Sperm morphometry is affected by increased inbreeding in the Retinta cattle breed: A molecular approach

Inbreeding depression, the genetic condition caused by mating related individuals, is particularly important in several cattle breeds with limited effective populations. This condition is often associated with decreases in performance, including fertility. Furthermore, sperm head morphometry was associated with fertility in several species. To our knowledge, the influence of inbreeding on sperm morphometry has not been widely reported in cattle. In this study, a Sperm Class Analyzer (SCA™, Microptic S.L., Barcelona, Spain) was used to characterise 7 sperm morphometry parameters in 59 Retinta bulls, including sperm head length, width, perimeter, ellipticity, elongation, regularity and rugosity. Two replicates of at least 100 sperm heads, from 2 frozen semen samples, were assessed per individual (n=200 sperm per bull). Additionally, all individuals were genotyped with the Axiom Bos1 high density SNP Array (Affymetrix, Santa Clara, CA, USA). The molecular-based inbreeding factor (FROH; mean 12.5%, range 1.75 to 33.0) had very low correlations with all sperm morphometry parameters. On average, sperm heads from bulls with higher FROH had a smaller area, but variability was high. Correlations between inbreeding and sperm shape were low and negative, length (r=−0.1449; P<0.01), width (r=−0.2494; P<0.01), and rugosity (r=−0.1086; P>0.01) being the most informative. Whole-genome association study (GWAS) analysis, performed using molecular inbreeding as co-factor, revealed 8 SNP, located on chromosomes BTA2, BTA5, BTA7, and BTA11, significantly associated with sperm head regularity and rugosity. Genomic analysis revealed that genes SLF1 and TMEFF2 are located close to enriched SNP. Gene SLF1 (SMC5-SMC6 complex localization factor 1) is associated with the regulation of protein complex and cytoskeleton assembly, whereas TMEFF2 (transmembrane protein with EGF like and 2 follistatin like domains 2) is associated with integral components of cell membrane and cell spreading and development. Therefore, we inferred that SLF1 and TMEFF2 may be involved in variations of sperm head shape. In this preliminary study, there was evidence of a potential influence of inbreeding on sperm morphometry in a beef cattle breed. However, additional studies, ideally including more individuals and additional breeds, are necessary to determine the validity of this potential association.

The Proclossiana eunomia (Esper, 1799) complex is currently composed of the several subspecies distributed throughout Palaearсtic region and North America. Despite the fact that some of the taxa have differences in wing pattern and body size, previous assumptions on taxonomy not supported by molecular data. Therefore, the identity of certain populations of this complex has remained unclear and the taxonomic status of several recently described taxa is debated. Here, we provide insights into systematics of some Palaearctic members of this group using molecular approach, based on the analysis of the barcoding fragment of the COI gene taking into account known morphological differences.

According to the current “Instruction” used in dairy cattle selection and breeding in the Republic of Kazakhstan, bulls-producers of dairy breeds are assessed according to the their offspring quality based on the principle of “peer daughter”. This means that the phenotypic indicators of the daughters of the tested bulls are compared with the corresponding indicators of their peers. In European countries with developed dairy cattle breeding, as well as in Canada, the USA, etc., to ensure a reliable forecast of the genetic value of individuals (primarily, bulls-producers), use is made of the best linear unbiased forecast method (BLUP method). This method implies that the breeding value of producers is determined by the deviation values of the development of traits of the examined animal from its average values in the population. Especially urgent area is the research aimed at improving breeding programs, including assessing the breeding value of bulls-producers of dairy breeds using BLUP methods based on the productive qualities of the mass of dairy cattle in the Republic of Kazakhstan. The research material included the data on the phenotypic indicators of the milk productivity of first-calf cows (the amount of milk yield, the content of fat and protein in milk, the yield of milk fat and protein) of the Holstein black-motley dairy cattle breed, obtained from the information and analytical database of the Republic of Kazakhstan for 2016–2017. It was found that when evaluating according to the official “Instruction”, 16 sires out of 256 bulls (6.2%) got the stud category in 2016, 14 sires (9.2%) out of 152 bulls in 2017, and – 30 sires of 249 bulls (12.0%) over the cumulative period. The results of the conducted research prove that the use of the classic “Instructions” in dairy cattle breeding has lower efficiency (by 42.8–90.0%) as compared with the assessment of the breeding value of bulls based on the BLUP method.The selection of sire bulls into breeding groups based on the “peer daughter” methodology is not reliable enough and rather ineffective. Comparing the results of assessing the breeding qualities of sire bulls, obtianed using two methods in all compared periods (2016, 2017, 2016–2017), the authors established a clear superiority of the BLUP method over the current Instruction used in the Republic of Kazakhstan.

