Young Merino ewes on five commercial properties in northern New South Wales were supplemented with selenium and their production was compared with untreated flock mates. They were studied from weaning till first lambing at about two years of age. Treatments of 5 mg selenium were given orally every six weeks for approximately 12 months. Copper treatments were also included to test for a possible concurrent deficiency or interaction with selenium. There were significant responses to selenium in liveweight in four of the five flocks and in wool production in two of the flocks at both shearings. Reproductive performance at first mating was also significantly better in two flocks. There were no beneficial effects of copper treatment nor were there any significant interactions with selenium treatment in any aspect of production measured.

Infection with Aleutian mink disease virus (AMDV) has negative effects on reproductive performance and survival rate of American mink (Neovison vison). The objectives of this study were to assess the effects of kelp (Ascophylum nodosum) supplementation on survival, growth rate, and reproductive performance of mink challenged with AMDV. AMDV-free female black mink (n = 75) were intranasally inoculated with a local AMDV strain. Mink were fed a commercial pellet supplemented with 1.5% or 0.75% kelp or were kept as controls (received no kelp) for 451 d. Body weight and rectal temperature were recorded on days 0, 31, 56, 99, 155, 366, and 451 post inoculation (PI). Annual mortality rates were 13.6%, 20.0%, and 31.8% for mink fed 1.5%, 0.75%, or 0.0% kelp, respectively (P = 0.29). Mink which were fed 1.5% kelp had a significantly (P < 0.01) greater daily weight loss during breeding and post-breeding periods (days 155–366 PI), and outperformed (P < 0.01) the other groups in regard to litter sizes at birth and weaning. Differences among treatments were not significant for the number of females mated, or whelped of those exposed to males, kit survival from birth to weaning, or rectal temperature. It was concluded that 1.5% kelp supplementation had beneficial effects on survival rate of adult mink and litter size.

Abstract Yucca is an important source of biologically active substances such as steroidal saponins and stilbenes providing many beneficial effects when administered to humans and other animals. These substances offer a great potential in the prevention and treatment of current civilized diseases as well as to their: antioxidant, hypocholesterolaemic, anti-inflammatory, phytoestrogenic, pro-apoptotic, anti-proliferative, and anti-carcinogenic properties. This review focuses on the roles of two main yucca constituent groups and their ability to modulate ovarian functions and female reproductive performance. Both the biological activity of yucca substances and the mechanisms of their actions on ovaries are still incompletely understood. Thus, the direct effects of yucca extract on ovarian cells in animal models under in vitro conditions, as well as actions after yucca consumption will be discussed.

AbstractDairy farming in tropical climates is challenging as heat stress can impair reproduction in cows. Previous studies have demonstrated the beneficial effects of beta-carotene supplementation on bovine reproductive performance. This study was performed in Thailand, where the temperature-humidity index (THI) during the experimental periods was measured to range from 78.4 to 86.1. Lactating Holstein cows classified as repeat breeders (previous artificial insemination [AI] failures) were randomly assigned into two treatments, control treatment (T1; received placebo, n = 200) and test treatment (T2; received 400 mg/h/day of beta-carotene, n = 200). All cows were subjected to a protocol for synchronization of ovulation and timed artificial insemination (TAI). The day of the 1st ovulation synchronized protocol was defined as day 0, and the total experimental period was 160 days. Daily placebo or beta-carotene supplements were given orally on day 0 and each subsequent day of the experiment. Diagnosis of pregnancy was performed using ultrasound on day 30 after insemination. Non-pregnant cows were subjected to further ovulation synchronizations (maximum of four) and TAI over a period of 160 days. Milk samples were collected every ten days throughout the experiment. The samples were analyzed for beta-carotene concentration, superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities. The pregnancies per AI of the cows in T2 were significantly greater than that of T1 from the 2nd to 4th TAI. During the entire experimental period, the pregnancies in T2 were significantly greater than that of T1. Cox's proportional hazards regression model data indicated a 44% greater probability of pregnancy for cows receiving beta-carotene. The concentrations of milk beta-carotene in T2 were significantly greater than T1 from the 2nd to 4th TAI. Significantly greater SOD and GPx activities were observed in T2 than T1, suggesting a reduction of oxidative stress in cows treated with beta-carotene. Dietary supplementation with beta-carotene thus improves the reproductive performance of repeat breeders exposed to heat stress, possibly by reducing oxidative stress.

Abstract Leydig cells play a critical role in male reproductive physiology, and their dysfunction is usually associated with male infertility. Melatonin has an important protective and regulatory role in these cells. However, the lack of suitable animal models impedes us from addressing the impact of endogenous melatonin on these cells. In the current study, by using AANAT overexpression transgenic sheep and AANAT knockout mice, we confirmed the regulatory effects of endogenously occurring melatonin on Leydig cells as well as its beneficial effects on male reproductive performance. The results showed that the endogenously elevated melatonin level was correlated with decreased Leydig cell apoptosis, increased testosterone production and improved quality of sperm in melatonin-enriched transgenic mammals. Signal transduction analysis indicated that melatonin targeted the mitochondrial apoptotic Bax/Bcl2 pathway and thus suppressed Leydig cell apoptosis. In addition, melatonin upregulated the expression of testosterone synthesis-related genes of StAR, SF1 and Gata4 in Leydig cells. This action was primarily mediated by the melatonin nuclear receptor RORα since blockade of this receptor suppressed the effect of melatonin on testosterone synthesis. All of these actions of melatonin cause Leydig cells to generate more testosterone, which is necessary for spermatogenesis in mammals. In contrast, AANAT knockout animals have dysfunctional Leydig cells and reduced reproductive performance.

Reproductive failure is a source of major economic loss to the UK pig industry, accounting for 0.5 of all first parity gilt cullings (MLC Pig Year Book, 1995). Previous research (Cameron et al, 1999) has shown that rearing gilts on a diet formulated to support maximal protein deposition has beneficial effects on ovulation rate at 3rd oestrus. The aim of the current experiment was to develop further the previous model to assess long and short-term effects of protein nutrition on reproductive performance, body composition and metabolic status.

The Family

For several decades, it has been known that oestrogens are essential for human health. The discovery that there are two oestrogen receptors (ERs), ERalpha and ERbeta, has facilitated our understanding of how the hormone exerts its physiological effects. The ERs belong to the family of ligand-activated nuclear receptors, which act by modulating the expression of target genes. Studies of ER-knockout (ERKO) mice have been instrumental in defining the relevance of a given receptor subtype in a certain tissue. Phenotypes displayed by ERKO mice suggest diseases in which dysfunctional ERs might be involved in aetiology and pathology. Association between single-nucleotide polymorphisms (SNPs) in ER genes and disease have been demonstrated in several cases. Selective ER modulators (SERMs), which are selective with regard to their effects in a certain cell type, already exist. Since oestrogen has effects in many tissues, the goal with a SERM is to provide beneficial effects in one target tissue while avoiding side effects in others. Refined SERMs will, in the future, provide improved therapeutic strategies for existing and novel indications.
