Toxicity assessment of 255 chemicals to pure cultured nitrifying bacteria using biosensor

The bioassay has been attracting attention as a method of toxicity assessments of micropollutants in the environment. In this study, we report the characteristics (selectivity and sensitivity) of the nitrifying bacteria biosensor for 255 kinds of chemicals as a model of chemical contaminant in the environment and the results of evaluation of mixed samples of several substances. In the nitrifying bacteria respiration inhibition test using the biosensor, 56 chemicals were detected. It was found that this biosensor is especially sensitive to seven chemicals that have a thiocarbonyl functional group (>C=S), such as a thioamide group of thiocarbamate group. These chemicals are considered to specifically inhibit AMO by chelation of copper. The samples consisted of a mixture of seven types of anilines that inhibit respiration in the bacteria, a mixture of five types of chlorophenols, and a mixture of eight types of substances that contain thiocarbonyl groups were examined. All of the mixed samples inhibited the respiration of the nitrifying bacteria more than 10% by the inhibition rate, and observed a synergistic effects of the substances in the samples.

<p>Gelatin  has  been  successfully  extracted  from  chicken  clawsby  acid  hydrolysis method through four major stages of degreasing, demineralization, extraction and drying. The yield of  extracted  gelatin obtained  from wet chicken claws  was about 6.21  %  (w/w). The  effectiveness  of  gelatin  as  preservative  applied  in  beef  and  fish  was  physically, chemically and microbiologically  studied  by  organoleptic  analysis,  peroxide test, bacteria inhibition  test  and  number  calculation, respectively. Organoleptic analysis by hedonic  test shows  there  were significant  differences  for  color  and  texture  parameters  between  gelatin coated  samples  and control  samples  at  concentrations  of  1:1  (w/w).  Peroxide number analysis  by  iodometry  indicates  the  use  of  gelatin  in  the  preservation  of  meat  and  fish decreased the oxidation number. The result of inhibition test againts Staphylococcus aureus bacteria  shows  the  gelatin  concentration  of  20  %  provided  greatest  halo  zone  diameter (inhibitory  region)  as  19.44  ±  2.11  mm. The colony bacteria test shows the use of gelatin potentially  inhibited  the growth  of  bacteria  up to  75  %.  Overall,  the results  conclude  that gelatin  extracted from chicken claws  has the potential  activities  as a  natural preservative for beef and fish.</p>

The activated sludge respiration inhibition test and the luminescent bacteria test with Vibrio fischeri are important bacterial test systems for evaluation of the toxicity of chemical compounds. These test systems were further optimized to result in better handling, reliability and sensitivity. Concerning the Vibrio fischeri test, media components such as yeast extract and bivalent cation concentrations like Ca2+ and Mg2+ were optimized. The cultivation, storage conditions and reactivation process of the stored bacteria were also improved, which enabled simpler handling and led to good reproducibility. Additionally, the respiration inhibition test with a prolonged incubation time was further analyzed using different chlorinated phenols as reference compounds. It could be stated that a longer incubation period significantly improved the sensitivity of the test system.
