Protective effects of ethyl acetate extract from Shorea roxburghii against diabetes induced testicular damage in rats

Background: Based on traditional beliefs, Biarum carduchrum (family Araceae) can strengthen the nervous system and prevent from sleep disorder and agitation. Objectives: This study was performed to investigate the protective effects of the ethyl acetate extract of Biarum carduchrum leaves against pentylenetetrazol (PTZ)-induced seizure in rats. Methods: The present experimental research was conducted on 50 male Wistar rats in Iran during 2019. The animals were examined in five groups of control, PTZ, intervention (PTZ with 100 and 200 mg/kg ethyl acetate extract for 10 days), diazepam (PTZ, 200 mg/kg extract, and diazepam). The research groups were compared in terms of behaviors. Furthermore, total phenol and flavonoid levels in the extract were determined using high-performance liquid chromatography. Results: According to the results, the intervention and diazepam groups had a significantly lower number of the whole body, tonic, head, and upper organ seizures (P<0.05), and jumps (P<0.05), compared to the PTZ group. Both doses of the extract also significantly decreased immobility time in tail suspension and secondary latency time and significantly enhanced spatial memory in Morris water maze test (P<0.05). This treatment also significantly reduced the levels of nitric oxide and malondialdehyde in the brain and serum (P<0.05) and showed antioxidant activity (IC50=200 μg/ml). The extract contained 42.63±0.7494 and 85.16±6.499 µg/mg phenol and flavonoid, respectively. The HPLC analysis also revealed the presence of quercetin (30 μg/g) in the extract. Conclusions: Based on the results, Biarum carduchrum extract can be used for depression control and improvement of learning and memory impairments in seizure patients after complementary testing.

Protective effects of Curcuma longa rhizomes ethyl acetate extract against alcohol induced oxidative stress and nephrotoxicity in female Wistar rats. Biofarmasi J Nat Prod Biochem 21: 5-12. This study aimed to evaluate the protective effect of Curcuma longa Linn. (syn. Curcuma domestica Val.) rhizomes ethyl acetate extract (CLREAE) facing alcohol-induced oxidative stress and nephrotoxicity. Thirty female (30) Wistar rats were categorized randomly into six groups. Groups 1, 2, 3, 4, 5 and 6 were treated with normal saline; 20% ethanol; 100 mg of CLREAE + 20% ethanol; 200 mg of CLREAE + 20%; 350 mg of CLREAE + 20% ethanol and 350 mg of CLREAE respectively for 14 days. A significant (p<0.05) decrease in the SOD, CAT and GPx activities and GSH concentration of rat treated with only 20% ethanol were found when compared to the normal control group, whereas a significant (P<0.05) increase in the groups pretreated with different doses of the CLREAE were also found when compared to groups with only 20% ethanol treatment. Thus, comparing to the normal control group, treatment with the CLREAE fetched a significant (p<0.05) decrease in the renal biomarkers (creatinine and urea).  Whilst, comparing to the groups with 20% methanol treatment, a significant (p<0.05) increase happened in the groups pretreated with different doses of the CLREAE. There was a significant (p<0.05) decrease on Kidney MDA level in rats pretreated with different doses of CLREAE compared with the normal control. It was shown in the results of the histology that there was a physiologic recovery in the kidney tissues as groups were treated with different doses of the CLREAE. Evidenced by reduced necrosis of tubular and glomerular epithelial, the signs of protection against toxicity were found on the rats. The study suggested that through in vivo free radical scavenging ability, the CLREAE has protective effects against alcohol-induced oxidative stress and nephrotoxicity in female Wistar rats.

Study to assess the larvicidal property of Lantana camara leaves against Aedes triseriatus larvae found that the ethyl acetate extract had profound larvicidal action with the crude extract having a LC50 value of 409.831ppm. GC-MS analysis of the ethyl acetate extract confirmed the presence of twenty-one compounds out of which beta-caryophyllene covered the highest percentage of the chromatogram area. Further tests with beta-caryophyllene against the mosquito larvae proved it to be the active ingredient of L. Camara with a LC50 value of 104.243ppm.

Marrubium vulgare (Lamiaceae) is frequently used in traditional medicine to treat many illnesses from ancient times. Its beneficial effects include antibacterial, antioedematogenic, and analgesic activities. This study was designed to evaluate the antioxidant and anti-inflammatory activities of organic and aqueous extracts of the leaves, the flowers, the stems, and the roots of Marrubium vulgare. The total phenolic and flavonoid contents as well as the antioxidant and the anti-inflammatory effects of methanol, chloroform, ethyl acetate, and aqueous extracts have been investigated by using different in-vitro methods. It was found that the ethyl acetate extract from Marrubium vulgare stems had the highest total phenolic content, while the ethyl acetate extract from the leaves yielded a high concentration of flavonoids. The ethyl acetate extract from the stems exhibited the highest activity in scavenging of 2,2-diphenyl- 1-picrylhydrazyl (DPPH), as well as in protecting erythrocytes. The leaves aqueous extract exhibited the highest ferrous chelating activity and its methanolic extract was found to be the strongest inhibitor of lipid peroxidation in β-carotene bleaching assay. The leaves chloroform extracts as well as the flowers methanol, chloroform, and ethyl acetate extracts were found to decrease the pro-inflammatory tumor necrosis factor alpha (TNF-α) cytokine levels in a dose-dependent manner. On the other hand, the flowers methanolic extract and the leaves methanol, ethyl acetate, and aqueous extracts decreased the interleukin-1 beta (IL- 1β) release. It was also found that the methanol extract from the flowers and the chloroform extract from the stems of Marrubium vulgare inhibited interleukin-8 (IL-8) release. This study provides a scientific basis for the traditional use of Marrubium vulgare as an anti-inflammatory agent and for the plant to be considered as an important resource of natural antioxidants.

Background: The only remedy for up surging problem of antibiotic resistance is the discovery of antibacterial agents of natural origin. Objective: The present study was aimed at finding antibacterial potential of crude and solvent extracts of mature leaves of Plumeria pudica. Methods: Antibacterial activity of three different solvent extracts were evaluated in four human and four fish pathogenic bacteria by measuring the zone of inhibition and determining Minimum Inhibitory Concentration and Minimum Bactericidal Concentration values. Standard antibiotics were used as positive control. Preliminary phytochemical screening of most effective extract i.e., ethyl acetate extract, Fourier Transform Infra Red analysis and GC-MS analysis of the Thin Layer Chromatographic (TLC) fraction of ethyl acetate extract were done meticulously. All experiments were done thrice and analyzed statistically. Results: Crude leaf extracts and solvent extracts caused good inhibition of bacterial growth in all selected bacteria. Ethyl acetate extract showed highest inhibition zones in all tested strains with maximum inhibition (19.50±0.29 mm) in Escherichia coli (MTCC 739). MBC/MIC of the extracts indicated that all three solvent extracts were bactericidal. Preliminary phytochemical tests revealed the presence of tannins, steroids and alkaloids and FT-IR analysis revealed presence of many functional groups namely alcoholic, amide, amine salt and aldehyde groups. From the GC-MS analysis of TLC fraction of ethyl acetate extract five different bioactive compounds e.g., 2,4-ditert –butylphenyl 5-hydroxypentanoate, Oxalic acid; allyl nonyl ester, 7,9-Ditert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione, Dibutyl phthalate and 2,3,5,8-tetramethyl-decane were identified. Conclusion: Leaf extracts of P. pudica contain bioactive compounds that can be used as broad spectrum bactericidal agent.
