Optimization of pectinase‐assisted extraction of Annona muricata L. juice and the effect of liquefaction on its pectin structure

The main goal of this work was to measure the total alkaloid content (TALC) from pulp, peel, seed, and columella of soursop fruit (Annona muricata L.) by ultrasound-assisted extraction (UAE) and to obtain the best conditions of the UAE with the response surface methodology (RSM). We evaluated the effect of amplitude (40%, 70%, and 100%), time (5, 10, and 15 min) and pulse-cycles (0.4, 0.7, and 1 s) and compared the best UAE conditions of alkaloids with a conventional extraction (maceration). The structural characterization of the raw material with the highest TALC was developed using nuclear magnetic resonance (NMR) techniques [1H, 13C, heteronuclear single quantum correlation (HSQC), heteronuclear multiple bond correlation (HMBC), and homonuclear correlation spectroscopy (COSY)]. According to the RSM, the best conditions in the UAE for extracting alkaloids varied depending on the type of plant tissue. It took 5 min with an amplitude of 70% and pulse-cycles of 1, 0.4, and 1 s, respectively, to extract the highest TALC in peel, seed, and columella while the optimal conditions for extracting the largest amount of alkaloids from the pulp were obtained at 5 min in UAE with pulse-cycles of 0.55 s and 100% amplitude. The TALC was highest in the peel (7.48 mg/g), which was followed by the seed (2.31 mg/g), the pulp (1.20 mg/g), and the columella (0.79 mg/g) and was positively correlated (R2 = 0.98–0.88) with the predicted values. In addition, the extraction alkaloids from the peel, pulp, seed, and columella using the UAE was 56.31, 5.45, 3.06, and 2.96 times higher, respectively, than the extraction by maceration. The alkaloids identified in the peel have not been reported and were nornuciferin, assimilobin, anonaine, and isolaureline. This study showed that the soursop fruit peel can be a source of alkaloids and that UAE has an important potential for extracting these compounds.

En México el 63% de los productores cultivan menos de cinco hectáreas agrícolas, por lo que es importante diseñar y desarrollar estrategias para aumentar la productividad en superficies pequeñas. En este trabajo se presentan los resultados de investigación obtenidos durante 2015 a 2017, en una huerta de guanábana (Annona muricata L.) establecida en alta densidad de plantación con 2,222 plantas ha-1 (3X1.5 m). El estudio se inició cuando las plantas tenían ocho años de edad y concluyó a la edad de 10. Con la densidad evaluada se obtuvo, en los tres años de estudio, una producción promediode fruto de 28 t ha-1. Un aspecto sobresaliente de la tecnología validada, es el sistema de podas, que permitió mantener arboles de talla baja y cosechar la fruta manualmente, sin usar implementos como escaleras o bajadores, evitando daños mecánicos; así mismo, esta tecnología permitió tener un control más eficiente de organismos plaga, facilitar el manejo de los programas de riego y fertilización. Cuando se aplica esta tecnología, después del octavo año de producción, el productor tiene ingresos anuales promedio de hasta $663,873.49 pesos (USD$33200.00) de utilidad por hectárea.

There is a worldwide demand for phenolic compounds (PC) because they exhibit several biological activities. This work aimed at extracting phenolic compounds from peanut meal. The methods of extraction were mainly: conventional solvent extraction (traditional methods) and ultrasound assisted extraction (recent methods) and comparing their results. Peanut meal (PM) was prepared by defatting with n-hexane, and then extracted by the two previous methods. First, the conventional solvents used were 80% methanol, ethanol, acetone, isopropanol, and distilled water. Then studied Different parameters such as meal: water ratio, also the effect of temperature and the pH on the extraction process. Second, ultrasonic assisted extractions (USAE), the parameters investigated were temperature, time and speed of sonication. Finally, all the extracts were analyzed by HPLC for their phenolic contents. Results indicated that the highest extracted PC achieved by solvents was in distilled water where 1:100, Meal: Water ratio which extracted 40 mg PC / g PM at 30& 35°C. Highest extracted PC was achieved by alkaline medium at pH 12 more than acidic and neutral medium. While (USAE) at speed 8 ultrasonication and temperature 30ᵒC, extracted 49.2mg PC /g PM. Sothe ultrasound assisted extraction exhibited great influence on the extraction of phenolic compounds from peanut meal. The ultrasonic peanut extract was examined for its antioxidant, antimicrobial and anticarcinogenic activities. The antioxidant activity of PM phenolic extract prepared by ultrasonic technique, was measured by, β-carotene, and DPPH methods, and reducing antioxidant power. Results revealed values: 84.57, 57.72 and 5960 respectively. The PM extract showed different levels of antimicrobial activity against the pathogenic bacteria used. As for the anticarcinogenic effect PM phenolic extract most effective on inhibiting colon carcinoma and lung carcinoma cell lines with IC50 = 20.7 and 20.8 µ/ml., respectively. This was followed by intestinal carcinoma and liver carcinoma cell lines with IC50= 39.6 and 40.2µ/ml.

Ficus benghalensis is one of those taboo plants in India, which was claimed to be possessed and have weird effects on human health. Apart from this ficus species has a great variety of chemical constituents and an abundant amount of antioxidants. Drying is the most critical stage of improving the activity or preventing the loss of chemical components from a drug. There is another stage of ensuring high chemical constituent content in the plant and that is the extraction procedure. So the point of focus in the current research is to find the effect of extraction method and drying on the anti-inflammatory potential of the plant. The result of the extraction method and drying method of the plant was investigated and found that the ultrasound-assisted extraction of the shade dried leaves was found to give the highest yield of flavonoids and activity.
