Abstract Because we recently observed two patients with severe diabetic hyperglycemia and spuriously elevated electronically determined hematocrit and mean corpuscular volume (MCV), we investigated the effect of hyperglycemia on two popular automated hematology systems, the Coulter S and Ortho ELT-8. Marked hyperglycemia (blood glucose 800-- 2000 mg/dl) caused consistent overestimation of the electronically determined MCV compared to that derived from a simultaneous spun microhematocrit. The resultant overestimation and underestimation, respectively, of the derived values for hematocrit and mean corpuscular hemoglobin concentration may be clinically misleading. The mechanism of MCV elevation in hyperglycemia appears to be swelling of hyperosmolar glucose “loaded” erythrocytes when diluted into “isotonic” counting medium. This effect is readily circumvented by determination of a spun microhematocrit.

Telah dilakukan penelitian untuk mengetahui nilai hematologi MCHC (Mean Corpuscular Hemoglobin Concentration), MCV (Mean Corpuscular Volume) dan MCH (Mean Corpuscular Haemoglobin) darah sapi bali yang dipelihara berbasis organik. Sistem pemeliharaan berbasis organik adalah manajemen pemberian pakan yang berasal dari lingkungan serta tidak menggunakan pestisida atau zat kimia lainnya. Nilai MCV, MCH dan MCHC darah sapi bali diukur dengan Veterinary hematology analyzer. Hasil penelitian menunjukan, nilai dari MCHC sebesar 39,17%/dl, MCV sebesar 48,44 fl dan nilai dari MCH sebesar 18,69pg. Dapat disimpulkan rerata nilai tersebut masih dalam batas normal nilai indeks eritrosit sapi pada umumnya.

Carbon tetrachloride (CCL4) is widely used in industry, toxic to the environment and humans, and most often used as a model of acute liver damage and liver fibrosis in experimental animals. Liver damage can deteriorate the hematological profile. The roots of cogongrass (Imperata cylindrica L.) have been used as traditional medicine due to its antioxidant activity. This study was conducted at the Faculty of Medicine, Universitas Padjadjaran, from January to March 2019. The study aimed to investigate whether the cogongrass roots ethanol extract (CGRE) can ameliorate the disturbance in the hematological profile in acute CCl4-injected mice. CGRE in dose 150 and 200 mg/kgBW was given orally to mice for four weeks before intraperitoneal injection of CCl4 1 mL/kgBW in olive oil (1:1 v/v). After 48 hours, mice were sacrificed, and the whole blood was drawn for hematological analysis. As a result, mean corpuscular volume (MCV) was reduced in CCl4-induction mice treated with CGRE in dose 150 mg/kgBW (49.25±3.06 vs 43.38±2.13 fl, p<0.05). This condition was followed by the improved hematocrit (Hct) and mean corpuscular hemoglobin concentration (MCHC). Platelet and platelet crit (Pct) levels were tended to decrease in CCl4-induction mice treated with CGRE in dose 150 mg/kgBW. In conclusion, CGRE dose 150 mg/kg BW can improve MCV, Hct, MCHC, platelet, and Pct in CCl4-injection mice. The antioxidant level in CGRE might facilitate it. EKSTRAK ETANOL AKAR ALANG-ALANG (IMPERATA CYLINDRICAL L.) MEMPERBAIKI PROFIL HEMATOLOGI PADA MENCIT YANG DIINJEKSI CARBON TETRACHLORIDECarbon tetrachloride (CCL4) banyak digunakan pada industri, bersifat toksik bagi lingkungan dan manusia, serta sering digunakan pada hewan coba untuk kerusakan liver akut dan fibrosis. Kerusakan liver dapat menyebabkan gangguan profil hematologi. Akar alang-alang (Imperata cylindrica L.) telah digunakan sebagai obat tradisional karena memiliki aktivitas antioksidan. Penelitian ini dilakukan di Fakultas Kedokteran Universitas Padjadjaran pada bulan Januari hingga Maret 2019. Tujuan penelitian ini adalah meneliti apakah ekstrak etanol akar alang-alang dapat memperbaiki gangguan profil hematologi pada mencit yang diinjeksi CCL4 secara akut. Ekstrak etanol akar alang-alang (EEAA) dosis 150 dan 200 mm/kgBB diberikan per oral kepada mencit selama empat minggu sebelum injeksi intraperitoneal CCl4 1 mL/kgBB yang dilarutkan dalam minyak zaitun (1:1 v/v). Setelah 48 jam, mencit dikorbankan dan diambil darahnya untuk pemeriksaan hematologi. Sebagai hasil, mean corpuscular volume (MCV) menurun pada mencit yang diinduksi CCl4 dengan perlakuan EEAA 150 mg/kgBB (49,25±3,06vs 43,38±2,13 fl, p<0,05). Keadaan ini diikuti dengan perbaikan hematokrit (Hct) dan mean corpuscular hemoglobin concentration (MCHC). Kadar platelet dan platelet crit (Pct) cenderung menurun pada mencit yang diinduksi CCl4 dengan perlakuan EEAA 150 mg/kgBB. Sebagai simpulan, EEAA dosis 150 mg/kgBB dapat memperbaiki MCV, Hct, MCHC, platelet, dan Pct pada mencit yang diinjeksi CCL4. Kemungkinan difasilitasi oleh antioksidan pada EEAA.

