Global fire regimes are changing, with increases in wildfire frequency and severity expected for many North American forests over the next 100 years. Fires can result in dramatic changes to C stocks and can restructure plant and microbial communities, which can have long-lasting effects on ecosystem functions. We investigated wildfire effects on soil microbial communities (bacteria and fungi) in an extreme fire season in the northwestern Canadian boreal forest, using field surveys, remote sensing, and high-throughput amplicon sequencing. We found that fire occurrence, along with vegetation community, moisture regime, pH, total carbon, and soil texture are all significant predictors of soil microbial community composition. Communities become increasingly dissimilar with increasingly severe burns, and the burn severity index (an index of the fractional area of consumed organic soils and exposed mineral soils) best predicted total bacterial community composition, while burned/unburned was the best predictor for fungi. Globally abundant taxa were identified as significant positive fire responders, including the bacteria Massilia sp. (64x more abundant with fire) and Arthrobacter sp. (35x), and the fungi Penicillium sp. (22x) and Fusicladium sp. (12x) Bacterial and fungal co-occurrence network modules were characterized by fire responsiveness as well as pH and moisture regime. Building on the efforts of previous studies, our results identify specific fire-responsive microbial taxa and suggest that accounting for burn severity improves our understanding of their response to fires, with potentially important implications for ecosystem functions.

AbstractLittle is known about the relationship between soil microbial communities and soil properties in southern boreal forests. To further our knowledge about that relationship, we compared the soil samples in southern boreal forests of the Greater Khingan Mountains—the southernmost boreal forest biome in the world. The forests can be divided into boardleaf forests dominated by birch (Betula platyphylla) or aspen (Populus davidiana) and coniferous forests dominated by larch (Larix gmelinii) or pine (Pinus sylvestris var. mongolica). Results suggested different soil microbial communities and soil properties between these southern boreal forests. Soil protease activity strongly associated with soil fungal communities in broadleaf and coniferous forests (p < 0.05), but not with soil bacterial communities (p > 0.05). Soil ammonium nitrogen and total phosphorus contents strongly associated with soil fungal and bacterial communities in broadleaf forests (p < 0.05), but not in coniferous forests (p > 0.05). Soil potassium content demonstrated strong correlations with both soil fungal and bacterial communities in broadleaf and coniferous forests (p < 0.05). These results provide evidence for different soil communities and soil properties in southern boreal forest, and further elucidate the explicit correlation between soil microbial communities and soil properties in southern boreal forests.

ABSTRACT The Athabasca oil sand deposit is one of the largest single oil deposits in the world. Following surface mining, companies are required to restore soil-like profiles that can support the previous land capabilities. The objective of this study was to assess whether the soil prokaryotic alpha diversity (α-diversity) and β-diversity in oil sand soils reconstructed 20 to 30 years previously and planted to one of three vegetation types (coniferous or deciduous trees and grassland) were similar to those found in natural boreal forest soils subject to wildfire disturbance. Prokaryotic α-diversity and β-diversity were assessed using massively parallel sequencing of 16S rRNA genes. The β-diversity, but not the α-diversity, differed between reconstructed and natural soils. Bacteria associated with an oligotrophic lifestyle were more abundant in natural forest soils, whereas bacteria associated with a copiotrophic lifestyle were more abundant in reconstructed soils. Ammonia-oxidizing archaea were most abundant in reconstructed soils planted with grasses. Plant species were the main factor influencing α-diversity in natural and in reconstructed soils. Nitrogen deposition, pH, and plant species were the main factors influencing the β-diversity of the prokaryotic communities in natural and reconstructed soils. The results highlight the importance of nitrogen deposition and aboveground-belowground relationships in shaping soil microbial communities in natural and reconstructed soils. IMPORTANCE Covering over 800 km2, land disturbed by the exploitation of the oil sands in Canada has to be restored. Here, we take advantage of the proximity between these reconstructed ecosystems and the boreal forest surrounding the oil sand mining area to study soil microbial community structure and processes in both natural and nonnatural environments. By identifying key characteristics shaping the structure of soil microbial communities, this study improved our understanding of how vegetation, soil characteristics and microbial communities interact and drive soil functions.
