Dry bean proteins: extraction methods, functionality, and application in products for human consumption

Two extraction methods for subsequent gas chromatographic (GC) determination of volatiles from freshly harvested and aged fennel fruit samples ( Foeniculum vulgare Mill.,ssp. vulgare var. dulce) have been compared. Hydrodistillation followed by GC-FID and GC-MS analysis was used as a standard method for essential oil characterization, while static headspace followed by GC (SHS-GC-FID) was used as a comparative method for determination of volatile components. As the fennel fruit ages, there is a gradual loss of the volatile components as indicated by the lower yield of essential oil and lower content of volatiles, as indicated by the alternative SHS-GC-FID analysis. Slight differences observed for the main components ( trans-anethole, estragole, fenchone, and limonene) using the two methods are negligible, indicating that these volatiles did not undergo chemical transformation during the sample preparation procedures. A difference in anisaldehyde content was observed when the composition of the hydrodistilled essential oil was compared with the SHS-GC-FIDanalysis of volatiles and explanation for the variation of anisaldehyde content and the origin of other compounds was suggested. Comparison of the obtained results showed that limonene oxides, carvone and carveolare detectable in SHS-GC-FID analysis of the aged fennel fruits, while in hydrodistilled samples analyzed by GC-FID they were not present. Another observed difference was the appearance of products in significant amounts with higher retention times than trans-anethole, namely threo- and erythro-anethole β-hydroxymethylether and anethole glycol that are not detectable in the essential oil obtained by hydrodistillation. So, the relative abundance of the major components is comparable between these two methods for fennel seed up to 3 years from harvest and they can be used interchangeably depending on the purpose and amount of material. Furthermore, SHS-GC-FID can be used for assessment of maximum storage time and quality of fennel fruit suitable for human consumption.

Edible insects represent an interesting alternative source of protein for human consumption but the main hurdle facing the edible insect sector is low consumer acceptance. However, increased acceptance is anticipated when insects are incorporated as a processed ingredient, such as protein-rich powder, rather than presented whole. To produce edible insect fractions with high protein content, a defatting step is necessary. This study investigated the effects of six defatting methods (conventional solvents, three-phase partitioning, and supercritical CO2) on lipid extraction yield, fatty profiles, and protein extraction and purification of house cricket (Acheta domesticus) and mealworm (Tenebrio molitor) meals. Ethanol increased the lipid extraction yield (22.7%–28.8%), irrespective of the insect meal used or the extraction method applied. Supercritical CO2 gave similar lipid extraction yields as conventional methods for Tenebrio molitor (T. molitor) (22.1%) but was less efficient for Acheta domesticus (A. domesticus) (11.9%). The protein extraction yield ranged from 12.4% to 38.9% for A. domesticus, and from 11.9% to 39.3% for T. molitor, whereas purification rates ranged from 58.3% to 78.5% for A. domesticus and from 48.7% to 75.4% for T. molitor.

High-quality fish oil for human consumption requires low levels of toxic elements. The aim of this study was to compare different oil extraction methods to identify the most efficient method for extracting fish oil of high quality with the least contamination. The methods used in this study were Soxhlet extraction, enzymatic extraction, wet reduction, and supercritical fluid extraction. The results showed that toxic elements in fish oil could be reduced using supercritical CO2 at a modest temperature (60°C) and pressure (35 MPa) with little reduction in the oil yield. There were significant reductions in mercury (85 to 100%), cadmium (97 to 100%), and lead (100%) content of the fish oil extracted using the supercritical fluid extraction method. The fish oil extracted using conventional methods contained toxic elements at levels much higher than the accepted limits of 0.1 μg/g.

The edible bird nest (EBN) from Aerodramus fuciphagus has been consumed as a Chinese traditional food for health and medicinal purposes due to its elevated nutritional value. The present study focused on the influence of characterization and extraction methods on protein profiling, which could be a guideline for grading the EBN. The proposed extraction method is similar to the common food preparation methods of consumers and thus can accurately establish the bioactive protein available upon human consumption. The characterization includes physicochemical analysis (physical, morphology, elemental composition, and microbial content) and chemical analysis (crude protein and amino acid). The morphology of half-cup EBN was found to be uniformly shaped and rich in calcium as compared to rough surface of stripe-shaped EBN, and there was no significant microbial growth in both types of EBN. The crude protein and amino acid content in half-cup EBN were significantly higher than stripe-shaped EBN. The full stew (FS) and stew (SE) extraction methods produced a maximal yield of soluble protein. Sialic acid content in SE extract (8.47%, w/w) and FS extract (7.91%, w/w) were recorded. About seven parent proteins (39.15 to 181.68 kDa) were identified by LC-MS/MS Q-TOF, namely 78 kDa glucose-regulated protein, lysyl oxidase-3, Mucin-5AC-like, acidic mammalian chitinase-like, 45 kDa calcium-binding protein, nucleobindin-2, and ovoinhibitor-like. In conclusion, the characteristics and extraction methods influence the availability of bioactive protein and peptides, demonstrating the potential usage of EBN in improving its biological activities and nutritional properties.

