Oncogenic viruses, like viruses in general, can be divided into two classes, those that contain deoxyribonucleic acid (DNA) and those that contain ribonucleic acid (RNA). The RNA viruses have been recovered readily from the tumors which they cause whereas, the DNA-virus induced tumors have not yielded the virus. Since DNA viruses cannot be recovered, the bulk of present day investigations have been concerned with RNA viruses.The Lucké renal adenocarcinoma is a spontaneous tumor which occurs in northern leopard frogs (Rana pipiens) and has received increased attention in recent years because of its probable viral etiology. This hypothesis was first advanced by Lucké after he observed intranuclear inclusions in some of the tumor cells. Tumors with inclusions were examined at the fine structural level by Fawcett who showed that they contained immature and mature virus˗like particles.The use of this system in the study of oncogenic tumors offers several unique features, the virus has been shown to contain DNA and it can be recovered from the tumor, also, it is temperature sensitive. This latter feature is of importance because the virus can be transformed from a latent to a vegetative state by lowering or elevating the environmental temperature.

Bergenin Exhibits Hepatoprotective Activity Against Ethanol-Induced Oxidative Stress in ICR Mice

Bergenin is a C-glucoside derivative of gallic acid but its antioxidant and hepatoprotective effects have not previously been compared with gallic acid. Male ICR mice were administered bergenin (10, 50, and 250 mg/kg/day) or gallic acid (100 mg/kg/day) for 7 consecutive days before a single administration of ethanol (5 g/kg). Liver sections were histopathologically examined. Aspartate aminotransferase, alanine aminotransferase, reactive oxygen species, and malondialdehyde levels were determined in plasma. Total glutathione, reduced glutathione, and oxidized glutathione levels were determined in liver homogenates. Ethanol induced hepatic injury with prominent histopathological markers including nuclear pyknosis and necrotic areas and this accorded with increases in the plasma levels of aspartate aminotransferase, alanine aminotransferase, reactive oxygen species, and malondialdehyde. Moreover, ethanol disturbed hepatic glutathione homeostasis by reducing glutathione stores. Hepatic injury in the ethanol-induced mice was prevented with bergenin and gallic acid by significant decreases in plasma aspartate aminotransferase, alanine aminotransferase, reactive oxygen species, and malondialdehyde levels and restoration of the hepatic glutathione profile through an increase in the reduced glutathione/oxidized glutathione ratio. Bergenin at 10 mg/kg/day showed comparable hepatoprotective activity to gallic acid in an ethanol-induced mouse model of oxidative stress. Therefore, bergenin might be a promising candidate for further development as a novel hepatoprotective product.

Metastasis of the kidney carcinoma of leopard frogs (Rana pipiens) has been induced by exposing tumor-bearing animals for approximately 50 days to a constant temperature of 28°C. Under these conditions 54 per cent of the frogs developed secondary growths, whereas in groups kept at 18° or at 7° metastatic dissemination was found in only 6 per cent. Moreover, at the elevated temperature the metastases were usually more numerous and more widely disseminated; they were also fairly uniform in size, suggesting that they had developed at nearly the same time. Dissemination of the kidney tumors was influenced by the nutritional state of the frogs, occurring more readily in well nourished than in poorly nourished animals. Periodic Roentgen ray examinations showed that the size of the primary tumors was not significantly or uniformly affected during the course of the experiments. No correlation was found between change in size of the kidney tumors and the incidence of their metastasis. Although the mechanism by which temperature induces metastasis of frog carcinoma cannot as yet be elucidated, previous experiments with this tumor indicate that certain factors at least may be involved: Elevation of temperature has been found to cause more ready detachment of cells of frog carcinoma in tissue culture; to bring about increased velocity of locomotion of the detached cells; to lead more promptly and efficiently to vascularization of transplants; and to effect their greater invasiveness.
