Maternal high-fat diet consumption and obesity have been shown to program long-term obesity and lead to impaired glucose tolerance in offspring. Many rodent studies, however, use non-purified, cereal-based diets as the control for purified high-fat diets. In this study, primiparous ICR mice were fed purified control diet (10–11 kcal% from fat of lard or butter origin) and lard (45 or 60 kcal% fat) or butter (32 or 60 kcal% fat)-based high-fat diets for 4 weeks before mating, throughout pregnancy, and for 2 weeks of nursing. Before mating, female mice fed the 32 and 60% butter-based high-fat diets exhibited impaired glucose tolerance but those females fed the lard-based diets showed normal glucose disposal following a glucose challenge. High-fat diet consumption by female mice of all groups decreased lean to fat mass ratios during the 4th week of diet treatment compared with those mice consuming the 10–11% fat diets. All females were bred to male mice and pregnancy and offspring outcomes were monitored. The body weight of pups born to 45% lard-fed dams was significantly increased before weaning, but only female offspring born to 32% butter-fed dams exhibited long-term body weight increases. Offspring glucose tolerance and body composition were measured for at least 1 year. Minimal, if any, differences were observed in the offspring parameters. These results suggest that many variables should be considered when designing future high-fat diet feeding and maternal obesity studies in mice.

Abstract Objectives Obesity is associated with impaired immune function. However, impact of obesity on blood T cell profile is not well studied. The objectives of this study were to investigate the effects of high fat diet (HFD)-induced obesity and long-term fruits and vegetable (FV) consumption on body composition and blood T cell profile. Methods This is partial report from an ongoing study. A total of 240 male C57BL/6J mice were randomly assigned to 4 groups: low fat control (LF-C) or high-fat control (HF-C) diet alone, or together with 15% of a unique mixture of FV (w/w, equivalent to 7–9 servings F&V/d for human) (LF-FV or HF-FV). The feeding will continue until 50% mortality is reached in one group. Body weight, body composition (using MRI), and blood T cell profile (using FACS) are monitored longitudinally at different time points. The results reported here are those assessed when mice were 7 months old. Results After 7 months of feeding, mice fed HF-C gained more weight compared to those fed LF-C. Mice fed HF-FV or LF-FV diets had significantly reduced weight gain and fat mass, and higher muscle mass compared to those fed HF-C or LF-C diet, respectively. Mice fed HF-C also had significantly lower percentage of blood CD3+, CD4+, and CD8 + T cells compared with the LF-C. FV supplementation prevented HFD-induced decrease in percentage of CD3+ and CD4+ cells. Furthermore, both % CD3+ and CD4+ cells were negatively correlated with body weight (P < 0.001) or percentage of fat mass (P < 0.001), and positively associated with percentage of lean mass (P < 0.001). Conclusions Our results suggest that consuming large amounts of a unique mixture of F&V curbs HFD-induced body weight gain, reduces fat mass, and favorably affects blood T cell population. Ongoing studies will assess these analytes when mice are 16 months old, and again when one group reaches 50% mortality, and determine their correlations with functional measures of T cell response, host resistance to infection, health span, and mortality. The vagal afferent pathway is important in short-term regulation of food intake, and decreased activation of this neural pathway with long-term ingestion of a high-fat diet may contribute to hyperphagic weight gain. We tested the hypothesis that expression of genes encoding receptors for orexigenic factors in vagal afferent neurons are increased by long-term ingestion of a high-fat diet, thus supporting orexigenic signals from the gut. Obesity-prone (DIO-P) rats fed a high-fat diet showed increased body weight and hyperleptinemia compared with low-fat diet-fed controls and high-fat diet-induced obesity-resistant (DIO-R) rats. Expression of the type I cannabinoid receptor and growth hormone secretagogue receptor 1a in the nodose ganglia was increased in DIO-P compared with low-fat diet-fed controls or DIO-R rats. Shifts in the balance between orexigenic and anorexigenic signals within the vagal afferent pathway may influence food intake and body weight gain induced by high fat diets.

