus alpinus), where production volume is low. The requirement to genotype a large number of animals for breeding practices necessitates cost effective genotyping approaches. In the current study, we used double digest restriction site-associated DNA (ddRAD) sequencing of either high or low coverage to genotype Arctic charr from the Swedish national breeding program and performed analytical procedures to assess their utility in a range of tasks. SNPs were identified and used for deciphering the genetic structure of the studied population, estimating genomic relationships and implementing an association study for growth-related traits. Missing information and underestimation of heterozygosity in the low coverage set were limiting factors in genetic diversity and genomic relationship analyses, where high coverage performed notably better. On the other hand, the high coverage dataset proved to be valuable when it comes to identifying loci that are associated with phenotypic traits of interest. In general, both genotyping strategies offer sustainable alternatives to hybridization-based genotyping platforms and show potential for applications in aquaculture selective breeding.
