Tenascin-C (TN-C), an extracellular matrix (ECM) glycoprotein, is specifically induced upon tissue injury and infection and during septic conditions. Carbon monoxide (CO) gas is known to exert various anti-inflammatory effects in various inflammatory diseases. However, the mechanisms underlying the effect of CO on TN-C-mediated inflammation are unknown. In the present study, we found that treatment with LPS significantly enhanced TN-C expression in macrophages. CO gas, or treatment with the CO-donor compound, CORM-2, dramatically reduced LPS-induced expression of TN-C and proinflammatory cytokines while significantly increased the expression of IL-10. Treatment with TN-C siRNA significantly suppressed the effects of LPS on proinflammatory cytokines production. TN-C siRNA did not affect the CORM-2-dependent increase of IL-10 expression. In cells transfected with IL-10 siRNA, CORM-2 had no effect on the LPS-induced expression of TN-C and its downstream cytokines. These data suggest that IL-10 mediates the inhibitory effect of CO on TN-C and the downstream production of proinflammatory cytokines. Additionally, administration of CORM-2 dramatically reduced LPS-induced TN-C and proinflammatory cytokines production while expression of IL-10 was significantly increased. In conclusion, CO regulated IL-10 expression and thus inhibited TN-C-mediated inflammationin vitroandin vivo.

Gas CO is formed as a result of incomplete combustion. The material containing carbon monoxide is gas emission from motorcycle vehicle which is toxic to humans. This study aims to analyze the correlation between CO gas that also known as Carbon monoxide gas in the air with COHb level of blood parking basement officers in Surabaya "x" mall. This study was analytical observational with cross sectional design and the data was analyzed by Chi-Square test. The number of samples used was 15 parking basement officers and a control group of 15 people as a comparison. The results of measurement of CO gas levels in 3 parking points were more than standards recommendation. While the other 3 points in the control area were below the quality standard. All of samples in the parking basement officers had COHb levels >5% and for control areas there were 11 persons who had COHb levels of <5% and 4 people who had COHb levels of >5%. The results suggested that there was a significant correlation between the characteristics of individuals with COHb levels, such as duration of work (p = 0.040), nutritional status (p = 0.047) and smoking habits (p = 0.046). While variable of age (p = 0.070) and personal protective equipment (PPE) use (p = 0.128) showed no significant correlation. In addition, there was a significant correlation between exposure of CO gas and blood COHb levels in basement car park officers in Surabaya “X” Mall (p = 0.000), and there was a distinction between COHb level of blood basement car park officers in the "x" mall building and the control group with a value of p = 0.000. Based on the results, it is suggested for car park officers to cease smoking, go to places with more oxygen, maintain diet and do exercise regularly. It is also suggested and recommended for agencies is to put more attention regarding exhaust fans’ maintenance and sufficiency.

Resonance frequency shift of a zinc oxide- (ZnO-) functionalized microcantilever as a response to carbon monoxide (CO) gas has been investigated. Here, ZnO microrods were grown on the microcantilever surface by a hydrothermal method. The measurement of resonance frequency of the microcantilever vibrations due to the gas was carried out in two conditions, that is, gas flow with and without air pumping into an experiment chamber. The results show that the resonance frequency of the ZnO-functionalized microcantilever decreases because of CO in air pumping condition, while it increases when CO is introduced without air pumping. Such change in the resonance frequency is influenced by water vapor condition, and a possible model based on water-CO combination was proposed.

: The quest to find novel strategies to tackle respiratory illnesses has led to the exploration of the potential therapeutic effects of carbon monoxide (CO) as an endogenous signaling molecule and a cytoprotective agent. Further, several studies have demonstrated the pharmacological efficacy of CO in animal models of respiratory disorders such as acute lung injury and pulmonary hypertension. Because of the gaseous nature of CO and its affinity for multiple targets, its controlled delivery has been a challenge. Past studies have employed different delivery modalities including CO gas, HO-1 inducers, and CO donors, sometimes leading to substantive variations of the resulting pharmacological effects for various reasons. Herein, this review summarizes and analyzes the differences among the profiles of various CO-delivery modalities in terms of their efficacy, dosing regimen, and pharmacokinetics in airways models. We believe that analysis of these issues will help in understanding the fundamental roles of CO in airways and eventually contribute to its development as a medicine for respiratory diseases.

The incidence of poisoning due to carbon monoxide gas arising from drilling activities on the first floor of a building in the Kelapa Gading beauty clinic in Jakarta resulted in 17 people experiencing poisoning. In this study developing a device on the sensor used to detect CO and SO2 gas, in the air of a closed room using gas sensor MQ 135 and MQ 136. The results of testing the CO and SO2 gas gauges using samples of cigarette smoke and sulfur powder using MQ 135 and MQ 136 sensors with fuzzy rule base logic for motor speed to produce CO and SO2 gas, that obtained a value of 0.233 ppm SO2 gas safe conditions and gas input CO with the sensor obtained a value of 0.513 ppm, the condition is safe so that the output is 49.8 ppm, the condition of the fan blower does not rotate. Whereas when the reading value of 5.0 ppm is very concentrated and the CO gas input with the sensor is 13.8 ppm the condition is very concentrated producing an output of 228 ppm the very danger.