In the Republic of Kazakhstan over the past decade positive changes have been observed in increasing the number of farm animals, increasing their productivity, and increasing the gross production of milk, meat and other products. At the same time the development of dairy cattle breeding is mainly due to the breeding of Black-and-White, Red Steppe, Simmental breeds and their intra-breed types as well as imported from abroad Holstein breed. The purpose of the research was to analyze ways to improve the intra-breed type Karatomar of Black-and-White breed in Northern Kazakhstan. Research work has been carried out on breeding animals such as Karatomar type cattle of Black-and-White breed in LLP “Experimental farm Zarechnoe” in the Kostanay district in the Kostanay region. The dynamics of growth and development indicators, reproductive traits of heifers of a new intra-breed type Karatomar of Black-and-White cattle have been studied. A comprehensive assessment of their economically useful traits in the first lactation, determined by the resistance of animals to climatic and forage conditions, the efficiency of breeding of animals of different genotypes has been conducted. It has been found that the replacement heifers – daughters of American breeding sires inherited precocity and showed higher growth energy after 6 months of age and continued to grow rapidly until 18 months. The advantage in metabolism was in the daughters of Holstein sires of the American selection Orbit 4078, and the increased immune status of the organism in the daughters of the sire of the domestic selection Yamal 975 as the most adapted to the conditions of Northern Kazakhstan. The best ability to increasing the milk yield, stable lactation activity, high milk productivity, gross content of dry matter and nonfat milk solids have been characterized by the daughters of sires of the American selection Orbit 4078 and Lowrider 4129. Analysis of economic indicators of milk production has been shown that from cows – daughters of sires Orbit 4078 and Lowrider 4129 for 305 days of lactation received 15,8 and 15,2 % more milk of base fat, the expenditures of EFU per 1 kg of production was lower by 10,2 and 5,9 %. A higher level of profitability has been observed in the group of daughters of the sires Orbit 4078 – 35,5 % and Lowrider 4129 – 33,1 %, which was 8,5 and 10,8 % higher than in the group of daughters of the sire Yamal 975.

Целью проведенных исследований являлась отработка и адаптация применения методик BLUP AM (Animal Model — модель животного) и TDM (Test-Day Model — модель тестового дня) для прогнозирования племенной ценности быков-производителей и оценки селекционно-генетических параметров на популяции скота симментальской породы четырех регионов РФ. Проведен сравнительный анализ указанных методов с более ранним методом BLUP SM (Sire Model — модель отца). Рассчитана племенная ценность быков и коров симментальской породы по признакам молочной продуктивности: удой за 305 дней, выход молочного жира, выход молочного белка. Анализ полученных средних значений достоверности оценок быков-производителей, рассчитанных на основе сопоставляемых методов, свидетельствует, что достоверность для признака «удой за 305 дней» при переходе от метода SM1 к AM1 увеличивается на 2,4%, а при переходе от SM1 к TDM1 — на 7,8%. Даны варианты генетического тренда по удою с использованием различных уравнений моделей расчета племенной ценности. На основании полученных данных можно сделать вывод о том, что модель тестового дня позволяет не только повысить точность оценок быков, но и более рельефно выявить их ранги, а также несколько уменьшить срок получения достоверных оценок производителей по качеству потомства по продуктивным признакам. The purpose of this research was to develop and adapt the application of BLUP AM (Animal Model) and TDM (Test-Day Model) methods for predicting the sires breeding value and evaluating genetic parameters for Simmental cattle population in four regions of the Russian Federation. A comparative analysis of these methods with the earlier BLUP SM (Sire Model) method is performed. The breeding value for sires and cows of Simmental breed was calculated by milk production traits: milk yield for 305 days; milk fatyield; milk proteinyield. The sires reliability of average breeding value calculated by different methods reveal that milk yield for 305 days when switching from the SM1 to AM1 method increases by 2.4%, and when switching from SM1 to TDM1 — by 7.8%.The variants of the genetic trend for milk yield are given using various equations of BLUP and TDM. Based on the obtained data, it can be concluded that the Test-Day model allows increasing the accuracy of bull’s evaluation and also more clearly identifying their ranks, as well as slightly reducing the time for obtaining reliable estimates of bulls by offspring for production traits.


Combination of genotypes of main types’ genetic markers causes milk productivity in cattle. The article studies correlation between kappa casein genotypes (C3N3), beta-lactoglobulin (LGB), their complexes and milk production indices in cows bred under equal conditions at a breeding farm. The calculation of parental milk yield and fat indices between the three breeds showed a significant difference (P>0,999) of PCI of Ayrshire and Holstein cattle breed milk yield - 3395 kg, PCI of fat - 0,22%; between Holstein and Yaroslavl cattle breeds - 3200 kg, PCI offat - 0,16%. A significant difference of realization indices of protein between all three breeds was not stated. The highest realization ofparental milk yield index was stated - between Ayrshire and Yaroslavl cattle breeds- 113%, Holstein cattle breed - 88%. As for the fat and protein indices, all three cattle breeds showed a high realization of PCI from 98% to 109%. However, Holstein cattle breed showed the highest percentage: protein - 101%, fat - 109%. A significant difference of PCI realization of milk productivity in CSN3 and LGB genotypes was not stated. It was also stated that there is a tendency of high level parental indices realization when there is a higher number of B-allele variants in genotypes of cows.