The aim of the current study was to compare feline hematologic variables in blood collected in microcapillary tubes (20 μl) and conventional blood tubes with the Medonic CA620/530 Vet in-house hematologic analyzer. A comparison of results obtained in 60 cats presented at the clinics of the veterinary school showed that the correlations between the 2 methods were 0.97 for white blood cell, 0.95 for red blood cell, and 0.93 for platelet counts; 0.92 for hemoglobin concentration; and 0.99 for mean corpuscular volume. No clinically relevant differences between the 2 blood sampling techniques were observed for any variable, which suggests that both techniques are interchangeable in cats. Moreover, microcapillary tubes would allow easier repeated sampling in the same cat and would likely be useful in other small species.

Abstract Background Dengue fever (DF) is an acute viral disease which can lead to severe illness, including dengue hemorrhagic fever, marked by thrombocytopenia and hemolytic anemia, as well as end-organ damage. Despite the well-known presentation and prevalence, changes in hematologic markers across the DF course have not been well-described in children. We sought to investigate the association of clinical laboratory values over time with dengue disease progression and outcome in a pediatric population in the Dominican Republic. Methods Pediatric participants were enrolled at Hospital Infantil Dr. Robert Reid Cabral in Santo Domingo, Dominican Republic, in a prospective, observational case-based study. Laboratory values, including complete blood count (CBC) indices and dengue titer results, were collected over the course of hospital stay. Using linear mixed models, we assessed whether 13 different CBC values and time trajectories differed by dengue status, including age and sex as covariates. To account for multiple testing, p≤0.0033 was considered significant. Results A total of 575 children ages 0 to 211 months met inclusion criteria; 51.8% (n=298) were male, and the median (IQR) age was 59 (14-93) months. Eighty-two percent (n=472) of participants had DF. CBC values across days 1 to 10 of fever in those with and without DF are depicted in Figure 1. Those with DF showed levels dropping more quickly across days of fever for hematocrit and hemoglobin (p≤ 0.002), with a more rapid decline in those with severe DF (p &lt; 0.0001). Those with DF had levels increasing more quickly for mean corpuscular hemoglobin concentration (MCHC), monocyte number, and white blood cell counts (p ≤ 0.003), with those with severe DF having a more rapid increase (p &lt; 0.001). The direction of the change across time differed by DF status for mean corpuscular volume and red blood cell distribution width (RDW) (p ≤ 0.0003), with those with severe DF showing an increase in RDW across day of fever (p= 0.0004). Measurements of copper and zinc in transfusion-dependent thalassemia (TDT) show contradictory results.Aim of the study: To examine serum levels of these minerals in TDT in relation to iron overload indices and erythron variables. Methods: This study recruited 60 children with TDT and 30 healthy children aged 3-12 years old.Results: Zinc was significantly higher in TDT children than in control children, whilst copper and the copper to zinc ratio were significantly lowered in TDT. Serum zinc was significantly associated with the number of blood transfusions and iron overload variables (including serum iron and TS%) and negatively with erythron variables (including hemoglobin, mean corpuscular volume, mean corpuscular hemoglobin). Serum copper was significantly and negatively associated with the same iron overload and erythron variables. The copper to zinc ratio was significantly correlated with iron, TS%, ferritin, hemoglobin, mean corpuscular volume, and mean corpuscular hemoglobin. Albumin levels were significantly higher in TDT children than in control children. Conclusion: Our results suggest that the increase in zinc in children with TDT may be explained by iron loading anemia and hemolysis and the consequent shedding of high amounts of intracellular zinc into the plasma. Increased albumin levels and treatment with Desferral may further contribute towards higher zinc levels in TDT. We suggest that the elevations in zinc in TDT are a compensatory mechanism protecting against infection, inflammation, and oxidative stress. Previous proposals for prophylactic use of zinc supplements in TDT may not be warranted.