Abstract Objectives Many cell culture and animal studies have demonstrated that Trans-resveratrol (R) has the potential to induce beige cell formation and activity. Although human studies indicate that R can maintain metabolic health, evidence is inconclusive regarding its browning effectiveness mainly due to its low aqueous solubility and high hepatic metabolism in humans. To combat the shortcomings of R, we have successfully synthesized biocompatible and biodegradable R-encapsulated liposomes (Rlipo). We will directly inject Rlipo into inguinal white adipose tissue (iWAT) in this project. The purpose of this study to evaluate the anti-obesity effects of resveratrol-encapsulated liposomes in female APOE*3Leiden.CETP transgenic mice, which have human-like lipoprotein metabolism. Methods Rlipo was prepared using R and soy phosphatidylcholine (soy-PC) dissolved in ethanol. After mixing and drying with nitrogen, deionized water was added followed by a sonication step. Ultrafiltration was used to remove any unencapsulated R. The void liposomes (Vlipo) were prepared using only soy-PC. Female APOE*3Leiden.CETP mice (n = 40) were fed with a high fat diet (45% of calorie from fat) throughout the study. After 4 weeks of the high-fat diet administration, mice were randomly divided into 4 groups (n = 10) and received iWAT injections of Rlipo, Vlipo, free R and saline (control) once per week for 5 weeks. R concentration was 17.5 mg/kg body weight/week. Body weight and food intake were measured weekly. Body composition of mice was measured using an EchoMRITM every other week. Paired sample t-test and One-way ANOVA were used to analyze differences between means. Results After 5-weeks of treatment compared to baseline, fat percentage differences were 1.99 ± 0.93%, 1.85 ± 0.58%, 1.45 ± 0.67%, and 1.40 ± 0.68% in control, free R, Vlipo and Rlipo groups, respectively. Body weight and fat mass showed a similar trend of change. Although control group showed an increase in lean mass (0.25 ± 0.95 g), RLipo group showed a decrease (−0.14 ± 0.52 g). Food intake was similar among four groups. Conclusions Nanoencapsulation of R can enhance R's anti-obesity effects. However longer treatment time might be necessary to see more prominent results. Abstract Objectives The objective of this study was to investigate the main and interactive effects of fat and sodium content of the diet on food intake, body weight and composition, glucose metabolism and blood pressure in male Wistar rats. Methods Male Wistar Rats (n = 48, initial body weight: 115.30 ± 1.73 g) were allocated into 4 groups (n = 12/group) and received one of the following diets: Normal sodium normal fat (NSNF), normal sodium high fat (NSHF), high sodium normal fat (HSNF), high sodium high fat (HSHF) diet for 12 weeks. Body weight (BW) and food intake (FI) were measured weekly. Short-term food intake (1, 2 and 12 hours food intake after 12 hours fasting) was measured at week 6. Body composition and organs’ weight were measured at week 12. Systolic (SBP) and diastolic (DBP) blood pressure, pulse and fasting blood glucose (FBG) were measured and oral glucose tolerance test (OGTT) was conducted at weeks 1, 4, 8 and 12. Results Regardless of sodium content, a greater FI (both gram and cal) was observed in rats fed normal fat diet compared with those fed high fat diet. Consistently, FI (g) at 1, 2 and 12 hours was higher in rats fed a normal fat diet. However, no difference in calorie intake was observed at any time point. Higher BW and fat (%) was observed in high fat diet groups. Moreover, greater kidneys’ weights was observed in high sodium diet groups. Fasting blood glucose was higher in rats fed a normal sodium diet compared with those fed a high sodium diet while the tAUC glucose response to glucose preload was higher in rats fed a high fat diet compared with those fed a normal fat diet which is consistent with higher body weight in high fat diet groups. Regardless of fat content of the diet, pulse was higher in rats fed a high sodium diet compared with those fed a normal sodium diet. No effect of either dietary sodium or fat content of the diet on SBP or DBP was observed. Conclusions Fat but not sodium content of the diet is a determining factor in regulation of FI and BW. Moreover, both fat and sodium content of the diet influence the glucose metabolism potentially through different mechanisms. While pulse is influenced by sodium content, the results of this study do not support the effect of sodium or fat content of the diet on either SBP or DBP. Aronia melanocarpa are a rich source of anthocyanins that have received considerable interest for their relations to human health. In this study, the anti-adipogenic effect of cyanidin-3-O-galactoside-enriched Aronia melanocarpa extract (AM-Ex) and its underlying mechanisms were investigated in an in vivo system. Five-week-old male C57BL/6N mice were randomly divided into five groups for 8-week feeding with a control diet (CD), a high-fat diet (HFD), or a HFD with 50 (AM-Ex 50), 100 (AM-Ex 100), or 200 AM-Ex (AM-Ex 200) mg/kg body weight/day. HFD-fed mice showed a significant increase in body weight compared to the CD group, and AM-Ex dose-dependently inhibited this weight gain. AM-Ex significantly reduced the food intake and the weight of white fat tissue, including epididymal fat, retroperitoneal fat, mesenteric fat, and inguinal fat. Treatment with AM-Ex (50 to 200 mg/kg) reduced serum levels of leptin, insulin, triglyceride, total cholesterol, and low density lipoprotein (LDL)-cholesterol. Real-time reverse transcription-polymerase chain reaction (RT-PCR) analysis revealed that AM-Ex suppressed adipogenesis by decreasing CCAAT/enhancer binding protein α, peroxisome proliferator-activated receptor γ, sterol regulatory element-binding protein-1c, peroxisome proliferator-activated receptor gamma coactivator-1α, acetyl-CoA carboxylase 1, ATP-citrate lyase, fatty acid synthase, and adipocyte protein 2 messenger RNA (mRNA) expressions. These results suggest that AM-Ex is potentially beneficial for the suppression of HFD-induced obesity by modulating multiple pathways associated with adipogenesis and food intake.

Early life nutrition impacts on subsequent risk of obesity and hypertension. Several brain chemicals responsible for both feeding and cardiovascular regulation are altered in obesity. We examined effects of early postnatal overnutrition on blood pressure, brain neuropeptide Y (NPY), and adiposity markers. Rat pup litters were adjusted to either 3 or 12 male animals (overnutrition and control, respectively) on day 1 of life. After weaning, rats were given either a palatable high-fat diet or standard chow. Smaller litter pups were significantly heavier by 17 days of age. By 16 wk, the effect of litter size was masked by that of diet, postweaning. Small and normal litter animals fed a high-fat diet had similar increases in body weight, plasma insulin, leptin, and adiponectin concentrations, leptin mRNA, and fat masses relative to chow-fed animals. An increase in 11β-hydroxysteroid dehydrogenase-1 mRNA in white adipose tissue, and a decrease in uncoupling protein-1 mRNA in brown adipose tissue in both small litter groups at 16 wk of age, may represent a programming effect of the altered litter size. NPY concentration in the paraventricular nucleus of the hypothalamus was reduced in high fat-fed groups. Blood pressure was significantly elevated at 13 wk in high-fat-fed animals. This study demonstrates that overnourishment during early postnatal development leads to profound changes in body weight at weaning, which tended to abate with maturation. Thus the effects of long-term dietary intervention postweaning can override those of litter size-induced obesity.
