The purpose of work was the research of the immune status and allocation of immunologic phenotypes at patients with the intolerance of stomatologic materials (ISM). Examination 91 patients aged from 46 up to 65 years, with complaints to ISM is conducted. As a result of a research of the T-cellular link of system of immunity at patients with an intolerance of stomatologic materials, but without the taped hypersensitivity to their components, taped the following patterns: statistically reliable (р<0,05) depression of the CD3+ levels of the T-general (47,9±3,7 at norm in control group – 66,2±4,5) and CD4+ of T-helpers (30,6±2,8 at norm of donors 38,2±3,2). It is noted reliable (р<0,05) depression of IRI (1,49±0,32 at norm of donors 1,76±0,36). At patients with complaints to an intolerance of stomatologic materials and the taped hypersensitivity, by results of application assays and reaction of allergenindutsirovanny damage of leucocytes the immune status was characterized reliable (р<0,05) by rising of quantity of the lymphocytes carrying CD25+ markers for 36,2% in comparison with those in control group (р<0,05). The augmentation of IRI (2,33±0,54) in comparison with control group (1,76±0,36) (р<0,05), generally at the expense of the increased CD4+ level of T-helpers and relative depression the maintenance of CD8+ of T lymphocytes to 18,3±3,6% is established (patients of control group have 21,7±3,09%). At that group depression of the IgA level (р<0,05) in blood serum to 1,36±0,12 g/l and in oral liqid to 0,042±0,0023 g/l is taped, and patients of control group have 2,57±0,06 g/l and 0,11±0,04 g/l respectively. The lowered activity of neutrocytes by results of spontaneous (8,4±3,5%) and induced (41,4±5,3%) is established to test NST at patients with ISM without hypersensitivity to dental alloys that authentically (р<0,05) differed with similar indicators of patients with ISM and the taped hypersensitivity to components of dental alloys. Indicators of the increased metabolic activity of neutrophils at patients with complaints to NSM and the taped hypersensitivity to dental alloys and their components defined in the NST-test correlated with: levels IgA of stomatic liquid (r=-0,55; р<0,05); quantity of dentoprosthetic units (r=0,72; р<0,05); terms of use of dentoprosthetic designs (r=0,47; р<0,05). The data obtained by us show two immunologic phenotypes of ISM: one with a hypersensitivity to them, the second with its absence. At both phenotypes signs of deficiency of local and general immunity which differed at these phenotypes are taped.

The tumor immunological microenvironment in hepatocellular carcinoma (HCC) is the T-helper (Th) 2 dominant inhibition state. Improving the immunosuppressive tumor microenvironment represents an important strategy for HCC treatment. TSLP-OX40L pathway is a target to improve Th2 immunosuppression. Yupingfeng granule (YPF) is clinically used to effectively improve the immune status of HCC. In this study, YPF increased the percentage of mature dendritic cells (DCs) and decreased levels of TSLP, TSLPR, and OX40L in tumor and adjacent tissues of the orthotopic-HCC mice model. This occurs together with the decreased levels of Th2 cytokines and increased levels of Th1 cytokines and Th1/Th2 ratio. In vitro experiment showed that YPF not only increased the percentage of mature DCs and stimulated IL-12 secretion in DCs but also reduced the positive rate of OX40L expression, decreased the proportion of CD4+ IL-13+ T cells, increased levels of Th1 cytokines, and decreased levels of Th2 cytokines from TSLP-treated DCs. In summary, these findings demonstrated that YPF promoted the maturation of DCs, decreased OX40L in TSLP-induced DCs, and improved the immunosuppressive state of Th2 in HCC microenvironment. Our results suggest that the mechanism underlying the improving effect of YPF on the immunosuppression is related to the DC-mediated TSLP-OX40L pathway.

