Five Saccharomyces cerevisiae strains (Chinese indigenous yeasts SC5, WC5, SC8, CC17 and commercial starter F15) were inoculated into Cabernet sauvignon grape must and fermented at pilot scale. For the first time, combination of 1H NMR, HS-SPME/GC-MS and HPLC-DAD-ESI-MS/MS metabonomic profiling techniques was performed to analyze the global chemical fingerprints of sampled wines at the end of alcoholic and malolactic fermentation respectively, then 13 non-volatile flavor compounds, 52 volatile organic aromas and 43 polyphenolic molecules were identified and determined correspondently. All principal component analysis (PCA) of two fermentation stages based on the analytical results of 1H NMR, HS-SPME/GC-MS and HPLC-DAD-ESI-MS/MS divided these strains into three clusters: (1) SC5 and SC8, (2) WC5 and F15 and (3) CC17. The wine fermented by indigenous yeast, CC17, showed a very unique chemical profile, such as low pH and high color intensity, reduced amino acids (including proline) and the lowest total higher alcohols levels, most of the fixed acids, glycerol, ethyl esters and anthocyanins concentrations. The statistical results indicate that CC17 strain possesses very special anabolism and catabolism abilities on such substances in grape juice and has potentiality to produce characteristic wines with high qualities.

ABSTRACT Fatty acid ethyl esters (FAEEs) were produced in this study by the use of an engineered Escherichia coli p(Microdiesel) strain. Four fed-batch pilot scale cultivations were carried out by first using glycerol as sole carbon source for biomass production before glucose and oleic acid were added as carbon sources. Cultivations yielded a cell density of up to 61 ± 3.1 g of cell dry mass (CDM) per liter and a maximal FAEE content of 25.4% ± 1.1% (wt/wt) of CDM.

Fatty acid ethyl esters (FAEE), esterification products of fatty acids and ethanol, are in use as fatty acid supplements, but they also have been implicated as toxic mediators of ethanol ingestion. We hypothesized that hydrolysis of orally ingested FAEE occurs in the gastrointestinal (GI) tract and in the blood to explain their apparent lack of toxicity. To study the in vivo inactivation of FAEE by hydrolysis to free fatty acids and ethanol, we assessed the hydrolysis of FAEE administered as an oil directly into the rat stomach and when injected within the core of low-density lipoprotein particles into the circulation of rats. Our studies demonstrate that FAEE are rapidly degraded to free fatty acids and ethanol in the GI tract at the level of the duodenum with limited hydrolysis in the stomach. In addition, FAEE are rapidly degraded in the circulation, with a half-life of only 58 s. Thus the degradation of FAEE in the GI tract and in the blood provides an explanation for the apparent lack of toxicity of orally ingested FAEE.

Many current treatment options for lung inflammation and thrombosis come with unwanted side effects. The natural omega-3 fatty acids (O3FA) are generally anti-inflammatory and antithrombotic. The O3FA are always administered orally and occasionally by intravenous (IV) infusion. The main goal of this study is to determine if O3FA administered by inhalation of a nebulized formulation mitigates LPS-induced acute lung inflammation in male Wistar rats. Inflammation was triggered by intraperitoneal injection of LPS once a day for 14 days. One hour later, rats received nebulized treatments consisting of egg lecithin emulsified O3, budesonide and Montelukast, and blends of O3 and melatonin or Montelukast or Cannabidiol; O3 was in the form of free fatty acids for all groups except one group with ethyl esters. Lung histology and cytokines were determined in n=3 rats per group at day 8 and day 15. All groups had alveolar histiocytosis severity scores half or less than that of the disease control (Cd) treated with LPS and saline only inhalation. IL-6, TNF-α, TGF-β, and IL-10 were attenuated in all O3 groups. IL-1β was attenuated in most but not all O3 groups. O3 administered as ethyl ester was overall most effective in mitigating LPS effects. No evidence of lipid pneumonia or other chronic distress was observed. These preclinical data suggest that O3FA formulations should be further investigated as treatments in lung inflammation and thrombosis related lung disorders, including asthma, chronic obstructive pulmonary disease, lung cancer and acute respiratory distress like COVID-19.

The enzymes released into both human and rabbit plasmas by heparin injection hydrolyzed, in addition to triglyceride moieties of lipoproteins, a number of mono- and diglycerides of C16 and C18 fatty acids after in vitro addition of the unemulsified glycerides to the plasma. In human postheparin plasma, these enzymes also hydrolyzed glycerides of butyric and caproic acids. The pure triglycerides and methyl or ethyl esters of C16 and C18 fatty acids were not substrates. The heparin-released activities for the hydrolysis of glycerides added in vitro persisted after all activity for the lipolysis of lipoproteins had been destroyed by heat. These activities also differed from lipoprotein lipase activity with respect to the effects of 1 m NaCl, dialysis, and aging the plasma at 4 C. It appears that heparin releases into the blood more than one enzyme or more than one form of an enzyme which may be involved in a stepwise degradation to fatty acids and glycerol of the triglyceride moieties of lipoproteins of density less than 1.007 g/ml.

Chinese dome digesters are usually operated at long hydraulic retention times (HRT) and low influent total solids (TS) concentration because of limited mixing. In this study, a newly optimised Chinese dome digester with a self-agitating mechanism was investigated at a pilot scale (digester volume = 500 L) and compared with a conventional Chinese dome digester (as blank) at 15% influent TS concentration at two retention times (30 and 40 days). The reactors were operated at ambient temperature: 27–33 °C. The average specific methane production, volatile fatty acids and percentage of volatile solids (VS) reduction are 0.16 ± 0.13 and 0.25 ± 0.05L CH4/g VS; 1 ± 0.5 and 0.7 ± 0.3 g/L; and 51 ± 14 and 57 ± 10% at 40 days HRT (day 52–136) for the blank and optimised digester, respectively. At 30 days HRT (day 137–309) the results are 0.19 ± 0.12 and 0.23 ± 0.04 L CH4/g VS; 1.2 ± 0.6 and 0.7 ± 0.3 g/L; and 51 ± 9 and 58 ± 11.6%. Overall, the optimised digester produced 40% more methane than the blank, despite the high loading rates applied. The optimised digester showed superior digestion treatment efficiency and was more stable in terms of VFA concentration than the blank digester, can be therefore operated at high influent TS (15%) concentration.