Decreasing of harpy eagle (Harpia harpyja) populations in natural environments, mainly in non-preserved areas, makes captive population management an important contribution to genetic diversity conservation. The aim of this study is to evaluate hematological parameters for captive harpy eagles maintained at the wild animals breeding center of Itaipu Binacional, Paraná State, Brazil. Fourteen blood samples from nine harpy eagles were collected from animals of both sexes, of different ages and with no clinical signs of disease. Significant variations were found in haematological values of hematocrit, hemoglobin, mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), leukocyte, a relative number of heterophils, absolute and relative number of lymphocytes, monocytes, eosinophils, basophils and plasma protein between groups of young (less than six months old) and adult birds. Comparing males and females there was variation in the values of erythrocytes, hemoglobin, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) on heterophils, absolute and relative number of lymphocytes, eosinophils and basophils. There was also variation in the values of red blood cells, hematocrit, hemoglobin, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), leukocyte count, absolute number of lymphocytes, eosinophils and basophils among birds that study compared to another reference birds. Due to the limited information available on harpy eagle hematology, this study will be useful to the clinical assessment of birds maintained in captivity.

Background: Animal models are widely used in scientific research because of the ability to generate information from an organism like everything under a given experimental condition. Hematological and biochemical tests in laboratory animals are essential for the validation of several scientific studies. In addition, it standardizes physiological values   for these animals according to their sex, age, lineage, environment, and nutritional status. The present work aims to establish reference values   for biochemical and hematological standards in Balb/c mice, for males and females.Materials, Methods & Results: A total of 50 male and female mice were used at reproductive age. The procedures for collecting, processing, and analyzing the samples were standardized.  The collected blood samples were immediately transferred to eppendorf tubes containing heparin, and intended for hematological and biochemical evaluation. The hematological evaluation consisted of Red blood cell count (RBC), Leukocyte counts (WBC), Platelet counts (PLT), Hematocrit (HCT), Hemoglobin concentration (HGB), Mean corpuscular volume (MCV), and Mean corpuscular hemoglobin concentration (MCHC). Already the quantified biochemical parameters were: urea, creatinine, alanina aminotransaminase (ALT), aspartato aminotransaminase (AST) and fosfatase alcalina (FAL). The differential leukocyte count was also performed. Hematological results obtained for males and females were: 9.19 ± 3.35 (106/mm³) and 7.3 ± 2.01(106/mm³) of RBC; 35.8 ± 6.7% and 38.44 ± 3.93% of HCT; 11.51 ± 2.17 g/dL and 11.85 ± 1.56 g/dL of HGB; 45.83 ± 15.03 fL and 60.26 ± 18.25 fL of VCM; 31.80 ± 1.15% and 31.88 ± 0.99% of MCHC; and, 5380 ± 1994.21(10³/mm³) and 3564 ± 1071(10³/mm³) of WBC.  