Purpose. Mesenchymal-epithelial transition (MET), a reverse biological process to epithelial-mesenchymal transition (EMT), is involved in tumor metastasis and invasion. However, the role of MET-related genes (MRGs) in hepatocellular carcinoma (HCC) prognosis remains unclear. Methods. In this research, we obtained MRGs data and clinical information from public databases. In the TCGA dataset, a prognostic signature for HCC was constructed by the least absolute shrinkage and selection operator (LASSO) method and externally verified using the ICGC dataset. Results. There were 148 differentially expressed MRGs (DEMRGs), out of which 37 MRGs were found associated with overall survival (OS) in the univariate Cox analysis. A novel signature integrating of 5 MRGs was constructed, which split patients into high- and low-risk groups. Kaplan–Meier analysis revealed that high-risk patients had unfavorable OS than those low-risk counterparts. Receiver operating characteristic curve (ROC) showed great performance of this signature in predictive ability. Multivariate Cox analysis confirmed that this signature could independently predict HCC prognosis. The analysis of immune cell infiltration demonstrated that immune status varied differently between high- and low-risk groups. The analysis of clinicopathological characteristics suggested that tumor grade, clinical stage, and T stage were different between risk groups. The analysis between this signature and chemotherapeutic efficacy and immunosuppressive molecules indicated that this signature could serve as a promising predictor. Conclusions. In conclusion, we constructed and verified a novel signature from the perspective of MET, which was significantly associated with HCC prognosis, clinicopathological features, immune status, chemotherapeutic efficacy, and immunosuppressive biomarkers.

This study examined the immunity dynamics of bovine brucellosis-complicated leukemia, as well as the efficacy of treatment with betulin. Blood serum samples were taken from cows with specific brucellosis antibodies and bovine leukemia virus antibodies based on the results of complex diagnostic studies. Animals (n = 5) selected based on serological studies were injected subcutaneously with betulin-PEG three times with an interval of 7 days at a dose of 10 mg/kg. Blood was taken for hematological and immunological studies before the injection, and after the first, second and third injections. It was found that leukemia and brucellosis infections were accompanied by a significant increase in the number of leukocytes and lymphoid cells, as well as strong fluctuations in the leukocyte functional and metabolic activity parameters (from insignificant in some individual animals to significantly increased in others). Immunological restructuring of the body after triplicate administration of betulin-PEG was characterized by the restoration of the number of lymphoid cells to the level of clinically healthy animals, as well as a significant increase in the functional and metabolic activity of the neutrophilic granulocytes, especially their oxygen-dependent systems. Based on the results, the product is recommended as an effective means to restrain the leukemia process that is complicated with associated infection, as well as to use for seronegative animals in order to increase nonspecific resistance to BLV infection. BackgroundImmunotherapy and sorafenib exert anti-tumor effects via ferroptosis, but reliable biomarkers for the individual treatment and prognosis prediction of hepatocellular carcinoma (HCC) based on the ferroptosis and immune status remain lacking.MethodsFerroptosis-related genes (FRGs) were identified by downloading data from FerrDb and by searching and reading original articles from PubMed. Immune-related genes (IRGs) were downloaded from ImmPort. Prognostic FRGs and IRGs in the GSE14520 (n = 220) and The Cancer Genome Atlas (TCGA, n = 365) datasets were identified. Least absolute shrinkage and selection operator (LASSO) Cox regression and multivariate Cox regression were used for model construction. Ferroptosis expression profiles, the infiltration of immune cells, and the somatic mutation status were analyzed and compared.ResultsTwenty-seven prognostic ferroptosis- and immune-related signatures were included to construct a comprehensive index of ferroptosis and immune status (CIFI). A subgroup of patients was identified as having a high CIFI value, which was associated with a worse prognosis. This subgroup of patients had significantly up-regulated expressions of many suppressors of ferroptosis and higher fractions of immunosuppressive cells, such as cancer-associated fibroblasts (CAFs) and myeloid-derived suppressor cells (MDSCs). Notably, somatic mutation analysis indicated that high-CIFI patients had higher levels of tumor heterogeneity and higher mutation frequencies of genes like TP53.ConclusionIn this work, a novel prognostic classifier was developed based on ferroptosis- and IRGs in HCC, and this classifier could be used for prognostic prediction and the selection of patients for immunotherapies and targeted therapies.
