Abstract The current drug therapy for schizophrenia effectively treats acute psychosis and its recurrence; however, this mental disorder’s cognitive and negative symptoms are still poorly controlled. Antipsychotics present important side effects, such as weight gain and extrapyramidal effects. The essential oil of Alpinia zerumbet (EOAZ) leaves presents potential antipsychotic properties that need further preclinical investigation. Here, we aimed to determine the effects of EAOZ in the prevention and reversal of schizophrenia-like symptoms (positive, negative, and cognitive) induced by ketamine (KET) repeated administration in mice and putative neurobiological mechanisms related to this effect. To this end, we evaluated antioxidant (GSH, nitrite levels), anti-inflammatory [interleukin (IL)-6], and neurotrophic [brain-derived neurotrophic factor (BDNF)] effects of this oil in hippocampal tissue. The atypical antipsychotic olanzapine (OLZ) was used as standard drug therapy. EOAZ, similarly to OLZ, prevented and reversed most KET-induced schizophrenia-like behavioral alterations, i.e., sensorimotor gating deficits and social impairment. EOAZ had a modest effect on the prevention of KET-associated working memory deficit. Compared to OLZ, EOAZ showed a more favorable side effects profile, inducing less cataleptic and weight gain changes. EOAZ efficiently protected the hippocampus against KET-induced oxidative imbalance, IL-6 increments, and BDNF impairment. In conclusion, our data add more mechanistic evidence for the anti-schizophrenia effects of EOAZ, based on its antioxidant, anti-inflammatory, and BDNF up-regulating actions. The absence of significant side effects observed in current antipsychotic drug therapy seems to be an essential benefit of the oil.

In phytotherapy, the essential oil from the leaves of Alpinia zerumbet ( Alpinia speciosa K. Schum.) (EOAZ) is used for neuropsychiatric symptoms, such as depression, stress and anxiety, and chronic problems that are associated with reproductive hormone imbalances in women. The chemical composition of EOAZ was analyzed by GC/MS, and the EOAZ properties inducing behavioral alterations in mice were examined by behavioral observations (BO) and an elevated plus-maze task (EPM), widely used as a method for assessing anxiolytic-like behaviors. Five major compounds, p-cymene (28.0 ± 5.0%), 1,8-cineole (17.9 ± 4.2%), terpinen-4-ol (11.9 ± 6.3%), limonene (6.3 ± 2.2%), and camphor (5.2 ± 2.1%) were identified by retention indices, mass spectra and comparison with standards. Inhalational administration of EOAZ (8.7 ppm) induced unique jumping behaviors in mice. To further investigate the behavioral regulatory mechanisms of EOAZ, we administered an intraperitoneal injection of either 10 mg/kg 5-HTP or 10 mg/kg fluoxetine prior to the EOAZ inhalations. By 5-HTP or fluoxetine pretreatments, the jumping frequencies were significantly decreased. In EPM, EOAZ (0.087 and 8.7 ppm) obviously showed the anxiolytic-like activity in mice.

Alpinia zerumbet (Pers.) B.L. Burtt. & R.M. Sm is an important perennial ginger plant in the tropics with various uses as foods, dietary supplement, cosmetics, essential oil production and traditional medicines. In this study, essential oils, phenolic acids, fatty acids, kavains, total phenols and antioxidant activity in A. zerumbet plants exposed to UV-C were investigated. Number and weights of essential oils increased rapidly from 1 to 3 days of exposure. Major extend of essential oils was recorded in methyl cinnamate and followed by benzyl acetone. The correspondence to the accumulation of palmitic acid was 15 folds, in contrast to the strong reduction of stearic quantity was 10 folds at 3 days of treatment. Changes in chemical components and antioxidant capacity in A. zerumbet are differed reversely between the exposure of UV-C and heavy toxic chemicals. The exposure of UV-C is beneficial for the increasing of essential oil production in A. zerumbet.