The platelet counts were 878.92 ± 84.19 and 678.28 ± 227.21, for males and females respectively. And for differential leukocyte counts, for males and females: eosinophils 2.12 ± 1.09% and 2.16 ± 1.71%; monocytes 2.84 ± 1.03% and 2.68 ± 1%; lymphocytes 68 ± 8.36% and 71.76 ± 5.9%; neutrophils 27.04 ± 8.55% and 22.96 ± 5.54%. Basophils were not quantified in the samples. As for the biochemical parameters, values of 54.16 ± 27.8 UI/L and 29.72 ± 4.4 UI/L of  ALT; 89.56 ± 47.73 UI/L and 71.32 ± 8.12 UI/L of AST; 3.76 ± 2.08 UI/L and 2.32 ± 0.85 UI/L of  FAL; 31.76 ± 21.08 mg/dL and 41.48 ± 13.61 mg/dL of urea; and 0.76 ± 0.18 mg/dL and 0.44 ± 0.11 mg/dL of creatinine.Discussion: The mean corpuscular volume, mean corpuscular hemoglobin concentration, leukocyte, and platelet counts diverged from those   found in literature. For the biochemical values, it was observed that creatinine values were different from those exhibited by other authors. Such divergences might be explained by the activity of endocrine organs, such as the production and/or release of activation/differentiation factors, and stress, applied methodology, lineage, or individual variability.  In addition, differences in the methodologies applied may be responsible for variations in hematological and biochemical values, requiring the standardization of the equipment and reagents used, as well as the adoption of a range that represents the minimum and maximum values within the normal physiological standard for given mouse lineage. In conclusion, the values presented in the present work are within the variation curve for rodents, and can be used as reference for other studies that use these animals.

<em>Skin flaps </em>merupakan salah satu prosedur bedah rekonstruksi untuk penutupan luka. Tujuan penelitian ini adalah melihat profil hematologi selama proses kesembuhan luka menggunakan teknik <em>skin flaps H˗plasty</em> dan <em>linear closure </em>pada kucing (<em>Felis catus</em>). Pembuatan luka dengan luas 2x2 cm dilakukan pada daerah toraks 6 kucing lokal jantan sehat yang dibagi dalam dua kelompok teknik penutupan luka, <em>skin flaps</em> <em>H-plasty</em> dan <em>linear closure</em>. Pengambilan darah sebanyak 1 ml dilakukan secara aseptis pada <em>vena saphena</em> pada hari ke˗0 pra operasi, hari ke˗3, 6, 9, dan 12 pascaoperasi. Parameter pengamatan pada nilai sel darah merah (SDM), hemoglobin (Hb), hematokrit (Hct), Mean Corpuscular Volume (MCV), Mean Corpuscular Hemoglobin (MCH), Mean Corpuscular Hemoglobin Concentration (MCHC) dan trombosit. Hasil penelitian menunjukkan bahwa rata-rata SDM, hemoglobin, hematokrit, MCHC, dan trombosit tidak terjadi perbedaan yang signifikan (P&gt;0.05) antar kelompok perlakuan <em>H-plasty</em> dengan <em>linear closure</em>. Parameter MCV dan MCH menunjukkan perbedaan signifikan (P&lt;0.05) antar kelompok perlakuan. Pengamatan rata-rata SDM, hemoglobin, hematokrit, MCV, MCH, MCHC dan trombosit menunjukkan perbedaan yang signifikan (P&lt;0.05) diantara hari pengamatan. Kedua teknik <em>skin flaps</em> menunjukkan nilai rata˗rata profil hematologi yang fisiologis selama proses kesembuhan luka.