Application of dry bean desiccants just prior to crop maturity is common practice by Canadian producers. As dry beans are grown for human consumption it is critical that producers pick desiccants that do not affect crop yield, seed quality, or result in desiccant seed residue levels above accepted levels. In this study the efficacy of glyphosate, diquat, glufosinate, carfentrazone, flumioxazin, and saflufenacil as desiccants was examined for navy, cranberry, pinto, and great northern dry bean. Seed herbicide residues were also tested for each of the dry bean classes tested. Navy, cranberry, pinto, and great northern dry bean yields were not impacted by use of the desiccants diquat, carfentrazone, flumioxazin, or saflufenacil when applied at labelled rates and application timings. Additionally, herbicide residues in seed following application remained lower than maximum residue limits (MRL) established by primary Canadian dry bean export partners. Generally, dry bean colour, irrespective of class, was not altered by desiccant use; diquat and flumioxazin caused minor increases in the degree of red and yellow seed pigmentation for cranberry bean only. Although colour differences were noted using a Chroma meter the differences were slight and would not likely be of economic importance. Application of glyphosate did not affect crop yield, and seed residue levels were below MRLs for navy, pinto, and great northern bean. However, seed glyphosate residue levels were above the MRL for cranberry bean when glyphosate was applied alone or tankmixed with carfentrazone, flumioxazin, or saflufenacil. Seed residue levels were also above listed MRLs for some export countries when glufosinate was applied to navy, cranberry, and pinto bean, although crop yield and seed quality remained unaffected. These findings suggest that growers and contractors should avoid using glufosinate as a dry bean desiccant at least for some markets and that care should be taken when selecting glyphosate as a desiccant, especially for cranberry bean. Across all market classes desiccation progress of bean leaf, stem, and pod tissue was slowest when glyphosate and carfentrazone were used.

The Leguminosae plant family comprises a large number of agricultural species, including important crops of field, vegetable and staple plants. According to the biological characteristics and significance for human consumption, the most prominent crop is bean, traditionally grown in Serbia either as dry bean or as a vegetable - green beans. Production of dry and green beans in our country have the trend of reduction of sowed areas, with small average yields and enormous international trade deficits. Due to these reasons and high domestic demand, there is great potential for dry and green bean production in Serbia. In many parts of the world, plant pathogenic fungi and fungi-like organisms cause high, and in some cases total, annual yield losses. On the other hand, mycoses of bean in our country are greatly understudied, even on basic levels such as presence and distribution of fungal pathogens. In this paper, we summarize the current knowledge on the most important pathogenic fungi infecting beans worldwide, particularly related to losses, symptomatology, survival and dispersal means. Furthermore, appropriate and available disease control measures are discussed as an encouragement and support for domestic production in obtaining high and safe yields.

Dry bean (Phaseolus vulgaris L.) consumption is associated with reduced risk for a number of chronic diseases. In westernised societies, dry bean consumption is particularly low (approximately 2–4 kg/capita per year) and little information is available about the safety of increasing dietary intake in humans to achieve levels that prevent and control chronic diseases. In anticipation of a human intervention study to address the safety and efficacy of increasing bean consumption, a dose–response study with dietary beans was conducted to establish whether increased bean consumption in rats exhibits changes indicative of hepatic stress or toxicity. Transcript levels from a panel of stress and toxicity-related genes were analysed in female Sprague–Dawley rats fed a dose range of dietary beans that bracketed amounts relevant to human consumption globally. Cooked red bean was incorporated into a purified diet formulation at 0, 7·5, 15, 30 or 60 % w/w for the assessment of adaptive patterns of gene expression using quantitative PCR array. Of the eighty-four genes evaluated, the expressions of Cyp3a11, Cyp7a1, Fmo1, Gstm1, Mif and Ugt1a6 were elevated, whereas the expression of Hspa8 was down-regulated. Liver gene expression was not modulated in a manner indicative of an adverse response. Only the expression of the cholesterol 7α hydoxylase and UDP-glucuronosyltransferase genes increased in a dose-dependent manner at nutritionally relevant dietary bean concentrations. These candidate genes may contribute to the health benefits attributed to increased bean consumption.
