Recently, the antibacterial activity of a composite resin containing prereacted glass ionomer (S-PRG) filler was revealed. We examined the effect of an S-PRG eluate on various biologic activities ofStreptococcus mutansandPorphyromonas gingivalis. Adherence ability ofS. mutanswas evaluated by microtiter plate assay; protease and gelatinase activities ofP. gingivaliswere examined by synthetic substrate hydrolysis and gelatin film spot assay, respectively. Coaggregation ofP. gingivaliswithFusobacterium nucleatumwas also examined. S-PRG eluate was found to suppress streptococcal adherence. S-PRG eluate inhibited the protease and gelatinase activities ofP. gingivalisand the coaggregation betweenP. gingivalisandF. nucleatum. These results indicate that S-PRG eluate suppresses streptococcal adherence and inhibits the protease and coaggregation activities ofP. gingivalis. These findings may prompt research into novel strategies for preventing caries and periodontitis.

SUMMARY The purpose of this study was to investigate the effects of surface roughness of resin composite on biofilm formation of Streptococcus mutans in the presence of saliva. To provide uniform surface roughness on composites, disks were prepared by curing composite against 400-grit silicon carbide paper (SR400), 800-grit silicon carbide paper (SR800), or a glass slide (SRGlass). The surface roughness was examined using confocal laser microscopy. For biofilm formation, S. mutans was grown for 24 hours with each disk in a biofilm medium with either glucose or sucrose in the presence of fluid-phase or surface-adsorbed saliva. The adherent bacteria were quantified via enumeration of the total viable counts of bacteria. Biofilms were examined using scanning electron microscopy. This study showed that SR400 had deeper and larger, but fewer depressions than SR800. Compared to SRGlass and SR800, biofilm formation was significantly increased on SR400. In addition, the differences in the effect of surface roughness on the amount of biofilm formation were not significantly influenced by either the presence of saliva or the carbohydrate source. Considering that similar differences in surface roughness were observed between SR400 and SR800 and between SR800 and SRGlass, this study suggests that surface topography (size and depth of depressions) may play a more important role than surface roughness in biofilm formation of S. mutans.

Abstract Although fluoride has been widely used in the prevention of dental caries, the effect of fluoride on the activity of biofilm in different stages of cariogenic biofilm formation is less studied. This study aimed to investigate the antibiofilm activity of sodium fluoride during early and mature Streptococcus mutans (S. mutans) biofilms formation. S. mutans biofilms were formed on saliva-coated hydroxyapatite disks. In the early (0 ~ 46 h) and mature (46 ~ 94 h) biofilm stages, the biofilm was treated with different concentrations of fluoride (250, 500, 1000, 2000 ppm; 5 times in total, 1 min/treatment). Acidogenicity, dry weight, colony-forming units, water-soluble/insoluble extracellular polysaccharides (EPS), and intracellular polysaccharides were analyzed and confocal laser scanning microscopy images were obtained of the two stages of biofilms (early and mature biofilms). To determine the antibiofilm activity of sodium fluoride during the formation of early and mature biofilms, and to evaluate the relationship between different concentrations of sodium fluoride and antibiofilm activity. In the early cariogenic biofilm formation stage, all fluoride concentration test groups (250, 500, 1000, 2000 ppm) significantly inhibited the growth of S. mutans biofilm. The antibiofilm and anti-EPS formation activities of the brief fluoride treatment increased in a concentration-dependent pattern. At the mature biofilm stage, only the 2000 ppm fluoride treatment group significantly inhibited biofilm accumulation, activity, and intracellular/extracellular polysaccharide content compared with the control and other fluoride treatment groups. The antimicrobial activity of fluoride is related to the formation stage of cariogenic biofilm. The early formation stage of cariogenic biofilm is more susceptible to the inhibition of fluorine than the mature stage. The fluoride treatment in the early formation stage of cariogenic biofilm may be an effective means to control the development of cariogenic biofilm and prevent caries.
