The Difference

Aim. To evaluate the potential utility of fibrin matrices containing 10, 20, and 25 mg/ml of fibrinogen (fibrin-10, fibrin-20, and fibrin-30, respectively) in vascular tissue engineering (VTE). Materials and Methods. Fibrinogen was isolated using the method of ethanol cryoprecipitation and polymerized using a solution of thrombin and CaCl2. The fibrin structure was studied in a scanning electron microscope, and the physical and mechanical properties of the material were tested on a Zwick/Roell test machine. The metabolic activity of endothelial cells (EC) on the fibrin surface was evaluated by the MTT assay, and the viability of fibroblasts in the thickness of fibrin and possibility for migration by in fluorescent and light microscopy. Percent of fibrin shrinkage was determined from the difference in the sample volumes before and after removal of moisture. Results. The fiber diameter did not differ among all fibrin samples, but the pore diameter in fibrin-30 was smaller than those in fibrin-10 and fibrin-20. A possibility for migration of fibroblasts into the depth of the fibrin matrix and preservation of 97-100% viability of cells at a depth 5 mm was confirmed. The metabolic activity of EC on the surface of fibrin-20 and fibrin-30 exceeded that on collagen, fibronectin, and fibrin-10. All fibrin samples shrank in volume to 95.5-99.5%, and the highest shrinkage was seen in fibrin-10. The physical and mechanical properties of fibrin were inferior to those of human A. mammaria by a factor of 10. Conclusion. Fibrin with fibrinogen concentrations of 20 and 30 mg/ml maintains a high metabolic and proliferative activity of EC on the surface and also a high viability of fibroblasts in the matrix. Its availability, ease of preparation, and a number of other favorable properties make fibrin a promising material for VTE. However, the problem of insufficient strength requires further investigations.

AbstractEndothelial cells are among the fundamental building blocks for vascular tissue engineering. However, a clinically viable source of endothelium has continued to elude the field. Here, we demonstrate the feasibility of sourcing autologous endothelium from human fat – an abundant and uniquely dispensable tissue that can be readily harvested with minimally invasive procedures. We investigate the challenges underlying the overgrowth of human adipose tissue-derived microvascular endothelial cells by stromal cells to facilitate the development of a reliable method for their acquisition. Magnet-assisted cell sorting strategies are established to mitigate the non-specific uptake of immunomagnetic microparticles, enabling the enrichment of endothelial cells to purities that prevent their overgrowth by stromal cells. This work delineates a reliable method for acquiring human adipose tissue-derived microvascular endothelial cells in large quantities with high purities that can be readily applied in future vascular tissue engineering applications.

Vascular diseases with their high morbidity and mortality are a major challenge for medical science, engaging the best minds in modern medicine. The development of antithrombogenic surfaces still remains a huge challenge in the vascular tissue engineering field. Various researchers have expanded surface coating procedures allowing endothelial cells (EC) adhesion and retention on vascular substitutes or by incorporating some of the mechanisms employed by vascular endothelial cells i.e. heparin. The short in vivo patency of these grafts is related. Our group study evaluates a new surface modification based on polyelectrolyte building. The layer by layer self assembly and the result in polyelectrolyte multilayer films (PEM) became also in a recent past a challenging, simple and versatile way to engineer surfaces with highly specific properties. Previous studies indicated that the poly(sodium-4 styrene sulfonate)/poly (allylamine hydrochloride) PSS/PAH multilayered films when ended by PAH induce strong adhesion and retention of mature EC which spread and keep their phenotype as well on glass [1,2], on expanded polytetrafluoroethylene ePTFE [3] and on cryopreserved arteries [4,5]. The mechanical properties (compliance), leading to early intimal hyperplasia and graft failure, were lost after artery cryopreservation. We have demonstrated the compliance restoration of PEM treated cryopreserved close to native arteries [5]. The use of an autologous EC source avoids the immunological rejections of the grafts. With an autologous origin, high proliferation capacity and potentialities to proliferate and differentiate into matures EC, the endothelial progenitor cells (EPC) have raised huge interest and offer new opportunities in vascular engineering. Currents protocols for isolation and differentiation of EPC from peripheral blood requires at least 1 month to observe an endothelium-like morphology and about 2 months for confluent EC monolayer. The EPC cultivated on PEM treated glasses showed a monolayer development after only 14 days of culture. The morphological appearance and mature phenotype markers expression and repartition of the monolayer cells are close to mature EC [6]. These main results have led to French patent deposit in June 2007[7].
