The mature thread-like spermatozoon of Paragonimus ohirai was shown to be composed of 7 distinct segments which have morphologically different characteristics, following staining with silver nitrate. The first segment (1S) is a needle-shaped structure with a perforatorium. The 2S is composed of two filaments loosely paired; 3S consists of a somewhat coarse thread (vestige of the cytoplasmic projection); 4S is composed of two flagella tightly paired; in 5S, two silver-positive filaments are independently seen as in 2S; 6S has a ‘spiral’ structure composed of two flagella; in 7S, two filaments are seen dissociated from each other. The sperm nucleus exists within the ‘spiral’ structure of 6S and this structure may serve to hold and/or protect the nucleus. The technique presented here is useful for gross morphological observations of the long sperm of flatworms.

The technique of surrounding virus particles with a neutralised electron dense stain was described at the Fourth International Congress on Electron Microscopy, Berlin 1958 (see Home & Brenner, 1960, p. 625). For many years the negative staining technique in one form or another, has been applied to a wide range of biological materials. However, the full potential of the method has only recently been explored following the development and applications of optical diffraction and computer image analytical techniques to electron micrographs (cf. De Hosier & Klug, 1968; Markham 1968; Crowther et al., 1970; Home & Markham, 1973; Klug & Berger, 1974; Crowther & Klug, 1975). These image processing procedures have allowed a more precise and quantitative approach to be made concerning the interpretation, measurement and reconstruction of repeating features in certain biological systems.

After intoxication of rabbits with certain substances such as convulsant agents (3-acetylpyridine), centrally acting drugs (reserpine), or toxic metal compounds (tetraethyl lead) a significant observation by phase microscope is the loss of contrast of the hippocampal mossy fiber layer. It has been suggested that this alteration, as well as changes seen with the electron microscope in the hippocampal mossy fiber boutons, may be related to a loss of neurotransmitters. The purpose of these experiments was to apply the OsO4-zinc-iodide staining technique to the study of these structural changes since it has been suggested that OsO4-zinc-iodide stain reacts with neurotransmitters (acetylcholine, catecholamines).Domestic New Zealand rabbits (2.5 to 3 kg) were used. Hippocampal tissue was removed from normal and experimental animals treated with 3-acetylpyridine (antimetabolite of nicotinamide), reserpine (anti- hypertensive/tranquilizer), or iproniazid (antidepressant/monamine oxidase inhibitor). After fixation in glutaraldehyde hippocampal tissue was treated with OsO4-zinc-iodide stain and further processed for phase and electron microscope studies.

Silver impregnation techniques have been used for the demonstration of the complex carbohydrates in electron microscopy. However, the silver stains were believed to be technically sensitive and time consumming to perform. Currently, due to the need to more specifically evaluate immune complex for localization in certain renal diseases, a simplified procedure in conjunction with the use of the microwave has been developed and applied to renal and other biopsies. The procedure is as follows:Preparation of silver methenamine solution:1. 15ml graduated, clear polystyrene centrifuge tube (Falcon, No. 2099) was rinsed once with distilled water.2. 3% hexamethylene tetramine (methenamine) was added into the centrifuge tube to the 6ml mark.3. 3% silver nitrate was added slowly to the methenamine to the 7ml mark while agitating. (Solution will instantly turn milky in color and then clear rapidly by mixing. No precipitate should be formed).4. 2% sodium borate was added to the solution to the 8ml mark, mixed and centrifuged before use.

Hemocyanin, the respiratory pigment of the scorpion Androctonus australis is composed of 24 kidney shaped subunits. A model of architecture supported by many indirect arguments has been deduced from electron microscopy (EM) and immuno-EM. To ascertain, the disposition of the subunits within the oligomer, the 24mer was submitted to three-dimensional reconstruction by the method of single-exposure random-conical tilt series.A sample of native hemocyanin, prepared with the double layer negative staining technique, was observed by transmisson electron microscopy under low-dose conditions. Six 3D-reconstructions were carried out indenpendently from top, side and 45°views. The results are composed of solid-body surface representations, and slices extracted from the reconstruction volume.The main two characters of the molecule previously reported by Van Heel and Frank, were constantly found in the solid-body surface representations. These features are the presence of two different faces called flip and flop and a rocking of the molecule around an axis passing through diagonnally opposed hexamers. Furthermore, in the solid-body surface of the top view reconstruction, the positions and orientations of the bridges connecting the half molecules were found in excellent agreement with those predicted by the model.

Aim: The aimed study was to evaluate the influence of silver nitrate on surfacehardness and tensile strength of acrylic resins.Materials and methods: A total of 60 specimens were made from heat polymerizingresins. Two mechanical tests were utilized (surface hardness and tensile strength)and 4 experimental groups according to the concentration of silver nitrate used.The specimens without the use of silver nitrate were considered as control. Fortensile strength, all specimens were subjected to force till fracture. For surfacehardness, the specimens were tested via a durometer hardness tester. Allspecimens data were analyzed via ANOVA and Tukey tests.Results: The addition of silver nitrate to acrylic resins reduced significantly thetensile strength. Statistically, highly significant differences were found among allgroups (P≤0.001). Also, the difference between control and experimental groupswas highly significant (P≤0.001). For surface hardness, the silver nitrate improvedthe surface hardness of acrylics. Highly significant differences were statisticallyobserved between control and 900 ppm group (P≤0.001); and among all groups(P≤0.001)with exception that no significant differences between control and150ppm; and between 150ppm and 900ppm groups(P>0.05).Conclusion: The addition of silver nitrate to acrylics reduced significantly the tensilestrength and improved slightly the surface hardness.

Silver nanoplates (SNPs) having different size were synthesized by a seed-mediated method. The seeds -silver nanoparticles with 4 – 6 nm diameters were synthesized first by reducing silver nitrate with sodium borohydride in the present of Trisodium Citrate and Hydrogen peroxide. Then these seeds were developed by continue reducing Ag\(^+\) ions with various amount of L-Ascorbic acid to form SNPs. Our analysis showed that the concentratrion of L-Ascorbic acid, a secondary reducing agent, played an important role to form SNPs. In addition, the size and in-plane dipole plasmon resonance wavelenght of silver nanoplates were increased when the concentration of added silver nitrate increased. The characterization of SNPs were studied by UV-Vis, FE-SEM, EDS and TEM methods.
